    Widely tunable, non-degenerate three-wave mixing microwave device operating near the quantum limit

    We present the first experimental realization of a widely frequency tunable, non-degenerate three-wave mixing device for quantum signals at GHz frequency. It is based on a new superconducting building-block consisting of a ring of four Josephson junctions shunted by a cross of four linear inductances. The phase configuration of the ring remains unique over a wide range of magnetic fluxes threading the loop. It is thus possible to vary the inductance of the ring with flux while retaining a strong, dissipation-free, and noiseless non-linearity. The device has been operated in amplifier mode and its noise performance has been evaluated by using the noise spectrum emitted by a voltage biased tunnel junction at finite frequency as a test signal. The unprecedented accuracy with which the crossover between zero-point-fluctuations and shot noise has been measured provides an upper-bound for the noise and dissipation intrinsic to the device.Comment: Accepted for Physical Review Letters.     DSS-28: a novel wide bandwidth radio telescope devoted to educational outreach

    We have recently equipped the 34-meter DSS-28 radio telescope at the Goldstone Deep Space Communications Complex with a novel wide bandwidth radiometer and digital signal processor as part of the Goldstone Apple Valley Radio Telescope (GAVRT) educational outreach program operated by the Jet Propulsion Laboratory and the Lewis Center for Educational Research. The system employs a cryogenically cooled wide bandwidth quad-ridge feed and InP low noise amplifiers to achieve excellent noise performance from 2.7 to 14 GHz; a fractional bandwidth better than 4:1. Four independently tunable dual-polarization receivers each down-convert a 2 GHz block to baseband, providing access to 8 GHz of instantaneous bandwidth. A flexible FPGA-based signal processor has been constructed using CASPER FPGA hardware and tools to take advantage of this enormous bandwidth. This system demonstrates many of the enabling wide bandwidth technologies that will be crucial to maximizing the utility of future large centimeter-wavelength arrays, in particular the Square Kilometer Array. The GAVRT program has previously used narrow bandwidth total power radiometers to study flux variability of quasars and the outer planets. The versatility of DSS-28 will enable other projects including spectroscopy and SETI. Finally, the wide instantaneous bandwidth available makes this system uniquely suited for studying transient radio pulses. A configuration of the digital signal processor has been developed which provides the capability of recording a burst of raw baseband voltage data triggered by a real-time incoherent dedispersion system which is very sensitive to pulses from a known source, such as the Crab Nebula pulsar

    Topological Orthoalgebras

    We define topological orthoalgebras (TOAs) and study their properties. While every topological orthomodular lattice is a TOA, the lattice of projections of a Hilbert space is an example of a lattice-ordered TOA that is not a toplogical lattice. On the other hand, we show that every compact Boolean TOA is a topological Boolean algebra. We also show that a compact TOA in which 0 is an isolated point is atomic and of finite height. We identify and study a particularly tractable class of TOAs, which we call {\em stably ordered}: those in which the upper-set generated by an open set is open. This includes all topological OMLs, and also the projection lattices of Hilbert spaces.     Discovery of interstellar mercapto radicals (SH) with the GREAT instrument on SOFIA

    We report the first detection of interstellar mercapto radicals, obtained along the sight-line to the submillimeter continuum source W49N. We have used the GREAT instrument on SOFIA to observe the 1383 GHz Doublet Pi 3/2 J = 5/2 - 3/2 lambda doublet in the upper sideband of the L1 receiver. The resultant spectrum reveals SH absorption in material local to W49N, as well as in foreground gas, unassociated with W49N, that is located along the sight-line. For the foreground material at velocities in the range 37 - 44 km/s with respect to the local standard of rest, we infer a total SH column density ~ 2.6 E+12 cm-2, corresponding to an abundance of ~ 7 E-9 relative to H2, and yielding an SH/H2S abundance ratio ~ 0.13. The observed SH/H2S abundance ratio is much smaller than that predicted by standard models for the production of SH and H2S in turbulent dissipation regions and shocks, and suggests that the endothermic neutral-neutral reaction SH + H2 -> H2S + H must be enhanced along with the ion-neutral reactions believed to produce CH+ and SH+ in diffuse molecular clouds.Comment: Accepted for publication in Astronomy and Astrophysics (SOFIA/GREAT special issue

    Exosomal αvβ6 integrin is required for monocyte M2 polarization in prostate cancer

    Therapeutic approaches aimed at curing prostate cancer are only partially successful given the occurrence of highly metastatic resistant phenotypes that frequently develop in response to therapies. Recently, we have described αvβ6, a surface receptor of the integrin family as a novel therapeutic target for prostate cancer; this epithelial-specific molecule is an ideal target since, unlike other integrins, it is found in different types of cancer but not in normal tissues. We describe a novel αvβ6-mediated signaling pathway that has profound effects on the microenvironment. We show that αvβ6 is transferred from cancer cells to monocytes, including β6-null monocytes, by exosomes and that monocytes from prostate cancer patients, but not from healthy volunteers, express αvβ6. Cancer cell exosomes, purified via density gradients, promote M2 polarization, whereas αvβ6 down-regulation in exosomes inhibits M2 polarization in recipient monocytes. Also, as evaluated by our proteomic analysis, αvβ6 down-regulation causes a significant increase in donor cancer cells, and their exosomes, of two molecules that have a tumor suppressive role, STAT1 and MX1/2. Finally, using the Ptenpc−/− prostate cancer mouse model, which carries a prostate epithelial-specific Pten deletion, we demonstrate that αvβ6 inhibition in vivo causes up-regulation of STAT1 in cancer cells. Our results provide evidence of a novel mechanism that regulates M2 polarization and prostate cancer progression through transfer of αvβ6 from cancer cells to monocytes through exosomes

    Primary cilia signaling mediates intraocular pressure sensation

    Lowe syndrome is a rare X-linked congenital disease that presents with congenital cataracts and glaucoma, as well as renal and cerebral dysfunction. OCRL, an inositol polyphosphate 5-phosphatase, is mutated in Lowe syndrome. We previously showed that OCRL is involved in vesicular trafficking to the primary cilium. Primary cilia are sensory organelles on the surface of eukaryotic cells that mediate mechanotransduction in the kidney, brain, and bone. However, their potential role in the trabecular meshwork (TM) in the eye, which regulates intraocular pressure, is unknown. Here, we show that TM cells, which are defective in glaucoma, have primary cilia that are critical for response to pressure changes. Primary cilia in TM cells shorten in response to fluid flow and elevated hydrostatic pressure, and promote increased transcription of TNF-α, TGF-β, and GLI1 genes. Furthermore, OCRL is found to be required for primary cilia to respond to pressure stimulation. The interaction of OCRL with transient receptor potential vanilloid 4 (TRPV4), a ciliary mechanosensory channel, suggests that OCRL may act through regulation of this channel. A novel disease-causing OCRL allele prevents TRPV4-mediated calcium signaling. In addition, TRPV4 agonist GSK 1016790A treatment reduced intraocular pressure in mice; TRPV4 knockout animals exhibited elevated intraocular pressure and shortened cilia. Thus, mechanotransduction by primary cilia in TM cells is implicated in how the eye senses pressure changes and highlights OCRL and TRPV4 as attractive therapeutic targets for the treatment of glaucoma. Implications of OCRL and TRPV4 in primary cilia function may also shed light on mechanosensation in other organ systems
