    Fall prevention in the community: what older people say they need

This study aimed to explore older people’s perceptions of the facilitators and barriers to participation in fall prevention interventions. We undertook a qualitative study with older people who had taken part in, declined to participate or adhere to fall prevention interventions using semi-structured interviews (n=65), and 17 focus groups (n=122) with older people (including 32 South Asian and 30 Chinese older people) in primary and community care settings in the South of England. A number of factors acted as either barriers or facilitators to uptake of interventions. Older people also made recommendations for improving access to interventions. Community nurses are ideally placed to screen older people, identify those at risk of falling and refer them to appropriate interventions as well as providing health promotion and education.Peer reviewe

    Parameters determining the efficiency of gene targeting in the moss Physcomitrella patens

    In the moss Physcomitrella patens, transforming DNA containing homologous sequences integrates predominantly by homologous recombination with its genomic target. A systematic investigation of the parameters that determine gene targeting efficiency shows a direct relationship between homology length and targeting frequency for replacement vectors (a selectable marker flanked by homologous DNA). Overall homology of only 1 kb is sufficient to achieve a 50% yield of targeted transformants. Targeting may occur through homologous recombination in one arm, accompanied by non-homologous end-joining by the other arm of the vector, or by allele replacement following two homologous recombination events. Allele replacement frequency depends on the symmetry of the targeting vector, being proportional to the length of the shorter arm. Allele replacement may involve insertion of multiple copies of the transforming DNA, accompanied by ectopic insertions at non-homologous sites. Single-copy and single insertions at targeted loci (targeted gene replacements, ‘TGR’) occur with a frequency of 7–20% of all transformants when the minimum requirements for allele replacement are met. Homologous recombination in Physcomitrella is substantially more efficient than in any multicellular eukaryote, recommending it as the outstanding model for the study of homologous recombination in plants

    Tert-butyl benzoquinone: mechanism of biofilm eradication and potential for use as a topical antibiofilm agent

    Objectives: Tert-butyl benzoquinone (TBBQ) is the oxidation product of tert-butyl hydroquinone (TBHQ), an antimicrobial food additive with >40 years of safe use. TBBQ displays potent activity against Staphylococcus aureus biofilms in vitro. Here, we report on studies to further explore the action of TBBQ on staphylococcal biofilms, and provide a preliminary preclinical assessment of its potential for use as a topical treatment for staphylococcal infections involving a biofilm component. Methods: The antibacterial properties of TBBQ were assessed against staphylococci growing in planktonic culture and as biofilms in the Calgary Biofilm Device. Established assays were employed to measure the effects of TBBQ on biofilm structure and bacterial membranes, and to assess resistance potential. A living skin equivalent was used to evaluate the effects of TBBQ on human skin. Results: TBBQ eradicated biofilms of S. aureus and other staphylococcal species at concentrations ≤64 mg/L. In contrast to other redox-active agents exhibiting activity against biofilms, TBBQ did not cause substantial destructuring of the biofilm matrix; instead, the antibiofilm activity of the compound was attributed to its ability to kill slow- and non-growing cells via membrane perturbation. TBBQ acted synergistically with gentamicin, did not damage a living skin equivalent following topical application, and exhibited low resistance potential. Conclusions: The ability of TBBQ to eradicate biofilms appears to result from its ability to kill bacteria regardless of growth state. Preliminary evaluation suggests that TBBQ represents a promising candidate for development as a topical antistaphylococcal biofilm agent

    DLCQ Strings, Twist Fields and One-Loop Correlators on a Permutation Orbifold

    We investigate some aspects of the relationship between matrix string theory and light-cone string field theory by analysing the correspondence between the two-loop thermal partition function of DLCQ strings in flat space and the integrated two-point correlator of twist fields in a symmetric product orbifold conformal field theory at one-loop order. This is carried out by deriving combinatorial expressions for generic twist field correlation functions in permutation orbifolds using the covering surface method, by deriving the one-loop modification of the twist field interaction vertex, and by relating the two-loop finite temperature DLCQ string theory to the theory of Prym varieties for genus two covers of an elliptic curve. The case of bosonic Z(2) orbifolds is worked out explicitly and precise agreement between both amplitudes is found. We use these techniques to derive explicit expressions for Z(2) orbifold spin twist field correlation functions in the Type II and heterotic string theories.Comment: 48 pages, 1 figure; v2: typos correcte

    Tumour markers in breast cancer.

    The clinical usefulness of 8 potential tumour markers has been evaluated in 69 patients with Stage I and II breast cancer and 57 patients with Stage III and IV. Serum CEA concentrations were raised in 13% of patients with local and 65% of those with advanced breast cancer. In patients with clinical evidence of progression or regression of tumour, serum CEA levels changed appropriately in 83% of cases. Taking 4 of the markers (carcinoembryonic antigen (CEA), lactalbumin, alpha subunit and haptoglobin) serum concentrations of one or more were raised in 33% of patients with local disease and 81% of those with advanced breast cancer. However, marker concentrations were often only marginally raised, and are unlikely to provide sensitive guide to tumour burden. CEA, lactalbumin and alpha subunit were detectable in 68%, 43% and 40% respectively of extracts of primary breast cancers

    Land management strategies can increase oil palm plantation use by some terrestrial mammals in Colombia

    While the conservation role of remaining natural habitats in anthropogenic landscapes is clear, the degree to which agricultural matrices impose limitations to animal use is not well understood, but vital to assess species’ resilience to land use change. Using an occupancy framework, we evaluated how oil palm plantations affect the occurrence and habitat use of terrestrial mammals in the Colombian Llanos. Further, we evaluated the effect of undergrowth vegetation and proximity to forest on habitat use within plantations. Most species exhibited restricted distributions across the study area, especially in oil palm plantations. Habitat type strongly influenced habitat use of four of the 12 more widely distributed species with oil palm negatively affecting species such as capybara and naked-tailed armadillo. The remaining species showed no apparent effect of habitat type, but oil palm and forest use probabilities varied among species. Overall, generalist mesocarnivores, white-tailed deer, and giant anteater were more likely to use oil palm while the remaining species, including ocelot and lesser anteater, showed preferences for forest. Distance to nearest forest had mixed effects on species habitat use, while understory vegetation facilitated the presence of species using oil palm. Our findings suggest that allowing undergrowth vegetation inside plantations and maintaining nearby riparian corridors would increase the likelihood of terrestrial mammals’ occurrence within oil palm landscapes
