Dynamic additional loads influencing the fatigue life of gears in an electric vehicle transmission

Get PDF
In recent years the implementation of the electric engine in the automotive industries has been increasingly marked. The speed of the electric motors is much higher than the combustion engine ones, bringing transmission gears to be subjected to high dynamic loads. For this reason the dynamic effects on fatigue life of these components have be taken into account in a more careful way respect to what is done with the usual gears. In the present work the overload effects due to both speed and meshing in a gear couple of an electric vehicle transmission have been analyzed. The electric vehicle is designed for urban people mobility and presents all the requirements to be certified as M1 vehicle (a weight less than 600 kg and a maximum speed more than 90 Km/h). To investigate the overload effects of teeth in contact, the reference gear design Standards (ISO 6336) introduce a specific multiplicative factor to the applied load called Internal Dynamic Factor (K v ). Aim of this work is to evaluate how dynamic overloads may influence the fatigue life of the above quoted gears in term of durability. To this goal, Kv values have been calculated by means of the analytical equations (ISO 6336 Methods B and C) and then they have been compared with the results coming from multibody simulations, involving full rigid and rigid-flexible models

Upwelling of melt-depleted mantle under Iceland

Seafloor anomalies along mid-ocean ridges with exceptionally thick and compositionally distinct basaltic crust, for example, at Iceland, suggest that the underlying mantle is hotter and chemically different from the adjacent subridge mantle. Here we present hafnium and neodymium isotope ratios of peridotites from the Charlie Gibbs Transform Zone, which is located at the southern end of the Reykjanes Ridge south-west of Iceland. These peridotites are strongly depleted in incompatible elements with extremely high hafnium isotope ratios, suggesting that they had already melted to a large extent before being incorporated into the plume, at least 1 billion years ago, and thereby also became less dense. We argue that seismic velocity anomalies, geodynamic models and geochemical affinities of ridge basalts connect the peridotites from the Charlie Gibbs Transform Zone to the 'Iceland plume'. The thermochemical buoyancy of the moderately hot Iceland plume, but also that of other plumes worldwide, may therefore be strongly influenced by composition. Variable peridotite depletion along the rising Iceland plume could also cause the transient, density-driven pulses in plume flux, which have formed the V-shaped Reykjanes Ridge south of Iceland. Overall, expansion of a ridge-centred plume along adjacent ridges and melting of heterogeneous plume material explains the topographic swell, the seismic anomaly and the formation of V-shaped ridges, as well as the regional distribution of basalts with Icelandic affinity. The upwelling mantle beneath Iceland underwent melt depletion at least 1 billion years ago and is therefore compositionally buoyant, according to a study of neodymium and hafnium isotope ratios in peridotites from the Charlie Gibbs Transform Zone

Identification of a Novel p53 Modulator Endowed with Antitumoural and Antibacterial Activity through a Scaffold Repurposing Approach

Intracellular pathogens, such as Chlamydia trachomatis, have been recently shown to induce degradation of p53 during infection, thus impairing the protective response of the host cells. Therefore, p53 reactivation by disruption of the p53-MDM2 complex could reduce infection and restore pro-apoptotic effect of p53. Here, we report the identification of a novel MDM2 inhibitor with potential antitumoural and antibacterial activity able to reactivate p53. A virtual screening was performed on an in-house chemical library, previously synthesised for other targets, and led to the identification of a hit compound with a benzo[a]dihydrocarbazole structure, RM37. This compound induced p53 up-regulation in U343MG glioblastoma cells by blocking MDM2-p53 interaction and reduced tumour cell growth. NMR studies confirmed its ability to dissociate the MDM2-p53 complex. Notably, RM37 reduced Chlamydia infection in HeLa cells in a concentration-dependent manner and ameliorated the inflammatory status associated with infection

The tectonic puzzle of the Messina area (Southern Italy): Insights from new seismic reflection data

Get PDF
The Messina Strait, that separates peninsular Italy from Sicily, is one of the most seismically active areas of the Mediterranean. The structure and seismotectonic setting of the region are poorly understood, although the area is highly populated and important infrastructures are planned there. New seismic reflection data have identified a number of faults, as well as a crustal scale NE-trending anticline few km north of the strait. These features are interpreted as due to active right-lateral transpression along the north-eastern Sicilian offshore, coexisting with extensional and right-lateral transtensional tectonics in the southern Messina Strait. This complex tectonic network appears to be controlled by independent and overlapping tectonic settings, due to the presence of a diffuse transfer zone between the SE-ward retreating Calabria subduction zone relative to slab advance in the western Sicilian side

Background: In March 2020, the World Health Organization declared a global pandemic due to the novel coronavirus SARS-CoV-2 and several governments planned a national quarantine in order to control the virus spread. Acute psychological effects of quarantine in frail elderly subjects with special needs, such as patients with dementia, have been poorly investigated. The aim of this study was to assess modifications of neuropsychiatric symptoms during quarantine in patients with dementia and their caregivers. Methods: This is a sub-study of a multicenter nation-wide survey. A structured telephone interview was delivered to family caregivers of patients with diagnosis of Alzheimer disease (AD), dementia with Lewy bodies (DLB), frontotemporal dementia (FTD), and vascular dementia (VD), followed regularly at 87 Italian memory clinics. Variations in behavioral and psychological symptoms (BPSD) were collected after 1 month since quarantine declaration and associations with disease type, severity, gender, and caregiver’s stress burden were analyzed. Results: A total of 4,913 caregivers participated in the survey. Increased BPSD was reported in 59.6% of patients as worsening of preexisting symptoms (51.9%) or as new onset (26%), and requested drug modifications in 27.6% of these cases. Irritability, apathy, agitation, and anxiety were the most frequently reported worsening symptoms and sleep disorder and irritability the most frequent new symptoms. Profile of BPSD varied according to dementia type, disease severity, and patients’ gender. Anxiety and depression were associated with a diagnosis of AD (OR 1.35, CI: 1.12–1.62), mild to moderate disease severity and female gender. DLB was significantly associated with a higher risk of worsening hallucinations (OR 5.29, CI 3.66–7.64) and sleep disorder (OR 1.69, CI 1.25–2.29), FTD with wandering (OR 1.62, CI 1.12–2.35), and change of appetite (OR 1.52, CI 1.03–2.25). Stress-related symptoms were experienced by two-thirds of caregivers and were associated with increased patients’ neuropsychiatric burden (p<0.0001). Conclusion: Quarantine induces a rapid increase of BPSD in approximately 60% of patients and stress-related symptoms in two-thirds of caregivers. Health services need to plan a post-pandemic strategy in order to address these emerging needs

Continuous vs Intermittent Meropenem Administration in Critically Ill Patients With Sepsis

Importance: Meropenem is a widely prescribed β-lactam antibiotic. Meropenem exhibits maximum pharmacodynamic efficacy when given by continuous infusion to deliver constant drug levels above the minimal inhibitory concentration. Compared with intermittent administration, continuous administration of meropenem may improve clinical outcomes. Objective: To determine whether continuous administration of meropenem reduces a composite of mortality and emergence of pandrug-resistant or extensively drug-resistant bacteria compared with intermittent administration in critically ill patients with sepsis. Design, setting, and participants: A double-blind, randomized clinical trial enrolling critically ill patients with sepsis or septic shock who had been prescribed meropenem by their treating clinicians at 31 intensive care units of 26 hospitals in 4 countries (Croatia, Italy, Kazakhstan, and Russia). Patients were enrolled between June 5, 2018, and August 9, 2022, and the final 90-day follow-up was completed in November 2022. Interventions: Patients were randomized to receive an equal dose of the antibiotic meropenem by either continuous administration (n = 303) or intermittent administration (n = 304). Main outcomes and measures: The primary outcome was a composite of all-cause mortality and emergence of pandrug-resistant or extensively drug-resistant bacteria at day 28. There were 4 secondary outcomes, including days alive and free from antibiotics at day 28, days alive and free from the intensive care unit at day 28, and all-cause mortality at day 90. Seizures, allergic reactions, and mortality were recorded as adverse events. Results: All 607 patients (mean age, 64 [SD, 15] years; 203 were women [33%]) were included in the measurement of the 28-day primary outcome and completed the 90-day mortality follow-up. The majority (369 patients, 61%) had septic shock. The median time from hospital admission to randomization was 9 days (IQR, 3-17 days) and the median duration of meropenem therapy was 11 days (IQR, 6-17 days). Only 1 crossover event was recorded. The primary outcome occurred in 142 patients (47%) in the continuous administration group and in 149 patients (49%) in the intermittent administration group (relative risk, 0.96 [95% CI, 0.81-1.13], P = .60). Of the 4 secondary outcomes, none was statistically significant. No adverse events of seizures or allergic reactions related to the study drug were reported. At 90 days, mortality was 42% both in the continuous administration group (127 of 303 patients) and in the intermittent administration group (127 of 304 patients). Conclusions and relevance: In critically ill patients with sepsis, compared with intermittent administration, the continuous administration of meropenem did not improve the composite outcome of mortality and emergence of pandrug-resistant or extensively drug-resistant bacteria at day 28.
