    Culturing Keratinocytes and Fibroblasts in a Three-Dimensional Mesh Results in Epidermal Differentiation and Formation of a Basal Lamina-Anchoring Zone

    The purpose of this study was to characterize an in vitro co-culture model in which fibroblasts grown in a three-dimensional nylon mesh were recombined with human keratinocytes. The cultures were kept for 3 and 5 weeks and then processed for electron microscopy. Keratinocytes showed reconstruction of an epidermis consisting of a basal layer with hemidesmosome's, a stratified epithelium with tonofilament's and desmosomes, a granular layer with keratinosomes and keratohyaline granules, and a transitional stratum corneum. Anchoring filaments, lamina densa, anchoring fibrils, bundles of elastin-associated microfibrils (diameters 10 nm) and fine collagen fibrils were formed. Collagen fibrils near the epidermis were much thinner than those in the lower levels.The present study shows that the dermal model containing metabolically active fibroblasts in their natural environment will support epidermal morphogenesis and differentiation including the formation of a basal lamina and anchoring zone

    The Circadian Rhythm of Blood Pressure During Pregnancy

    To review the literature on the circadian rhythm of blood pressure during pregnancy. Data Sources : Computerized searches on MEDLINE, CINAHL, and MIRLYN. Study Selection : Selected studies from 1969 to 1997 were evaluated. Data Extraction : Data were extracted and information was organized under the following areas: definition of and the interconnection between circadian rhythm and blood pressure; the circadian variability of blood pressure throughout the trimesters; the patterns of the circadian rhythm of blood pressure in pregnancies defined as normal and those complicated by chronic hypertension and preeclampsia; and clinical implications. Data Synthesis : The circadian rhythm of blood pressure in pregnancy is the same as in the nonpregnant state, with a nocturnal decrease, especially during sleep. In patients with chronic hypertension, the nocturnal fall in blood pressure may be steeper. Patients with mild preeclampsia may experience a less pronounced nocturnal decrease in blood pressure. Patients with severe preeclampsia may display a reversed circadian rhythm, with no decrease and/or an increase in nocturnal blood pressure. Conclusions : The patterns of the circadian rhythm of blood pressure during normal pregnancy and pregnancies complicated by chronic hypertension and preeclampsia warrant consideration when monitoring patients and implementing management plans.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/71412/1/j.1552-6909.2000.tb02771.x.pd

    Les consultations pédopsychiatriques aux urgences pédiatriques du CHU de Montpellier

    En France et dans le monde, les urgences pédiatriques sont confrontées à une augmentation des consultations d'enfants et adolescents se présentant pour des motifs pédopsychiatriques. Nous avons voulu quantifier ces consultations au CHU de Montpellier et connaître les caractéristiques socio démographiques et médicales de ces patients afin de mieux comprendre leurs difficultés et leurs besoins. Pour cela, nous avons inclus tous les patients âgés de moins de 15 ans et 3 mois se présentant aux urgences pédiatriques pour des motifs pédopsychiatriques entre 2008 et 2010. Nous avons focalisé une partie de notre étude sur les adolescents suicidants et sur leur devenir à 6 mois et à 1 an. Notre cohorte réunit 549 patients reçus sur 3 ans, avec une augmentation de 34%. Elle contient 65% de filles, âgés de 13 ans en moyenne. 75% des consultations ont lieu durant la période scolaire, dans 70% des cas en journée. Les patients consultent pour 4 motifs principaux: 39,3% tentative de suicide, 23,1% troubles du comportement, 13,1% intoxication alcoolique/toxiques, 10,4% angoisse. 56,3% présentent des antécédents psychiatriques personnels, 44% ont des antécédents psychiatriques familiaux, et 60% bénéficient déjà d'un suivi psychiatrique. 75% des patients sont orientés en consultation, 22% en hospitalisation. 40% des adolescents suicidants se représenteront aux urgences dans l'année suivant leur geste, malgré lm taux de suivi ambulatoire de 95%. Les consultations pour motifs pédopsychiatriques, bien que ne représentant qu'une faible partie de l'activité des urgences, mobilise des ressources importantes.     High-resolution magnetic resonance imaging at 2 Tesla: potential for atherosclerotic lesions exploration in the apolipoprotein E knockout mouse

    International audienceINTRODUCTION: The aim of the present study was to evaluate the potential of high-resolution MRI at 2 Tesla (T) for direct noninvasive imaging of the aortic wall in a mouse model of atherosclerosis.MATERIAL AND METHODS: A specific mouse antenna was developed and sequence parameters were adjusted. T(1)- and T2-weighted images of abdominal aorta were obtained at 2 T with a spatial resolution of 86 x 86 x 800 microm3 in vivo. With a dedicated small coil, ex vivo MRI of the aorta was performed with a spatial resolution of 54 x 54 x 520 microm3.RESULTS: In vivo, the aortic wall was clearly defined on T(2)-weighted images in 15 of 16 mice: along the aorta the lumen circumference ranged from 1.07 to 3.61 mm and mean wall thickness from 0.11 to 0.67 mm. In vivo measurements of plaque distribution were confirmed by ex vivo MR imaging and by histology, with a good correlation with histology regarding lumen circumference (r = 0.94) and wall thickness (r = 0.97).CONCLUSION: Magnetic resonance imaging at 2 T to analyze in vivo atherosclerotic lesions in mice is possible with a spatial resolution of 86 x 86 x 800 microm3 and thus can be used for noninvasive follow-up in evaluation of new drugs
