    A psychological approach to providing self-management education for people with type 2 diabetes : the diabetes manual

    Background: The objectives of this study were twofold (i) to develop the Diabetes Manual, a selfmanagement educational intervention aimed at improving biomedical and psychosocial outcomes (ii) to produce early phase evidence relating to validity and clinical feasibility to inform future research and systematic reviews. Methods: Using the UK Medical Research Council's complex intervention framework, the Diabetes Manual and associated self management interventions were developed through preclinical, and phase I evaluation phases guided by adult-learning and self-efficacy theories, clinical feasibility and health policy protocols. A qualitative needs assessment and an RCT contributed data to the pre-clinical phase. Phase I incorporated intervention development informed by the preclinical phase and a feasibility survey. Results: The pre-clinical and phase I studies resulted in the production in the Diabetes Manual programme for trial evaluation as delivered within routine primary care consultations. Conclusion: This complex intervention shows early feasibility and face validity for both diabetes health professionals and people with diabetes. Randomised trial will determine effectiveness against clinical and psychological outcomes. Further study of some component parts, delivered in alternative combinations, is recommended

    A Body of Text: Physical Culture and the Marketing of Mobility

    Drawing on Althusserian notions of ideology and Bourdieu’s understanding of bodily hexis, A Body of Text seeks to reframe Physical Culture as an artifact worthy of serious study, more complex and less reactionary than its beefcake-and-sentiment reputation might suggest. This dissertation addresses the story of Physical Culture magazine from three different perspectives, reading the magazine through lenses of media history, medical history and social context, in order to understand the ways in which class operated on and through the body. In contrast to nearly every other publication in the early twentieth century, Physical Culture suggested that class mobility was possible, and that success would naturally follow improvement of body and health. Whereas the idea of “fitness” in the eugenics movement very clearly indicated an essential condition, consequent to the quality of the germline, Physical Culture initiated the idea that fitness was obtainable – and commodifiable – through diet, exercise and what we now call “lifestyle.” Much of this argument is constructed by contrasting Physical Culture with Hygeia, the health magazine created by the American Medical Association for a lay audience, and by contrasting Physical Culture impresario Bernarr Macfadden with his counterpart at the AMA, Morris Fishbein. Whereas Macfadden’s story has been told and retold (albeit in biographies that are increasingly interesting but not yet definitive), Fishbein’s story has yet to be the subject of a responsible biography. First gestures in that direction open the door to further work on Fishbein as a subject, and to deeper studies of the relationship between medicine, marketing and modern consumerism. Far from suggesting that Macfadden is ipso facto a liberatory force or a sophisticated theorist, the likeliest explanation for the complex, unstable and evolving constructions of body politics in Physical Culture are twofold: first, coming himself from “unfit” germlines, Macfadden needs to enrich contemporary thinking about the body to make sense of (and room for) his own success; second, and more importantly: you can’t sell a bloodline

    Journal ArticleFarnesyl diphosphate synthase catalyzes the sequential head-to-tail condensation of two molecules of isopentenyl diphosphate with dimethylallyl diphosphate. In plants the presence of farnesyl diphosphate synthase isozymes offers the possibility of differential regulation. Three full-length cDNAs encoding putative isoprenoid synthases, FDS-1, FDS-2, and FDS-5, with greater than 89% similarity were isolated from a Big Sagebrush Artemisia tridentata cDNA library using a three-step polymerase chain reaction protocol. One of the open reading frames, FDS-5, encoded a protein with an N-terminal amino acid extension that was identified as a plastidial targeting peptide. Recombinant histidine-tagged versions of three proteins were purified, and their enzymatic properties were characterized. FDS-1 and FDS-2 synthesized farnesyl diphosphate as the final chain elongation product, but their kinetic behavior varied. FDS-1 prefers geranyl diphosphate over dimethylallyl diphosphate as an allylic substrate and is active at acidic pH values compared with FDS-2. In contrast, FDS-5 synthesized two irregular monoterpenoids, chrysanthemyl diphosphate and lavandulyl diphosphate, when incubated with dimethylallyl diphosphate and an additional product, the regular monoterpene geranyl diphosphate, when incubated with isopentenyl diphosphate and dimethylallyl diphosphate. Specific cellular functions are proposed for each of the three enzymes, and a scenario for evolution of isoprenyl synthases in plants is presented

    Geographic Survey of Oxyspirura Petrowi Among Wild Northern Bobwhites in the United States

    Eyeworms (Oxyspirura petrowi) are potentially associated with northern bobwhite (Colinus virginianus) declines. We examined hunter-donated bobwhites from the 2013–2015 hunting seasons in 9 states to document infection prevalence (% of bobwhites [of total n]) and intensity (mean no. of eyeworms ± SE). Four states harbored infected bobwhites: Texas (59.1% [n = 110], 15.6 ± 2.1), Oklahoma (52.1% [n = 121], 6.9 ± 1.2), Virginia (14.8% [n = 27], 2.5 ± 1.0), and Alabama (1.6% [n = 61], 2.0). Prevalence and intensity of eyeworms in the Texas Rolling Plains were greater (P \u3c 0.001 and P = 0.002, respectively) than in any other area sampled. Based on our survey, eyeworms are locally prevalent and abundant in bobwhites from the Rolling Plains ecoregion, but virtually nonexistent in many areas that we surveyed

    Genetic Locus Required for Antigenic Maturation of \u3cem\u3eRhizobium etli\u3c/em\u3e CE3 Lipopolysaccharide

    Rhizobium etli modifies lipopolysaccharide (LPS) structure in response to environmental signals, such as low pH and anthocyanins. These LPS modifications result in the loss of reactivity with certain monoclonal antibodies. The same antibodies fail to recognize previously isolated R. etli mutant strain CE367, even in the absence of such environmental cues. Chemical analysis of the LPS in strain CE367 demonstrated that it lacked the terminal sugar of the wild-type O antigen, 2,3,4-tri-O-methylfucose. A 3-kb stretch of DNA, designated as lpe3, restored wild-type antigenicity when transferred into CE367. From the sequence of this DNA, five open reading frames were postulated. Site-directed mutagenesis and complementation analysis suggested that the genes were organized in at least two transcriptional units, both of which were required for the production of LPS reactive with the diagnostic antibodies. Growth in anthocyanins or at low pH did not alter the specific expression of gusA from the transposon insertion of mutant CE367, nor did the presence of multiple copies of lpe3 situated behind a strong, constitutive promoter prevent epitope changes induced by these environmental cues. Mutations of the lpe genes did not prevent normal nodule development on Phaseolus vulgaris and had very little effect on the occupation of nodules in competition with the wild-type strain

    Seasonality in the Surface Energy Balance of Tundra in the Lower Mackenzie River Basin

    This study details seasonal characteristics in the annual surface energy balance of upland and lowland tundra during the 1998–99 water year (Y2). It contrasts the results with the 1997–98 water year (Y1) and relates the findings to the climatic normals for the lower Mackenzie River basin region. Both years were much warmer than the long-term average, with Y1 being both warmer and wetter than Y2. Six seasons are defined as early winter, midwinter, late winter, spring, summer, and fall. The most rapid changes in the surface energy balance occur in spring, fall, and late winter. Of these, spring is the most dynamic, and there is distinct asymmetry between rates of change in spring and those in fall. Rates of change of potential insolation (extraterrestrial solar radiation) in late winter, spring, and fall are within 10% of one another, being highest in late winter and smallest in spring. Rates of change in air temperature and ground temperature are twice as large in spring as in fall and late winter, when they are about the same. Rates of change in components of the energy balance in spring are twice and 4 times as large as in fall and late winter, respectively. The timing of snowpack ripening and snowmelt is the major agent determining the magnitude of asymmetry between fall and spring. This timing is a result of interaction between the solar cycle, air temperature, and snowpack longevity. Based on evidence from this study, potential surface responses to a 18C increase in air temperature are small to moderate in most seasons, but are large in spring when increases range from 7% to 10% of average surface energy fluxes

    Total Energy Expenditure and Body Composition of Children with Developmental Disabilities

    Background Obesity prevalence is increased in children with developmental disabilities, specifically in children with spina bifida and Down syndrome. Energy expenditure, a critical aspect of weight management, has been extensively studied in the typically developing population, but not adequately studied in children with developmental disabilities. Objective Determine energy expenditure, fat-free mass and body fat percentile and the impact of these findings on recommended caloric intake in children with spina bifida and Down syndrome. Methods/Measures This pilot study included 36 children, 18 with spina bifida, 9 with Down syndrome and 9 typically developing children. Half of the children with spina bifida were non-ambulatory. Doubly labeled water was used to measure energy expenditure and body composition. Descriptive statistics described the sample and MANOVA and ANOVA methods were used to evaluate differences between groups. Results Energy expenditure was significantly less for children with spina bifida who primarily used a wheelchair (p = .001) and children with Down syndrome (p = .041) when compared to children without a disability when adjusted for fat-free mass. However, no significant difference was detected in children with spina bifida who ambulated without assistance (p = .072). Conclusions Children with spina bifida and Down syndrome have a significantly decreased energy expenditure which directly impacts recommended caloric intake. No significant difference was detected for children with spina bifida who ambulated, although the small sample size of this pilot study may have limited these findings. Validating these results in a larger study is integral to supporting successful weight management of these children
