    Metabolic effects of diets differing in glycaemic index depend on age and endogenous GIP

    Aims/hypothesis High- vs low-glycaemic index (GI) diets unfavourably affect body fat mass and metabolic markers in rodents. Different effects of these diets could be age-dependent, as well as mediated, in part, by carbohydrate-induced stimulation of glucose-dependent insulinotrophic polypeptide (GIP) signalling. Methods Young-adult (16 weeks) and aged (44 weeks) male wild-type (C57BL/6J) and GIP-receptor knockout (Gipr −/− ) mice were exposed to otherwise identical high-carbohydrate diets differing only in GI (20–26 weeks of intervention, n = 8–10 per group). Diet-induced changes in body fat distribution, liver fat, locomotor activity, markers of insulin sensitivity and substrate oxidation were investigated, as well as changes in the gene expression of anorexigenic and orexigenic hypothalamic factors related to food intake. Results Body weight significantly increased in young-adult high- vs low-GI fed mice (two-way ANOVA, p < 0.001), regardless of the Gipr genotype. The high-GI diet in young-adult mice also led to significantly increased fat mass and changes in metabolic markers that indicate reduced insulin sensitivity. Even though body fat mass also slightly increased in high- vs low-GI fed aged wild-type mice (p < 0.05), there were no significant changes in body weight and estimated insulin sensitivity in these animals. However, aged Gipr −/− vs wild-type mice on high-GI diet showed significantly lower cumulative net energy intake, increased locomotor activity and improved markers of insulin sensitivity. Conclusions/interpretation The metabolic benefits of a low-GI diet appear to be more pronounced in younger animals, regardless of the Gipr genotype. Inactivation of GIP signalling in aged animals on a high-GI diet, however, could be beneficial

    Week 96 efficacy and safety results of the phase 3, randomized EMERALD trial to evaluate switching from boosted-protease inhibitors plus emtricitabine/tenofovir disoproxil fumarate regimens to the once daily, single-tablet regimen of darunavir/cobicistat/emtricitabine/tenofovir alafenamide (D/C/F/TAF) in treatment-experienced, virologically-suppressed adults living with HIV-1

    Darunavir/cobicistat/emtricitabine/tenofovir alafenamide (D/C/F/TAF) 800/150/200/10 mg was investigated through 96 weeks in EMERALD (NCT02269917). Virologically-suppressed, HIV-1-positive treatment-experienced adults (previous non-darunavir virologic failure [VF] allowed) were randomized (2:1) to D/C/F/TAF or boosted protease inhibitor (PI) plus emtricitabine/tenofovir-disoproxil-fumarate (F/TDF) over 48 weeks. At week 52 participants in the boosted PI arm were offered switch to D/C/F/TAF (late-switch, 44 weeks D/C/F/TAF exposure). All participants were followed on D/C/F/TAF until week 96. Efficacy endpoints were percentage cumulative protocol-defined virologic rebound (PDVR; confirmed viral load [VL] >= 50 copies/mL) and VL = 50 copies/mL (VF) (FDA-snapshot analysis). Of 1141 randomized patients, 1080 continued in the extension phase. Few patients had PDVR (D/C/F/TAF: 3.1%, 24/763 cumulative through week 96; late-switch: 2.3%, 8/352 week 52-96). Week 96 virologic suppression was 90.7% (692/763) (D/C/F/TAF) and 93.8% (330/352) (late-switch). VF was 1.2% and 1.7%, respectively. No darunavir, primary PI, tenofovir or emtricitabine resistance-associated mutations were observed post-baseline. No patients discontinued for efficacy-related reasons. Few discontinued due to adverse events (2% D/C/F/TAF arm). Improved renal and bone parameters were maintained in the D/C/F/TAF arm and observed in the late-switch arm, with small increases in total cholesterol/high-density-lipoprotein-cholesterol ratio. A study limitation was the lack of a control arm in the week 96 analysis. Through 96 weeks, D/C/F/TAF resulted in low PDVR rates, high virologic suppression rates, very few VFs, and no resistance development. Late-switch results were consistent with D/C/F/TAF week 48 results. EMERALD week 96 results confirm the efficacy, high genetic barrier to resistance and safety benefits of D/C/F/TAF

    Voting and Civic Engagement Among Utah Women: A 2021 Update

    In 2016, the Utah Women & Leadership Project (UWLP) released a research snapshot called “Voting and Civic Engagement Among Utah Women.” At the time, despite having a strong history of women’s political and civic involvement, Utah had fallen to the bottom of national rankings in terms of women’s political participation. In 2019, UWLP released an update of this report, which showed some improvement in voter participation rates for both the United States and for Utah women. Since 2016, the level of political engagement among women in Utah and the United States has grown, and civic engagement—a critical element of change for municipalities, counties, and states—has remained high. Importantly, Kamala Harris was elected the first female vice president, which made 2020 a historic year for women in the US. Additionally, Utah elected former state Senator Deidre Henderson its second female lieutenant governor

    Catch-effort estimation of white-tailed deer population size

    Estimation of population size is important for most research in population biology and in the management of game species. Using a stochastic, catch-effort, competing risks model (Dupont 1983), we estimated the population size of the Savannah River Site white-tailed deer (Odocotleus virginianus) herd for 1965-86. Population size varied markedly in response to changes in both hunting method and pressure. Still hunters preferentially harvested older animals compared to dog hunters. Deer were 2.37 times more susceptible to harvest from dog hunting than from still hunting. Hunter-induced mortality was estimated as 1.73 and 4.10 times as large as nonhunting mortality for still and dog hunting, respectively. The temporal pattern of estimated prehunt population sizes was significantly correlated with the temporal pattern of car-deer accidents recorded on the site during the same time period, suggesting that the temporal pattern of the population estimates is accurate. If the number of cohorts is large and an accurate estimate of hunter effort can be obtained, this technique may provide more reliable population estimates than previously available techniques because it imposes fewer and less stringent biological assumptions

    Experiential learning and the acquisition of managerial tacit knowledge

    Tacit knowledge is believed to be one factor that distinguishes successful managers from others. We sought to determine whether levels of accumulated managerial tacit knowledge (LAMTK) were associated with managers' dominant learning styles. Instruments used in the study, involving 356 Malaysian public sector employees, included Sternberg et al.'s (2000) Tacit Knowledge Inventory for Managers and a normative version of Kolb's (1999a) Learning Styles Inventory (LSI-Ill). Findings suggest that LAMTK is independent of the length of subjects' general work experience, but positively related to the amount of time spent working in a management context. Learning styles also had a significant relationship. Subjects who spent most of their time performing management functions and whose dominant learning styles were accommodating had significantly higher LAMTK than those with different learning styles. We also found support for the belief that learners with a strong preference for all four different abilities defined in Kolb's learning theory may be critical for effective experiential learning

    The influence of learning styles on knowledge acquisition in public sector management

    This research note outlines a project designed to investigate the role of training institutions in providing effective training and development programmes for managers. The investigation is being carried out in the light of recent criticisms levelled against the nature of formal learning environments prevalent in most institutional settings. The traditional role of trainers and developers as the providers of knowledge and skills for the development of competent managers runs contrary to recent findings, which suggest that managers learn more effectively in informal settings, rather than the formal settings evident in many development programmes. The idea that explicitly extracted competencies are the target every manager should aim for to improve their effectiveness is also challenged because competencies alone are no longer regarded as a sufficient criterion for success. Recent research has attached greater importance to the need for helping managers to see knowledge as a social phenomenon, and one factor that might distinguish successful managers from others is tacit knowledge (Wagner & Sternberg, 1987; Argyris, 1999). A major focus of this study is to explore the possibility that the level and content of tacit knowledge acquired by managers may be influenced by their individual learning styles, and the degree to which their dominant styles are matched with the context of their work environment

    Exposure age and climate controls on weathering in deglaciated watersheds of western Greenland

    Fine-grained sediments deposited by retreating glaciers weather faster than the global average and this weathering can impact the global carbon cycle and oceanic fluxes of nutrients and radiogenic isotopes. Much work has focused on subglacial and proglacial weathering of continental ice sheets, but little is known about weathering and resulting fluxes from deglacial watersheds, which are disconnected from the ice sheets and discharge only annual precipitation and permafrost melt. We investigate the effects of exposure age and precipitation on weathering intensity in four deglacial watersheds on Greenland that form a transect from the coast near Sisimiut toward the Greenland Ice Sheet (GrIS) near Kangerlussuaq based on evaluations of major ion compositions, Sr isotope ratios, and mineral saturation states of waters and sediments. The transect is underlain by Archean orthogneiss and is characterized by gradients in moraine ages (∼7.5–8.0 ky inland to ∼10 ky at the coast) and water balance (−150 mm/yr inland to +150 mm/yr at the coast). Anion compositions are generally dominated by HCO3, but SO4 becomes increasingly important toward the coast, reflecting a switch from trace carbonate dissolution to sulfide mineral oxidation. Coastal watersheds have a higher proportion of dissolved silica, higher Na/Cl, Si/Ca, and lower Ca/Sr ratios than inland watersheds, indicating an increase in the relative proportion of silicate weathering and an increase in the extent of weathering toward the coast. More extensive weathering near the coast is also apparent in differences in the 87Sr/86Sr ratios of stream water and bedload (Δ87Sr/86Sr), which decreases from 0.017 inland to 0.005 at the coast, and in increased saturation states relative to amorphous SiO2 and quartz. The steep weathering gradient from inland to coastal watersheds reflects enhanced weathering compared to that expected from the 2 to 3 ky difference in exposure age caused by elevated coastal precipitation. The gradient of weathering with exposure age, water budget and distance from the ice sheet indicates that oceanic and atmospheric fluxes will change as continental glaciers retreat, precipitation patterns across the deglacial region readjust, and the relative proportion of deglacial to proglacial runoff increases
