Intralysosomal accumulation of polyanions. I. Fusion of pinocytic and phagocytic vacuoles with secondary lysosomes
The long-term exposure of macrophages to low concentrations of a number of polyanions leads to their accumulation in high concentration within secondary lysosomes. This was associated with enlargement of the lysosomes, the presence of membranous whorls, and intense toluidine blue staining of the organelles at pH 1.0. After the ingestion of a particulate load by these cells, newly formed phagocytic vacuoles failed to fuse with polyanion-laden lysosomes. The lack of fusion was evident in both fluorescence and electron micrographic studies which followed the transfer of acridine orange or Thorotrast from 2° lysosomes to phagosomes. Agents that inhibited phagosome-lysosome (P-L) fusion included molecules containing high densities of sulfate, sulfonate, or carboxylate residues. Dextran sulfate (DS) in ”g/ml quantities was an excellent inhibitor, whereas nonsulfated dextran (D) was without effect at 1,000-fold higher concentrations. In contrast to their effects on P-L fusion, polyanions failed to influence the fusion of pinocytic vesicles with 2° lysosomes. The uptake, intravacuolar distribution, and intralysosomal digestion of fluid-phase pinocytic markers were unaltered in lysosomes containing either D or DS. Furthermore, subcellular fractionation studies showed that the fluid-phase pinocytic marker HRP was efficiently transferred from pinosomes to large, dense 2° lysosomes containing DS
Rebuilding Bridges:Toward a Feminist Research Agenda for Postwar Reconstruction
As feminists who think about war and peacebuilding, we cannot help but encounter the complex, entwined political economic processes that underlie warsâ causes, their courses, and the challenges of postwar reconstruction. For us, then, the increasing academic division between feminist security studies (FSS) and feminist (international) political economy (FPE/FIPE) has been a cause for concern, and we welcomed Politics &amp; Genderâs earlier Critical Perspectives section on efforts to bridge the two (June 2015). We noticed, however, that although violence was addressed in several of the special section's articles, war made only brief and somewhat peripheral appearances, and peacebuilding was all but absent. While three contributions (Hudson 2015; Sjoberg 2015; True 2015) mentioned the importance of political economy in the analysis of armed conflict, the aspects of war on which the articles focused were militarized sexualities (Sjoberg 2015) or conflict-related and postwar sexual and gender-based violence (Hudson 2015; True 2015).</jats:p
Creative group performances to assess core competencies in a first-year patient-centered medicine course
Introduction: A novel assessment of systems-based practice and practice-based learning and improvement learning objectives, implemented in a first-year patient-centered medicine course, is qualitatively described. Methods: Student learning communities were asked to creatively demonstrate a problem and solution for health care delivery. Skits, filmed performances, plays, and documentaries were chosen by the students. Video recordings were reviewed for themes and the presence of course competencies. Results: All performances demonstrated not only the index competencies of team work and facilitation of the learning of others, but many other core objectives of the course. The assignment was rated positively both by the faculty and the students, and has been added to the assessment modalities of the course
Lenvatinib Plus Pembrolizumab in Patients With Advanced Endometrial Cancer.
PURPOSE: Patients with advanced endometrial carcinoma have limited treatment options. We report final primary efficacy analysis results for a patient cohort with advanced endometrial carcinoma receiving lenvatinib plus pembrolizumab in an ongoing phase Ib/II study of selected solid tumors.
METHODS: Patients took lenvatinib 20 mg once daily orally plus pembrolizumab 200 mg intravenously once every 3 weeks, in 3-week cycles. The primary end point was objective response rate (ORR) at 24 weeks (ORR
RESULTS: At data cutoff, 108 patients with previously treated endometrial carcinoma were enrolled, with a median follow-up of 18.7 months. The ORR
CONCLUSION: Lenvatinib plus pembrolizumab showed promising antitumor activity in patients with advanced endometrial carcinoma who have experienced disease progression after prior systemic therapy, regardless of tumor MSI status. The combination therapy had a manageable toxicity profile
Rassenschande, genocide and the reproductive Jewish body: examining the use of rape and sexualized violence against Jewish women during the Holocaust
Rape and sexual violence against Jewish women is a relatively unexplored area of investigation. This article adds to the scant literature on this topic. It asks: how and why did women's reproductive bodies (gender), combined with their status as Jews (race), make them particularly vulnerable during the Holocaust? The law against Rassenschande (racial defilement) prohibited sexual relations between Aryans and non-Aryans. Yet, Jewish women were raped by German men. Providing a more nuanced account than is provided by the dehumanization thesis, this article argues that women were targeted precisely because of their Jewishness and their reproductive capabilities. In addition, this piece proposes that the genocidal attack on women's bodies in the form of rape (subsequently leading to the murder of impregnated women) and sexualized violence (forced abortions and forced sterilizations) must be interpreted as an attack on an essentialized group: woman-as-Jew
Interplay Between Gravity and Quintessence: A Set of New GR Solutions
A set of new exact analytical General Relativity (GR) solutions with
time-dependent and spatially inhomogeneous quintessence demonstrate 1) a static
non-empty space-time with a horizon-type singular surface; 2) time-dependent
spatially homogeneous `spheres' which are completely different in geometry from
the Friedmann isotropic models; 3) infinitely strong anti-gravity at a `true'
singularity where the density is infinitely large. It is also found that 4) the
GR solutions allow for an extreme `density-free' form of energy that can
generate regular space-time geometries
