    Genetic Divergence Among Accessions Of Cassava (manihot Esculenta Crantz) Sampled In The Tapajós Region, State Of Pará, Using Agronomic Characters And Microsatellite Markers

    The aim of this work was to estimate the genetic divergence among accessions of cassava sampled in the Tapajós region in the State of Pará, Brazil, and conserved at the Regional Germplasm Bank of Eastern Amazon, using agronomic descriptors and molecular markers. Twenty-two accessions of cassava were evaluated in the field for two successive years, based on six agronomic descriptors in twelve-months-old plants without a specific experimental design. Accessions were also evaluated with eleven microsatellite loci in an automatic DNA analyser. Descriptive and multivariate statistical analyses were applied. Based on principal components analysis, the character weight of the aerial portion of the plant contributed most to the phenotypical variation. The six traits were used in the analysis of genetic dissimilarity between accessions, and the correlation between matrices generated by morphological and molecular data was estimated. The matrices of genetic dissimilarity were used in the construction of dendrograms using the UPGMA method. We observed a high variation of agronomical descriptors and molecular markers evaluated, which were capable to separate the accessions into distinct groups. A weak positive correlation was detected among the two matrices of genetic distances, which indicates the possibility to explore the genetic diversity using crossings and accessions Amarelinha 36 and Olho roxo 13 are divergent and potentially promising for the generation of heterotic hybrids.3752989300

    Whole genome sequencing of Shigella sonnei through PulseNet Latin America and Caribbean: advancing global surveillance of foodborne illnesses

    Objectives Shigella sonnei is a globally important diarrhoeal pathogen tracked through the surveillance network PulseNet Latin America and Caribbean (PNLA&C), which participates in PulseNet International. PNLA&C laboratories use common molecular techniques to track pathogens causing foodborne illness. We aimed to demonstrate the possibility and advantages of transitioning to whole genome sequencing (WGS) for surveillance within existing networks across a continent where S. sonnei is endemic. Methods We applied WGS to representative archive isolates of S. sonnei (n = 323) from laboratories in nine PNLA&C countries to generate a regional phylogenomic reference for S. sonnei and put this in the global context. We used this reference to contextualise 16 S. sonnei from three Argentinian outbreaks, using locally generated sequence data. Assembled genome sequences were used to predict antimicrobial resistance (AMR) phenotypes and identify AMR determinants. Results S. sonnei isolates clustered in five Latin American sublineages in the global phylogeny, with many (46%, 149 of 323) belonging to previously undescribed sublineages. Predicted multidrug resistance was common (77%, 249 of 323), and clinically relevant differences in AMR were found among sublineages. The regional overview showed that Argentinian outbreak isolates belonged to distinct sublineages and had different epidemiologic origins. Conclusions Latin America contains novel genetic diversity of S. sonnei that is relevant on a global scale and commonly exhibits multidrug resistance. Retrospective passive surveillance with WGS has utility for informing treatment, identifying regionally epidemic sublineages and providing a framework for interpretation of prospective, locally sequenced outbreaks

    Salinomycin and virginiamycin for lactating cows supplemented on pasture

    Animals on pasture generally show higher feed efficiency as a result of the use of antibiotics. This study evaluated the effect of the antimicrobials salinomycin and/or virginiamycin on production and the ruminal parameters of supplemented dairy cows grazing on Panicum maximum cv. Tanzania. Twelve Holstein/Zebu multiparous cows were used, distributed in three Latin squares, one for the evaluation of ruminal parameters, and the others for production parameters. Cows on pasture were fed 50 % of their estimated intake with corn silage and concentrate supplements containing salinomycin, virginiamycin or a combination of additives, in doses of 120 and 150 mg kg−1, respectively. There were no differences in milk production and composition, energy and nitrogen balance, dry matter digestibility and feeding behavior. However, salinomycin and virginiamycin each reduced pasture and total dry matter intake by about 14 % and 10 %, with a consequent improvement in feed efficiency

    Variabilidade genética e fluxo gênico em populações híbridas e silvestres de pupunha acessada com marcadores RAPD

    As populações híbridas de pupunha (Bactris gasipaes Kunth) acumularam variabilidade genética provenientes de raças primitivas ao seu redor, o que deveria aumentar sua variabilidade. Para testar esta hipótese, avaliou-se a variabilidade genética de populações híbridas por meio de marcadores RAPD utilizando 176 plantas mantidas no Banco Ativo de Germoplasma do INPA, Manaus-AM, sendo quatro populações híbridas [Belém (n=26); Manaus (n=38); Iquitos, Peru (n=41); Yurimáguas, Peru (n=41)], duas populações silvestres (B. gasipaes variedade chichagui) tipos 1 (n=21) e 3 (n=7), e duas amostras de espécie afim, B. riparia, e compararam-se os parâmetros genéticos com estudos das raças primitivas. Oito iniciadores RAPD geraram 88 marcadores polimórficos e 11 monomórficos. O teste de replicabilidade apresentou uma similaridade de Dice 0,67, considerado aceitável. A heterozigosidade média das populações híbridas foi 0,34 e o polimorfismo foi 87,9%, maiores que nas silvestres (0,31; 74,7%). O dendrograma das similaridades de Dice não apresentou grupos que representassem claramente as populações híbridas. O fluxo gênico entre Iquitos e Yurimáguas (Nm=12,75) e entre Iquitos e Manaus (Nm=9,47) foi alto, enquanto o fluxo entre Belém e Manaus (Nm=7,72) foi menor que o esperado, possivelmente devido à influência da raça Solimões. O alto valor de heterozigosidade em Manaus (0,31) parece ser resultado da união de duas dispersões após a domesticação: a do oeste amazônico, com Iquitos e Yurimáguas, e a do leste amazônico, com Belém, que se juntam em Manaus. No entanto, essas populações não apresentaram acúmulo de variabilidade genética tão expressiva para diferenciá-las das raças primitivas
