Shedding light on the hidden benefit of Porphyridium cruentum culture
Microalgae can represent a reliable source of natural compounds with different activities.
Here, we evaluated the antioxidant and anti-inflammatory activity of sulfated exopolysaccharides
(s-EPSs) and phycoerythrin (PE), two molecules naturally produced by the red marine microalga
Porphyridium cruentum (CCALA415). In vitro and cell-based assays were performed to assess the
biological activities of these compounds. The s-EPSs, owing to the presence of sulfate groups, showed
biocompatibility on immortalized eukaryotic cell lines and a high antioxidant activity on cell-based
systems. PE showed powerful antioxidant activity both in vitro and on cell-based systems, but
purification is mandatory for its safe use. Finally, both molecules showed anti-inflammatory activity
comparable to that of ibuprofen and helped tissue regeneration. Thus, the isolated molecules from

Semantic gaps in the theory and the practice of physical and sports education in the Italian context
Nowadays, we face a profound fragmentation of knowledge, which is addressed in distinctive ways, conforming to the specifics of each field of knowledge and having a specific lexicon and ways to interpret reality. To better understand and communicate the complexity of the reality of motor and sports sciences in Italy, it is necessary to study its ontology based on a holistic and comprehensive approach such as System Thinking (ST). So, this preliminary review aimed to verify the coherence of terminology used by Italian academics (who study methodologies related to teaching) and practical communities (who live the reality of daily teaching) in motor and sports sciences, specifically regarding the didactic of physical and sports education. For this purpose, according to Design Based Research (DBR), a first investigation of the interpretative semantics of the disciplineâs specific terms was accomplished with the support of multiple sources of information such as documents, databases, and brainstorming performed by experts (representing the guiding team and stakeholders from all considered communities). Moreover, the frequency of some terms and keywords considered in European (ERC) and Italian (CUN) legislation was assessed to contextualize better the impact of common practice community keywords on the scientific and regulatory community. Successively, the studies that analyzed the terms from 2000 to 2020 in the leading scientific search engines (SCOPUS and Web of Science) were also investigated. Results indicate that in the CUN and ERC areas, there is a total absence of the most relevant lemma to the community of practice. Furthermore, significant terms such as play, sports education, psychomotricity, and gymnastics are less considered or unconsidered in the specific scientific and didactic fields. Possible solutions for a specific semantic model to reduce the gap are finally hypothesized and presented
Are the forearm muscles excited equally in different, professional piano players?
Background and objectives
Professional pianists tend to develop playing-related musculoskeletal disorders mostly in the forearm. These injuries are often due to overuse, suggesting the existence of a common forearm region where muscles are often excited during piano playing across subjects. Here we use a grid of electrodes to test this hypothesis, assessing where EMGs with greatest amplitude are more likely to be detected when expert pianists perform different excerpts.
Methods
Tasks were separated into two groups: classical excerpts and octaves, performed by eight, healthy, professional pianists. Monopolar electromyograms (EMGs) were sampled with a grid of 96 electrodes, covering the forearm region where hand and wrist muscles reside. Regions providing consistently high EMG amplitude across subjects were assessed with a non-parametric permutation test, designed for the statistical analysis of neuroimaging experiments. Spatial consistency across trials was assessed with the Binomial test.
Results
Spatial consistency of muscle excitation was found across subjects but not across tasks, confining at most 20% of the electrodes in the grid. These local groups of electrodes providing high EMG amplitude were found at the ventral forearm region during classical excerpts and at the dorsal region during octaves, when performed both at preferred and at high, playing speeds.
Discussion
Our results revealed that professional pianists consistently load a specific forearm region, depending on whether performing octaves or classical excerpts. This spatial consistency may help furthering our understanding on the incidence of playing-related muscular disorders and provide an anatomical reference for the study of active muscle loading in piano players using surface EMG
The XMM-Newton wide-field survey in the COSMOS field: V. Angular Clustering of the X-ray Point Sources
We present the first results of the measurements of angular auto-correlation
functions (ACFs) of X-ray point sources detected in the XMM-Newton observations
of the 2 deg2 COSMOS field (XMM-COSMOS). A significant positive signals have
been detected in the 0.5-2 (SFT) band, in the angle range of 0.5-24 arcminutes,
while the positive signals were at the 2 and 3 sigma levels in the 2-4.5 (MED)
and 4.5-10 (UHD) keV bands respectively. Correctly taking integral constraints
into account is a major limitation in interpreting our results. With power-law
fits to the ACFs without the integral constraint term, we find correlation
lengths of theta_c=1.9+-0.3, 0.8+0.5-0.4 and 6+-2 arcseconds for the SFT, MED,
and UHD bands respectively for gamma=1.8. The inferred comoving correlation
lengths, also taking into account the bias by the source merging due to
XMM-Newton PSF, are 9.8+-0.7, 5.8+1.4-1.7 and 12+-2 h-1 Mpc at the effective
redshifts of 1.1, 0.9, and 0.6 for the SFT, MED, and UHD bands respectively. If
we include the integral constraint term in the fitting process,assuming that
the power-law extends to the scale length of the entire XMM-COSMOSfield, the
correlation lengths become larger by 20%-90%. Comparing the inferred rms
fluctuations of the spatial distribution of AGNs with those of the underlying
mass, the bias parameters of the X-ray source clustering at these effective

Inside out porphyridium cruentum: Beyond the conventional biorefinery concept
Here, an unprecedented biorefinery approach has been designed to recover high-added value bioproducts starting from the culture ofPorphyridium cruentum. This unicellular marine red alga can secrete and accumulate high-value compounds that can find applications in a wide variety of industrial fields. 300 ± 67 mg/L of exopolysaccharides were obtained from cell culture medium; phycoerythrin was efficiently extracted (40% of total extract) and isolated by single chromatography, with a purity grade that allowed the crystal structure determination at 1.60 Ă
; a twofold increase in ÎČ-carotene yield was obtained from the residual biomass; the final residual biomass was found to be enriched in saturated fatty acids. Search for the optical counterpart of the GW170814 gravitational wave event with the VLT Survey Telescope
We report on the search for the optical counterpart of the gravitational event GW170814, which was carried out with the VLT Survey Telescope (VST) by the GRAvitational Wave Inaf TeAm. Observations started 17.5 h after the Laser Interferometer Gravitational-wave Observatory (LIGO) and Virgo alert and we covered an area of 99 deg2 that encloses ⌠77{{ per cent}} and ⌠59{{ per cent}} of the initial and refined localization probability regions, respectively. A total of six epochs were secured over nearly two months. The survey reached an average limiting magnitude of 22 AB mag in the r band. After assuming the model described in Perna, Lazzati & Farr, that derives as possible optical counterpart of a BBH (binary black hole) event a transient source declining in about one day, we have computed a survey efficiency of about 5{{ per cent}}. This paper describes the VST observational strategy and the results obtained by our analysis pipelines developed to search for optical transients in multi-epoch images. We report the catalogue of the candidates with possible identifications based on light-curve fitting. We have identified two dozens of SNe, nine AGNs, and one QSO. Nineteen transients characterized by a single detection were not classified. We have restricted our analysis only to the candidates that fall into the refined localization map. None out of 39 left candidates could be positively associated with GW170814. This result implies that the possible emission of optical radiation from a BBH merger had to be fainter than r ⌠22 (Loptical ⌠1.4 Ă 1042 erg s-1) on a time interval ranging from a few hours up to two months after the gravitational wave event
Lipoprotein(a) Genotype Influences the Clinical Diagnosis of Familial Hypercholesterolemia
Background Evidence suggests that LPA risk genotypes are a possible contributor to the clinical diagnosis of familial hypercholesterolemia (FH). This study aimed at determining the prevalence of LPA risk variants in adult individuals with FH enrolled in the Italian LIPIGEN (Lipid Transport Disorders Italian Genetic Network) study, with (FH/M+) or without (FH/M-) a causative genetic variant. Methods and ResultsAn lp(a) [lipoprotein(a)] genetic score was calculated by summing the number risk-increasing alleles inherited at rs3798220 and rs10455872 variants. Overall, in the 4.6% of 1695 patients with clinically diagnosed FH, the phenotype was not explained by a monogenic or polygenic cause but by genotype associated with high lp(a) levels. Among 765 subjects with FH/M- and 930 subjects with FH/M+, 133 (17.4%) and 95 (10.2%) were characterized by 1 copy of either rs10455872 or rs3798220 or 2 copies of either rs10455872 or rs3798220 (lp(a) score >= 1). Subjects with FH/M- also had lower mean levels of pretreatment low-density lipoprotein cholesterol than individuals with FH/M+ (t test for difference in means between FH/M- and FH/M+ groups <0.0001); however, subjects with FH/M- and lp(a) score >= 1 had higher mean (SD) pretreatment low-density lipoprotein cholesterol levels (223.47 [50.40] mg/dL) compared with subjects with FH/M- and lp(a) score=0 (219.38 [54.54] mg/dL for), although not statistically significant. The adjustment of low-density lipoprotein cholesterol levels based on lp(a) concentration reduced from 68% to 42% the proportion of subjects with low-density lipoprotein cholesterol level >= 190 mg/dL (or from 68% to 50%, considering a more conservative formula). ConclusionsOur study supports the importance of measuring lp(a) to perform the diagnosis of FH appropriately and to exclude that the observed phenotype is driven by elevated levels of lp(a) before performing the genetic test for FH
Towards green extraction methods from microalgae learning from the classics
Microalgae started receiving attention as producers of third generation of biofuel, but they are rich in many bioactive compounds. Indeed, they produce many molecules endowed with benefic effects on human health which are highly requested in the market. Thus, it would be important to fractionate algal biomass into its several high-value compounds: this represents the basis of the microalgal biorefinery approach. Usually, conventional extraction methods have been used to extract a single class of molecules, with many side effects on the environment and on human health. The development of a green downstream platform could help in obtaining different class of molecules with high purity along with low environmental impact. This review is focused on technical advances that have been performed, from classic methods to the newest and green ones. Indeed, it is fundamental to set up new procedures that do not affect the biological activity of the extracted molecules. A comparative analysis has been performed among the conventional methods and the new extraction techniques, i.e., switchable solvents and microwave-assisted and compressed fluid extractions.</p
Biosynthesis of Silver Nanoparticles Using Salvia pratensis L. Aerial Part and Root Extracts: Bioactivity, Biocompatibility, and Catalytic Potential
The aim of this research was the synthesis of silver nanoparticles (SPA- and SPR-AgNPs) using the aqueous extracts of the aerial (SPA) and the root (SPR) parts of the plant Salvia pratensis L., their characterization, reaction condition optimization, and evaluation of their biological and catalytic activity. UV-Vis spectroscopy, X-ray powder diffraction (XRPD), scanning electron microscopy with EDS analysis (SEM/EDS), and dynamic light scattering (DLS) analysis were utilized to characterize the nanoparticles, while Fourier transform infrared (FTIR) spectroscopy was used to detect some functional groups of compounds present in the plant extracts and nanoparticles. The phenolic and flavonoid contents, as well as the antioxidant activity of the extracts, were determined spectrophotometrically. The synthesized nanoparticles showed twice-higher activity in neutralizing 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+) compared with the respective extracts. SPR-AgNPs exhibited strong antimicrobial activity against almost all of the tested bacteria (<0.0039 mg/mL) and fungal strains, especially against the genus Penicillium (<0.0391 mg/mL). Moreover, they were fully biocompatible on all the tested eukaryotic cells, while the hemolysis of erythrocytes was not observed at the highest tested concentration of 150 ”g/mL. The catalytic activity of nanoparticles toward Congo Red and 4-nitrophenol was also demonstrated. The obtained results confirm the possibility of the safe application of the synthesized nanoparticles in medicine and as a catalyst in various processes
