Secretion Kinetics of Salmonella Effector Proteins and Their Homologs
The Type III Secretion System (T3SS) is a needle-like assembly that bacteria such as Salmonella use to put their infectious proteins in target cells. The proteins that are put through the T3SS are the focus of this study because they have an interesting evolutionary pressure. The T3SS is one amino acid wide, so any proteins that go through it must be unfolded before being excreted. This project studies how the proteins have evolved to be unfolded. Whether they have evolved to be mechanically labile or not. This study is done by putting a tag on a protein that is normally excreted by the T3SS and a tag on a protein that is homologous to the first, but is not normally excreted by the T3SS. The tag is able to go through the T3SS and will bind a fluorescent molecule that is membrane-permeable. The end-goal is to do an infection assay where the proteins will be highly concentrated proteins within the bacterial cells, causing a large fluorescence reading. When the proteins are excreted into the mammalian cells, their fluorescence readings will decrease because they become less concentrated. This decrease in fluorescence will be related to the secretion rate of the proteins. Based on those readings, it can be concluded whether or not the traditionally secreted proteins are more easily excreted compared to the non-traditionally secreted ones.</p
Identifying a Test to Monitor Weightlifting Performance in Competitive Male and Female Weightlifters
Monitoring tests are commonly used to assess weightlifterâs preparedness for competition. Although various monitoring tests have been used, it is not clear which test is the strongest indicator of weightlifting performance. Therefore, the purpose of this study was to (1) determine the relationships between vertical jump, isometric mid-thigh pull (IMTP) and weightlifting performance; and (2) compare vertical jumps to IMTP as monitoring tests of weightlifting performance in a large cohort of male and female weightlifters. Methods: Fifty-two competitive weightlifters (31 males, 21 females) participated in squat and countermovement jump testing (SJ, CMJ), and IMTP testing performed on force plates. All laboratory testing data was correlated to a recent competition where the athletes had attempted to peak. Results: Squat jump height (SJH) was the strongest correlate for men and women with the Sinclair Total (r = 0.686, p ⤠0.01; r = 0.487, p ⤠0.05, respectively) compared to countermovement jump height (r = 0.642, p ⤠0.01; r = 0.413, p = 0.063), IMTP peak force allometrically scaled to body mass (r = 0.542, p ⤠0.01; r = â0.044, p = 0.851) and rate of force development at 200 ms (r = 0.066, p = 0.723; r = 0.086, p = 0.711), respectively. Further, SJH was a stronger correlate of relative weightlifting performance compared to IMTP peak force in females (p = 0.042), but not male weightlifters (p = 0.191). Conclusions: Although CMJ and IMTP are still considered strong indicators of weightlifting performance, SJH appears to be the most indicative measure of weightlifting performance across a wide-range of performance levels. Thus, SJH can be used as a reliable measure to monitor weightlifting performance in male and female weightlifters

West Nile virus growth is independent of autophagy activation
AbstractWest Nile virus (WNV) is an arthropod-borne virus with a worldwide distribution that causes neurologic disease and death. Autophagy is a cellular homeostatic mechanism involved in antiviral responses but can be subverted to support viral growth as well. We show that autophagy is induced by WNV infection in cell culture and in primary neuron cultures. Following WNV infection, lysosomes co-localize with autophagosomes resulting in LC3B-II turnover and autolysosomal acidification. However, activation or inhibition of autophagy has no significant effect on WNV growth but pharmacologic inhibition of PI3 kinases associated with autophagy reduce WNV growth. Basal levels of p62/sequestosome1(SQSTM1) do not significantly change following WNV-induced autophagy activation, but p62 is turned over or degraded by autophagy activation implying that p62 expression is increased following WNV-infection. These data show that WNV-induces autophagy but viral growth is independent of autophagy activation suggesting that WNV-specific interactions with autophagy have diverged from other flaviviruses
Force Output Comparison between Six U.S. Collegiate Athletic Teams.
The aim of the study was to compare allometrically scaled peak force and the force at 250 ms between six U.S. collegiate sport teams using isometric mid-thigh pull. Ninety subjects performed maximum effort of isometric mid-thigh pull to measure force output. The data were averaged within the teams, and statistically compared between teams using one-way ANOVA (p=.01). Significant difference was found that menâs soccer and baseball produced higher allometrically scaled peak force, and menâs soccer, tennis, and baseball produced higher allometrically scaled force at 250 ms. The data indicates that not all sports possess similar strength characteristics because of the nature of the sports, and observed separation between gender. Teams such as volleyball and baseball showed higher coefficient of variation due to the various positions within their sports
"After my husband's circumcision, I know that I am safe from diseases": Women's Attitudes and Risk Perceptions Towards Male Circumcision in Iringa, Tanzania.
While male circumcision reduces the risk of female-to-male HIV transmission and certain sexually transmitted infections (STIs), there is little evidence that circumcision provides women with direct protection against HIV. This study used qualitative methods to assess women's perceptions of male circumcision in Iringa, Tanzania. Women in this study had strong preferences for circumcised men because of the low risk perception of HIV with circumcised men, social norms favoring circumcised men, and perceived increased sexual desirability of circumcised men. The health benefits of male circumcision were generally overstated; many respondents falsely believed that women are also directly protected against HIV and that the risk of all STIs is greatly reduced or eliminated in circumcised men. Efforts to engage women about the risks and limitations of male circumcision, in addition to the benefits, should be expanded so that women can accurately assess their risk of HIV or STIs during sexual intercourse with circumcised men

Iron is essential for life. Accessing iron from the environment can be a limiting factor that determines success in a given environmental niche. For bacteria, access of chelated iron from the environment is often mediated by TonB-dependent transporters (TBDTs), which are β-barrel proteins that form sophisticated channels in the outer membrane. Reports of iron-bearing proteins being used as a source of iron indicate specific protein import reactions across the bacterial outer membrane. The molecular mechanism by which a folded protein can be imported in this way had remained mysterious, as did the evolutionary process that could lead to such a protein import pathway. How does the bacterium evolve the specificity factors that would be required to select and import a protein encoded on another organism's genome? We describe here a model whereby the plant iron-bearing protein ferredoxin can be imported across the outer membrane of the plant pathogen Pectobacterium by means of a Brownian ratchet mechanism, thereby liberating iron into the bacterium to enable its growth in plant tissues. This import pathway is facilitated by FusC, a member of the same protein family as the mitochondrial processing peptidase (MPP). The Brownian ratchet depends on binding sites discovered in crystal structures of FusC that engage a linear segment of the plant protein ferredoxin. Sequence relationships suggest that the bacterial gene encoding FusC has previously unappreciated homologues in plants and that the protein import mechanism employed by the bacterium is an evolutionary echo of the protein import pathway in plant mitochondria and plastids
