    Kinematic characteristics of hailstorms in Northern Greece

    International audienceThe purpose of this study is the analysis of radar data, digitally recorded, during an operational hail suppression program in the region of Central Macedonia, Greece, for the warm period of the years 1997?2001. Kinematic characteristics, such as lifetime and distance traveled by hailstorms, as well as direction of motion and speed, have been related to type of storms and season. It has been found that singlecells are short-lived and travel short distances, while multicells are long-lived and travel long distances. On the contrary, their corresponding speed distributions are similar. The deviation of the direction of motion from mean wind is smaller for singlecells than for multicells. September and July exhibit the maximum and minimum average storm speeds as a direct implication of synoptic disturbances passage and convection, prevailing respectively. Finally, storms overcoming orographic barriers decelerate in general on the windward side and accelerate on the lee side of mountains

    Mutations in matrix and SP1 repair the packaging specificity of a Human Immunodeficiency Virus Type 1 mutant by reducing the association of Gag with spliced viral RNA

    <p>Abstract</p> <p>Background</p> <p>The viral genome of HIV-1 contains several secondary structures that are important for regulating viral replication. The stem-loop 1 (SL1) sequence in the 5' untranslated region directs HIV-1 genomic RNA dimerization and packaging into the virion. Without SL1, HIV-1 cannot replicate in human T cell lines. The replication restriction phenotype in the SL1 deletion mutant appears to be multifactorial, with defects in viral RNA dimerization and packaging in producer cells as well as in reverse transcription of the viral RNA in infected cells. In this study, we sought to characterize SL1 mutant replication restrictions and provide insights into the underlying mechanisms of compensation in revertants.</p> <p>Results</p> <p>HIV-1 lacking SL1 (NLΔSL1) did not replicate in PM-1 cells until two independent non-synonymous mutations emerged: G913A in the matrix domain (E42K) on day 18 postinfection and C1907T in the SP1 domain (P10L) on day 11 postinfection. NLΔSL1 revertants carrying either compensatory mutation showed enhanced infectivity in PM-1 cells. The SL1 revertants produced significantly more infectious particles per nanogram of p24 than did NLΔSL1. The SL1 deletion mutant packaged less HIV-1 genomic RNA and more cellular RNA, particularly signal recognition particle RNA, in the virion than the wild-type. NLΔSL1 also packaged 3- to 4-fold more spliced HIV mRNA into the virion, potentially interfering with infectious virus production. In contrast, both revertants encapsidated 2.5- to 5-fold less of these HIV-1 mRNA species. Quantitative RT-PCR analysis of RNA cross-linked with Gag in formaldehyde-fixed cells demonstrated that the compensatory mutations reduced the association between Gag and spliced HIV-1 RNA, thereby effectively preventing these RNAs from being packaged into the virion. The reduction of spliced viral RNA in the virion may have a major role in facilitating infectious virus production, thus restoring the infectivity of NLΔSL1.</p> <p>Conclusions</p> <p>HIV-1 evolved to overcome a deletion in SL1 and restored infectivity by acquiring compensatory mutations in the N-terminal matrix or SP1 domain of Gag. These data shed light on the functions of the N-terminal matrix and SP1 domains and suggest that both regions may have a role in Gag interactions with spliced viral RNA.</p

    Epidemiology of Patent Foramen Ovale in General Population and in Stroke Patients: A Narrative Review

    Introduction: Percutaneous closure of patent foramen ovale (PFO) in selected patients with cryptogenic cerebrovascular ischemic events (CEs) decreases the risk of recurrent stroke; however, optimal patient selection criteria are still under investigation. Candidates for PFO closure are usually selected from the pool of CE patients with a high risk of Paradoxical Embolism (RoPE) score. The RoPE score calculates the probability that PFO is causally related to stroke, based on PFO prevalence in patients with CE compared with that in healthy subjects. The latter has been set at 25% based on the average of autopsy and transesophageal echocardiography (TEE) studies. Methods: We conducted a comprehensive review of studies investigating PFO prevalence in general population and in patients with CE and non-CE using autopsy, TEE, transcranial Doppler (TCD) or transthoracic echocardiography (TTE). Studies were excluded if they (1) reported data from referred subjects with underlying cerebrovascular disease or (2) did not specify etiologically the events. Results: In healthy/control subjects, PFO prevalence was 24.2% (1,872/7,747) in autopsy studies, 23.7% (325/1,369) in TEE, 31.3% (111/355) in TCD, and 14.7% (186/1,267) in TTE studies. All diagnostic modalities included PFO prevalence was higher in CE compared with healthy/control population [odds ratio (OR) = 3.1, 95% confidence interval (CI) = 2.5–3.8] and compared with non-CE (OR = 2.3, 95% CI = 2.0–2.6). In patients with CE, PFO prevalence in the young compared to the old was higher when the diagnostic modality was TEE (48.9 vs. 27.3%, p &lt; 0.0001, OR = 2.6 with 95% CI = 2.0–3.3) or TCD (58.1 vs. 41%, OR = 1.9, 95% CI = 1.6–2.5), but not TTE (53.3 vs. 37.5%, p = 0.16). Regarding non-CE, PFO prevalence in the young compared to the old was higher when the diagnostic modality was TEE (20 vs. 12.9%, OR = 1.7, 95% CI = 1.0–2.8) but not TTE (10.4 vs. 7.8%, p = 0.75) or TCD (22.8 vs. 20.1%, p = 0.56). Conclusions: Given the limitations of autopsy and TEE studies, there is good reason not to take a fixed 25% PFO prevalence for granted. The estimation of degree of causality may be underestimated or overestimated in populations with PFO prevalence significantly lower or higher than the established. Given the high sensitivity, non-invasive nature, low cost, and repeatability of TCD, future large-scale TCD-based studies should investigate potential heterogeneity in PFO prevalence in different healthy racial/ethnic populations. © Copyright © 2020 Koutroulou, Tsivgoulis, Tsalikakis, Karacostas, Grigoriadis and Karapanayiotides

    A deep stratospheric intrusion event down to the earth&apos;s surface of the megacity of Athens

    This case study investigates a stratospheric intrusion event down to the earth&apos;s surface (near sea-level pressure) of the greater area of Athens (23. 43°E 37. 58°N), which occurred on 9 October 2003 and caused a remarkable increase in surface ozone concentrations not related to photochemical production. This event is among the rare case studies investigating, on the one hand, a deep stratospheric intrusion down to the earth&apos;s surface at near sea-level pressure and, on the other, an event affecting the near surface ozone of a megacity such as Athens. The synoptic situation is described by a deep upper lever trough at 300 and 500 hPa extending over Greece, which is related to a deep tropopause fold as revealed by vertical cross sections of potential vorticity, relative humidity, divergence and vertical velocity. The analysis of potential vorticity at several isentropic levels indicates a hook-shaped streamer of high PV values (greater than 4 pvu at the 315 K isentropic level) over southeast Europe, which coincides with a streamer of dry air as observed from satellite images of water vapor. The aforementioned structure characterizes a textbook case study of stratosphere-to-troposphere transport. The Lagrangian particle dispersion model FLEXPART was used to calculate the trajectories of air particles reaching the receptor site and the fraction of particles with stratospheric origin. It reveals an important direct stratospheric impact within 1 day related to the tropopause fold described in this study with the fraction of stratospheric particles reaching maximum values of 1.9 and 4.5% for threshold values of the dynamical tropopause 2 and 1.5 pvu, respectively. Furthermore, a larger indirect aged stratospheric contribution is also revealed 4 to 5 days prior to the release, related to stratospheric intrusion events at the western Atlantic Ocean, reaching maximum values of 2.5 and 6.9% of particles crossing the 2 and 1.5 pvu potential vorticity surfaces, respectively. © 2010 Springer-Verlag

    Prevalence of patent foramen ovale in the Greek population is high and impacts on the interpretation of the risk of paradoxical embolism (RoPE) score

    Background: The risk of paradoxical embolism (RoPE) score calculates the probability that patent foramen ovale (PFO) is causally related to stroke (PFO attributable fraction, PFOAF), based on PFO prevalence in patients with cryptogenic stroke (CS) compared with that in the general population. The latter has been estimated at 25%; however, PFO prevalence in nonselected populations varies widely. Methods: Since PFO prevalence in Greece remains unknown, we evaluated it and we calculated PFOAF stratified by RoPE score in a cohort of patients with CS ⩽55 years old. PFO was detected according to the international consensus transcranial Doppler (TCD) criteria in 124 healthy subjects (H), in 102 patients with CS, and in 56 patients with stroke of known cause (nonCS). Each subject underwent unilateral middle cerebral artery recording after infusion of agitated saline, at rest, and after a controlled Valsalva maneuver. We characterized PFO as large (&gt;20 microbubbles or curtain), moderate (11–20), and small (⩽10). Results: PFO was detected in 42.7% of H, 49% of CS, and 25% of nonCS (p = 0.013). Large PFOs were numerically higher in CS [28.4% (29/102)] compared with H [19.3% (24/124); p = 0.1] and to nonCS [7.1% (4/56), p = 0.04]. The median RoPE score in patients with CS and PFO was seven. Even patients with very high RoPE score (9–10) had moderate PFOAF (57%). For any individual stratum up to RopE score 8, PFOAF was &lt;33%. Conclusions: PFO prevalence in the Greek population is much higher than the widely accepted 25%. PFO may be the cause of stroke in one out of nine Greek patients with CS. Among Greek CS patients who harbor a PFO, the latter is causal in one out of five. The established RoPE score cutoff of ⩾7 for having a probable PFO-associated stroke may overestimate the probability in patients deriving from populations with high PFO prevalence. © The Author(s), 2020

    Fog characteristics at the airport of Thessaloniki, Greece

    A statistical approach in order to study fog event characteristics occuring at the airport of Thessaloniki, Northern Greece is presented in this work. To achieve this, the seasonal and diurnal characteristics of fog are investigated using 35 years (1971–2005) of observations of meteorological parameters such as: visibility, air temperature, dew point temperature, air relative humidity, wind vector, precipitation, and cloud base height and coverage. Hourly surface observations of fifteen (15) years (1991–2005) are used to identify fog events induced by various physical mechanisms. Fog events are classified into fog types through the application of objective criteria that are derived upon fog formation processes and under the influence of various physiographic features. The temporal variability of different fog type occurrences are examined and the events are characterized according to their duration and intensity. &lt;br&gt;&lt;br&gt; The results are somehow affected by regional and local factors. Fog is mainly formed in winter time (~64%) with an enhanced likelihood to appear also in late autumn (19%). The highest frequency of the fog events occurs around sunrise or 1 to 2 h before of it. The mean duration of the events is about 4.5 h. Most of them (75%) are dense (visibility &amp;lt;400 m). The overall fog phenomenon is a combination of various types, such as: advection fog, radiation fog, cloud-base lowering fog and precipitation fog. Advection fog (30%) and radiation fog (29%), which are the most common types, occur predominantly in winter and early spring time. With respect to the former type, it seems that, in many cases, already formed fog is advected from the nearby &lt;i&gt;Anthemountas&lt;/i&gt; valley. Moreover, a considerable number of fog events (22%) result from cloud-base lowering, and they frequently occur in late autumn and mid winter
