    Methodological Bias in Estimates of Strain Composition and Straying of Hatchery‐Produced Steelhead in Lake Michigan Tributaries

    Steelhead Oncorhynchus mykiss were first introduced into the Great Lakes in the late 1800s. Subsequently, natural recruitment of steelhead from spawning runs in streams across the basin has been regularly supplemented by hatchery production of strains derived from widely dispersed locales within the species’ native range. Estimates of hatchery contributions to the spawning runs of naturalized populations may be underrepresented by observations of marked fish, as not all hatchery fish are marked prior to release. To assess the potential bias in estimates of the hatchery contribution to steelhead spawning runs in four major rivers in Michigan, we used scale pattern analysis (SPA) to identify nonmarked hatchery fish and multilocus genotypes to estimate the proportional contributions of each hatchery strain to spawning runs. The four hatchery strains currently stocked are significantly genetically distinct (mean FST = 0.077), making it possible to identify specific strains by use of likelihood‐based assignment tests. The differences between direct (mark observations) and indirect (SPA and genetic analysis) estimates of hatchery contribution were mainly due to variations in the percentage of hatchery fish marked by states prior to release and the potential for confusion between certain marks and injuries. By combining direct and indirect assessment methodologies, we estimated that the percentage of hatchery fish returning to the four rivers ranged from 13% to 31% of total spawning runs.     This chapter provides a comprehensive review of both laboratory characterization and modelling of bulk material fracture in asphalt mixtures. For the purpose of organization, this chapter is divided into a section on laboratory tests and a section on models. The laboratory characterization section is further subdivided on the basis of predominant loading conditions (monotonic vs. cyclic). The section on constitutive models is subdivided into two sections, the first one containing fracture mechanics based models for crack initiation and propagation that do not include material degradation due to cyclic loading conditions. The second section discusses phenomenological models that have been developed for crack growth through the use of dissipated energy and damage accumulation concepts.     Inhibition of Microsomal Prostaglandin E2 Synthase Reduces Collagen Deposition in Melanoma Tumors and May Improve Immunotherapy Efficacy by Reducing T-cell Exhaustion

    The arachidonic acid pathway participates in immunosuppression in various types of cancer. Our previous observation detailed that microsomal prostaglandin E2 synthase 1 (mPGES-1), an enzyme downstream of cyclooxygenase 2 (COX-2), limited antitumor immunity in melanoma; in addition, genetic depletion of mPGES-1 specifically enhanced immune checkpoint blockade therapy. The current study set out to distinguish the roles of mPGES-1 from those of COX-2 in tumor immunity and determine the potential of mPGES-1 inhibitors for reinforcing immunotherapy in melanoma. Genetic deletion of mPGES-1 showed different profiles of prostaglandin metabolites from that of COX-2 deletion. In our syngeneic mouse model, mPGES-1-deficient cells exhibited similar tumorigenicity to that of COX-2-deficient cells, despite a lower ability to suppress PGE2 synthesis by mPGES-1 depletion, indicating the presence of factors other than PGE2 that are likely to regulate tumor immunity. RNA-sequencing analysis revealed that mPGES-1 depletion reduced the expressions of collagen-related genes, which have been found to be associated with immunosuppressive signatures. In our mouse model, collagen was reduced in mPGES-1-deficient tumors, and phenotypic analysis of tumor-infiltrating lymphocytes indicated that mPGES-1-deficient tumors had fewer TIM3+ exhausted CD8+ T cells compared with COX-2-deficient tumors. CAY10678, an mPGES-1 inhibitor, was equivalent to celecoxib, a selective COX-2 inhibitor, in reinforcing anti-PD-1 treatment. Our study indicates that mPGES-1 inhibitors represent a promising adjuvant for immunotherapies in melanoma by reducing collagen deposition and T-cell exhaustion

    The Taiwanese-American Occultation Survey (TAOS) project has collected more than a billion photometric measurements since 2005 January. These sky survey data—covering timescales from a fraction of a second to a few hundred days—are a useful source to study stellar variability. A total of 167 star fields, mostly along the ecliptic plane, have been selected for photometric monitoring with the TAOS telescopes. This paper presents our initial analysis of a search for periodic variable stars from the time-series TAOS data on one particular TAOS field, No. 151 (R.A. = 17^(h)30^(m)6^(s).7, decl. = 27°17'30", J2000), which had been observed over 47 epochs in 2005. A total of 81 candidate variables are identified in the 3 deg^2 field, with magnitudes in the range 8 < R < 16. On the basis of the periodicity and shape of the light curves, 29 variables, 15 of which were previously unknown, are classified as RR Lyrae, Cepheid, ή Scuti, SX Phonencis, semi-regular, and eclipsing binaries

    Search for Small Trans-Neptunian Objects by the TAOS Project

    The Taiwan-America Occultation Survey (TAOS) aims to determine the number of small icy bodies in the outer reach of the Solar System by means of stellar occultation. An array of 4 robotic small (D=0.5 m), wide-field (f/1.9) telescopes have been installed at Lulin Observatory in Taiwan to simultaneously monitor some thousand of stars for such rare occultation events. Because a typical occultation event by a TNO a few km across will last for only a fraction of a second, fast photometry is necessary. A special CCD readout scheme has been devised to allow for stellar photometry taken a few times per second. Effective analysis pipelines have been developed to process stellar light curves and to correlate any possible flux changes among all telescopes. A few billion photometric measurements have been collected since the routine survey began in early 2005. Our preliminary result of a very low detection rate suggests a deficit of small TNOs down to a few km size, consistent with the extrapolation of some recent studies of larger (30--100 km) TNOs.Comment: 4 pages, 3 figures, IAU Symposium 23

    The TAOS Project Stellar Variability I. Detection of Low-Amplitude delta Scuti Stars

    We analyzed data accumulated during 2005 and 2006 by the Taiwan-American Occultation Survey (TAOS) in order to detect short-period variable stars (periods of <~ 1 hour) such as delta Scuti. TAOS is designed for the detection of stellar occultation by small-size Kuiper Belt Objects (KBOs) and is operating four 50cm telescopes at an effective cadence of 5Hz. The four telescopes simultaneously monitor the same patch of the sky in order to reduce false positives. To detect short-period variables, we used the Fast Fourier Transform algorithm (FFT) inasmuch as the data points in TAOS light-curves are evenly spaced. Using FFT, we found 41 short-period variables with amplitudes smaller than a few hundredths of a magnitude and periods of about an hour, which suggest that they are low-amplitude delta Scuti stars (LADS).     A Close Binary Star Resolved from Occultation by 87 Sylvia

    The star BD+29 1748 was resolved to be a close binary from its occultation by the asteroid 87 Sylvia on 2006 December 18 UT. Four telescopes were used to observe this event at two sites separated by some 80 km apart. Two flux drops were observed at one site, whereas only one flux drop was detected at the other. From the long-term variation of Sylvia, we inferred the probable shape of the shadow during the occultation, and this in turn constrains the binary parameters: the two components of BD+29 1748 have a projected separation of 0.097" to 0.110" on the sky with a position angle 104 deg to 107 deg. The asteroid was clearly resolved with a size scale ranging from 130 to 290 km, as projected onto the occultation direction.     A Critical Review of Bioaccumulation and Biotransformation of Organic Chemicals in Birds

    A literature review of bioaccumulation and biotransformation of organic chemicals in birds was undertaken, aiming to support scoping and prioritization of future research. The objectives were to characterize available bioaccumulation/ biotransformation data, identify knowledge gaps, determine how extant data can be used, and explore the strategy and steps forward. An intermediate approach balanced between expediency and rigor was taken given the vastness of the literature. Following a critical review of \u3e500 peer-reviewed studies, \u3e25,000 data entries and 2 million information bytes were compiled on \u3e700 organic compounds for ~ 320 wild species and 60 domestic breeds of birds. These data were organized into themed databases on bioaccumulation and biotransformation, field survey, microsomal enzyme activity, metabolic pathway, and bird taxonomy and diet. Significant data gaps were identified in all databases at multiple levels. Biotransformation characterization was largely fragmented over metabolite/pathway identification and characterization of enzyme activity or biotransformation kinetics. Limited biotransformation kinetic data constrained development of an avian biotransformation model. A substantial shortage of in vivo biotransformation kinetics has been observed as most reported rate constants were derived in vitro. No metric comprehensively captured all key contaminant classes or chemical groups to support broad-scope modeling of bioaccumulation or biotransformation. However, metrics such as biota-feed accumulation factor, maximum transfer factor, and total elimination rate constant were more readily usable for modeling or benchmarking than other reviewed parameters. Analysis demonstrated the lack of bioaccumulation/biotransformation characterization of shorebirds, seabirds, and raptors. In the study of bioaccumulation and biotransformation of organic chemicals in birds, this review revealed the need for greater chemical and avian species diversity, chemical measurements in environmental media, basic biometrics and exposure conditions, multiple tissues/matrices sampling, and further exploration on biotransformation. Limitations of classical bioaccumulation metrics and current research strategies used in bird studies were also discussed. Forward-looking research strategies were proposed: adopting a chemical roadmap for future investigations, integrating existing biomonitoring data, gap-filling with non-testing approaches, improving data reporting practices, expanding field sampling scopes, bridging existing models and theories, exploring biotransformation via avian genomics, and establishing an online data repository
