    Sample size for family evaluation in potato breeding programs.

    Clonal families from a broad genetic base population in the Potato Breeding Program at the Universidade Federal de Lavras (UFLA), Brazil, were used in this trials. Twenty-five families were assessed in a 5 x 5 triple lattice design. Each plot consisted of 30 clones distributed in three rows of ten plants. Tuber yield per plant, percentage of large tubers, mean weight of large tubers, mean medium-sized tuber weight and tuber specific gravity were measured. Three hundred experiments were simulated varying the family sizes from three to 90 clones. The coefficients of experimental variation (CVe), the coefficients of genetic variation (CVg), heritabilities for family mean and the CVg/CVe ratio were estimated. Genetic parameters were stabilized with family sizes as small as six clones, depending on the trait. This indicates that the families can be adequately represented by a small sample of clones. Using the maximum curvature method it is possible to conclude that approximately 30 clones would be sufficient to represent each family, even for traits with the highest CVe. The genetic variance within family was greater than the genetic variance among families for all traits, indicating a favorable potential for within family selection. The correlation coefficients of the family means with the 5%, 10%, 15% and 20% best clones from each family, considering the five traits assessed, were always high, meaning that within the best families generally are the best clones

    Estudo do impacto no meio ambiente pelo processamento da farinha de mandioca no Município de São Felipe, Bahia.

    O município de São Felipe localiza-se a uma latitude 12º50'50" sul e a uma longitude 39º05'22" oeste estando a uma altitude de 195 metros. Possui uma área de 198,71 km² uma população de aproximadamente 21.000 habitantes. Das atividades agrícolas destaca-se o cultivo da mandioca com técnicas ainda rudimentares. O Brasil é o segundo produtor de mandioca do mundo, sendo a planta resistente nas diferentes condições de cultivo, encontrada em todos os estados brasileiros. As utilizações culinárias caseiras da mandioca não acumulam resíduo significativo, pela pequena quantidade processada. Contudo, quando o processamento é maior, os subprodutos podem vir a apresentar problemas de disposição e soluções deverão ser encontradas. O processamento industrial da mandioca causa sérios problemas ambientais na disposição de resíduos, como poluição de rios, do solo, lençol freático e mortandade de peixes. Apesar dos agricultores serem familiarizados com a mandioca, desconhecem as variedades, os impactos que seus resíduos causam ao meio ambiente e o aproveitamento dos resíduos como uma fonte de sustentabilidade.PDF.035

    Caracterização da estrutura genética do pinhão manso (Jatropha curcas L.): isolamento de fragmentos de dna enriquecidos com sequências de microssatélites.

    O pinhão manso (Jatropha curcas) é uma planta oleaginosa que vem se destacando por apresentar um alto potencial na produção de biocombustíveis. Por este motivo o interesse em sua exploração comercial vem aumentado consideravelmente. Assim, torna-se imprescindível o desenvolvimento de tecnologias moleculares de monitoramento e seleção de populações naturais e cultivares. Nesse contexto, marcadores moleculares como os microssatélites são amplamente utilizados. No entanto, os genomas de organismos eucariontes são muito amplos e, assim, o isolamento de seqüências de microssatélites específicas, que possam atuar como marcadores, torna-se uma tarefa complexa. O presente trabalho teve como objetivo estabelecer um protocolo para isolar fragmentos de DNA específicos contendo loci de microssatélites. Este procedimento tem por finalidade aumentar a incidência das seqüências alvo para futura clonagem. Foram testadas diferentes enzimas de restrição e sondas de DNA para o reconhecimento e captura de fragmentos contendo seqüências de microssatélites. Os resultados obtidos mostraram que a enzima RsaI apresentou o melhor padrão de digestão do DNA, enquanto as melhores sondas foram as compostas pelas repetições (AATG)6, (AAAC)6, (AATC)6 e (AAAG)6. Estes procedimentos constituem etapas fundamentais no processo de isolamento de microssatélites e contribuirão para a caracterização da estrutura genética do pinhão manso. Com isto, será possível avaliar quais os estoques mais adequados para seleção de linhagens viáveis para o cultivo, visando ganhos na produtividade

    Population connectivity and larval dispersal of the exploited mangrove crab Ucides cordatus along the Brazilian coast.

    Abstract: Background: The mangrove crab Ucides cordatus is considered a key species for the ecological balance of mangrove forests and a major source of employment and income for traditional crab collectors in Brazil. Several studies evidenced weak genetic variation among populations due to an efficient larval transport. However, gene flow patterns of the species is poorly understood, with no information about migration rates. The influence of the two main Brazilian currents in larval dispersion is also not clear. In order to provide baseline information for conservation, planning and management of this important fishery resource, the present study aimed to estimate and evaluate spatial distribution of genetic diversity, migration rates and gene flow directivity among populations of U. cordatus in Brazil. Methods:Nine microsatellites were used to resolve population structure of 319 crabs collected from six sites located along the Brazilian coast. The degree of geographical differentiation included estimates of genetic diversity, population structure and gene flow models, with spatial analysis of shared alleles (SAShA), isolation by distance tests, AMOVA, discriminant analysis of principal components (DAPC) and Bayesian clustering. We estimated the amount of ongoing gene flow between clusters using the coalescent-based method implemented in Migrate-N. Results:Loci were highly polymorphic (average of 12.4 alleles per locus) evidencing high genetic variability. There was significant differentiation among localities, despite of the low value of FST (= 0.019; P < 0.001). FST and Jost's D indexes were also estimated in pairwise comparisons and showed significant differences between most of the surveyed site pairs (P < 0.05). Structure evidenced a single genetic group among samples, however SAShA pointed to a non-panmictic condition (P = 0.011). AMOVA detected four statistical significant clusters with low level of differentiation (FCT = 0.037; P = 0.023). The gene flow model that best described the population connectivity was the island model, with ?24 crabs being exchanged among localities per generation.DIscussion:The high migration rates found among localities seem to be the main force acting to sustain the distribution of the genetic diversity of U. cordatus. Despite the high gene flow and the weak population structure among samples, the significant genetic differences found suggest that gene flow alone does not bypass the effects of genetic drift, natural selection and/or human exploitation. These findings are vital for the establishment of a database to be used in the development of conservation programs

    Couples that work full time and are happily married : myth or reality?

    Este trabalho apresenta parte de uma pesquisa quantitativa cujo principal objetivo foi avaliar a satisfação no casamento de homens e mulheres que optaram por relacionamentos de duplo trabalho. Participaram do estudo 222 homens e 222 mulheres casados/as, funcionários/as em diversas instituições públicas de Brasília - DF. A maior concentração de respondentes de ambos os sexos está na faixa etária entre 31 a 40 anos. Os participantes responderam a um questionário demográfico e à Escala de Ajustamento Diádico - DAS. Os resultados mostraram que a maioria dos participantes está satisfeita com seus relacionamentos, sendo que as mulheres apresentaram média de satisfação inferior à dos homens. Quanto à percepção do futuro do relacionamento, ficou evidente o comprometimento de homens e mulheres em investirem na manutenção do casamento. Os resultados questionam a idéia vigente de falência do casamento e da família e apontam para uma transformação das relações. _________________________________________________________________________________________________________ ABSTRACTThis paper presents part of a quantitative research regarding marriages in which men and women are engaged in full time work. The main objective was to evaluate marital satisfaction in dual worker couples. Subjects were 222 married men and 222 married women that work in several public institutions in Brasilia - DF. The majority of the participants were between 31 to 40 years old. They answered a demographic questionnaire and the Dyadic Adjustment Scale - DAS. Results showed that the majority of participants were satisfied with their relationships although women presented lower satisfaction scores then men. Regarding the perception of the future of the relationship, the results showed that subjects were committed to preserve their marriage. These data question current ideas that marriages and families are outmoded and point in the direction of a transformation in interpersonal relationships

    Nanocomposite Nafion-Silica membranes for direct methanol fuel cells

    Commercially available proton exchange membranes such as Nafion do not meet the requirements for high power density direct methanol fuel cells, partly due to their high methanol permeability. The aim of this work is to develop a new class of high-proton conductivity membranes, with thermal and mechanical stability similar to Nafion and reduced methanol permeability. Nanocomposite membranes were produced by the in-situ sol-gel synthesis of silicon dioxide particles in preformed Nafion membranes. Microstructural modification of Nafion membranes with silica nanoparticles was shown in this work to reduce methanol crossover from 7.48x10-6 cm2s^-1 for pure Nafion® to 2.86 x10-6 cm2s^-1 for nanocomposite nafion membranes (Methanol 50% (v/v) solution, 75 degrees C). Best results were achieved with a silica composition of 2.6% (w/w). We propose that silica inhibits the conduction of methanol through Nafion by blocking sites necessary for methanol diffusion through the polymer electrolyte membrane.
