Linee educative nella Lettera Enciclica sulla cura della casa comune Laudato si\u2019
Papa Francesco non \ue8 un pedagogista, eppure, nell'enciclica Laudato si' propone numerose e pressanti sollecitazioni di carattere educativo, profondamente preoccupato per l'uso irresponsabile oggi della terra, casa comune dell'umanit\ue0, tanto da parlare di una vera catastrofe ecologica La lucida analisi non cede a una lettura pessimistica, quanto piuttosto \ue8 in grado di cogliere elementi critici e positivi, non soffermandosi su un'amara denuncia, ma suggerendo possibili piste di nuovo impegno che la sfida educativa presenta. La prospettiva da cui muove \ue8 quella di una nuova solidariet\ue0 universale, che scaturisce dalla convinzione che nel mondo tutto \ue8 intimamente connesso, tutto \ue8 in relazione, tra le cose esistono molteplici relazioni. Il presente contributo intende individuare alcune possibili linee di impegno educativo volte a far assumere comportamenti concreti, partendo da una lettura dell'enciclica, dal punto di vista educativo. Da una visione che abbraccia la complessit\ue0 della problematica ecologica in prospettiva mondiale, conseguono non solo interessanti riflessioni, ma anche l'indicazione di alcune possibili buone pratiche che il Papa Francesco propone per educare bambini e ragazzi.Pope Francis is not a pedagogue, and yet, in the encyclical Praised be to You ' he offers many pressing urging of an educational nature, deeply concerned about the irresponsible use of the earth today, the common home of humanity, so much to speak of a real ecological catastrophe. The clear analysis does not yield to a pessimistic reading, but rather is able to capture critical and positive elements, not dwelling on a bitter complaint, but suggesting possible ways of new commitments that the educational challenge presents. The perspective from which it moves is to a new universal solidarity, which springs from the belief that everything in the world is intimately connected, everything is related, all is in interaction, and there is a multiplicity of relationship. This paper seeks to identify some possible lines of educational efforts aimed at making concrete attitudes, from a reading of the encyclical, from the educational point of view. 3,5-dicaffeoylquinic acid lowers 3T3-L1 mitotic clonal expansion and adipocyte differentiation by enhancing heme oxygenase-1 expression
Adipogenesis is a complex process in which cell commitment and mitotic clonal expansion (MCE) are in-sequence crucial events leading to terminal adipocyte differentiation. The molecules able to block some key signals in this cascade can hamper adipogenesis becoming promising agents to counteract hyperplasia and hypertrophy of adipose tissue. Mono- and di-caffeoylquinic acid isomers are biologically active polyphenols, displaying in vitro and in vivo antioxidant, hepatoprotective, anti-diabetic and anti-obesity properties. Among these isomers, 3,5-dicaffeoylquinic acid (DCQA) has been reported to inhibit lipid accumulation in adipose cells more successfully than others. Thus, we investigated DCQA effects and molecular mechanisms on 3T3-L1 pre-adipocytes induced to differentiate with a hormonal cocktail (MDI). Oil Red O incorporation assessed that DCQA pre-treatment inhibited lipid accumulation in 3T3-L1 cells induced to differentiate for 10 days. At this time, an increased phosphorylation of both AMP-activated kinase and acetyl-CoA carboxylase, as well as a strong decrease in fatty acid synthase protein level, were registered by immunoblotting, thereby suggesting that DCQA treatment can reduce fatty acid anabolism in 3T3-L1 adipocytes. Furthermore, BrdU incorporation assay, performed 48 h after hormonal stimulation, revealed that DCQA treatment was also able to hinder the 3T3-L1 cell proliferation during the MCE, which is an essential step in the adipogenic process. Thus, we focused our attention on early signals triggered by the differentiation stimuli. In the first hours after hormonal cocktail administration, the activation of ERK1/2 and Akt kinases, or CREB and STAT3 transcription factors, was not affected by DCQA pre-treatment. Whereas 24 h after MDI induction, DCQA pre-treated cells showed increased level of the transcription factor Nrf2, that induced the expression of the antioxidant enzyme heme oxygenase 1 (HO-1). In control samples, the expression level of HO-1 was reduced 24 h after MDI induction in comparison with the higher amount of HO-1 protein found at 2 h. The HO-1 decrease was functional by allowing reactive oxygen species to boost and allowing cell proliferation induction at the beginning of MCE phase. Instead, in DCQA-treated cells the HO-1 expression was maintained at high levels for a further 24 h; in fact, its expression decreased only 48 h after MDI stimulation. The longer period in which HO-1 expression remained high led to a delay of the MCE phase, with a subsequent inhibition of both C/EBP-α expression and adipocyte terminal differentiation. In conclusion, DCQA counteracting an excessive adipose tissue expansion may become an attractive option in obesity treatment

Exposure of GH-3 cells to epidermal growth factor for 4 consecutive days induced the expression of both D-2(415) and D-2(444) dopamine-receptor isoforms. Epidermal growth factor also promoted a remarkable increase in the content of Gi3 protein, which is responsible for receptor-induced activation of potassium channels in GH-3 cells. D-2 receptors in this model apparently activate a specific transducing pathway, leading to opening of potassium channels and inhibition of prolactin release by cAMP-independent mechanisms. This is shown by: 1) the selective D-2 agonist quinpirole, while inactive on vasoactive intestinal peptide-induced prolactin release, strongly inhibited the hormone secretion induced by neurotensin; 2) quinpirole, up to 100 microM, did not inhibit cAMP production evoked by vasoactive intestinal peptide both in intact cells and in broken cell membrane preparations; and 3) quinpirole and other D-2 agonists strongly potentiated Rb+ efflux when measured in a nominally calcium-free reaction solution containing 100 mM potassium (voltage-dependent component), but did not modify Rb+ efflux if measured in a reaction solution containing 1 mM calcium and 5 mM potassium (calcium-activated, cAMP-dependent component)
PPAR-α Contributes to the Anti-Inflammatory Activity of Verbascoside in a Model of Inflammatory Bowel Disease in Mice
The previous results suggest that peroxisome proliferator-activated receptor-alpha (PPAR)-α, an intracellular transcription factor activated by fatty acids, plays a role in control of inflammation. There is persuasive epidemiological and experimental evidence that dietary polyphenols have anti-inflammatory activity. In this regard, it has been demonstrated that verbascoside (VB) functions as intracellular radical scavenger and reduces the microscopic and macroscopic signs of experimental colitis. With the aim to characterize the role of PPAR-α in VB-mediated anti-inflammatory activity, we tested the efficacy of VB in an experimental model of inflammatory bowel disease induced by dinitrobenzene sulfonic acid, comparing mice lacking PPAR-α (PPAR-αKO) with wild type (WT) mice. Results indicate that VB-mediated anti-inflammatory activity is weakened in PPAR-αKO mice, compared to WT controls, especially in the inhibition of neutrophil infiltration, intestinal permeability and colon injury. These results indicate that PPAR-α can contribute to the anti-inflammatory activity of VB in inflammatory bowel disease
Robotic treatment of a rare paramedian cystic lesion of the lower male urogenital tract
Abstract Objectives To report the diagnosis and the mini-invasive treatment of a rare paramedian cystic lesion of the lower male urogenital tract (CLMGU). Material and methods This is a case of a 46 old male with low urinary tract symptoms, azoospermia and mild erectile dysfunction. MRI Imaging showed a midline high signal intensity cystic lesion of a tear drop shape, extending above the base of the prostate to the level of the seminal vesicles. The lesion was located between the seminal vesicles and the urinary bladder. A direct communication of the intra-abdominal portion of both vas deferens with the cyst was found. MRI also showed small solid nodule within the upper portion of the cyst. The patient underwent a robotic surgery of the CLMGU. A Retzius-sparing approach was used to gain access to the seminal vesicles in order to carefully dissect and excise the lesion without any dissection of the anterior compartment. The CLMGU was excised with a nerve sparing technique. Results Operative time was 115 min. Blood loss was minimal. Length of stay was three days. No post-operative complications occurred. One month after surgery patient's IPSS improved considerably. Final pathology showed a cystic lesion containing papillary projections with squamous metaplasia. At 2 months follow up, urinary symptoms improved with no postoperative complications. Conclusion Robotic surgery allows a direct access to the Douglas space with an easy removal of the neoplasia. Our video represents a case of possible application of robotic surgery to improve dissection, overall surgical precision and functional outcomes
