Characterization and Biological Activities of In Vitro Digested Olive Pomace Polyphenols Evaluated on Ex Vivo Human Immune Blood Cells

Olive pomace (OP) represents one of the main by-products of olive oil production, which still contains high quantities of health-promoting bioactive compounds. In the present study, three batches of sun-dried OP were characterized for their profile in phenolic compounds (by HPLC-DAD) and in vitro antioxidant properties (ABTS, FRAP and DPPH assays) before (methanolic extracts) and after (aqueous extracts) their simulated in vitro digestion and dialysis. Phenolic profiles, and, accordingly, the antioxidant activities, showed significant differences among the three OP batches, and most compounds showed good bioaccessibility after simulated digestion. Based on these preliminary screenings, the best OP aqueous extract (OP-W) was further characterized for its peptide composition and subdivided into seven fractions (OP-F). The most promising OP-F (characterized for its metabolome) and OP-W samples were then assessed for their potential anti-inflammatory properties in ex vivo human peripheral mononuclear cells (PBMCs) triggered or not with lipopolysaccharide (LPS). The levels of 16 pro-and anti-inflammatory cytokines were measured in PBMC culture media by multiplex ELISA assay, whereas the gene expressions of interleukin-6 (IL-6), IL-10 and TNF-α were measured by real time RT-qPCR. Interestingly, OP-W and PO-F samples had a similar effect in reducing the expressions of IL-6 and TNF-α, but only OP-W was able to reduce the release of these inflammatory mediators, suggesting that the anti-inflammatory activity of OP-W is different from that of OP-F

We present extensive spectroscopic and photometric monitoring of two famous and currently highly active luminous blue variables (LBVs) in the Large Magellanic Cloud (LMC), together with more limited coverage of three further, lesser known members of the class. R127 was discovered as an Ofpe/WN9 star in the 1970s but entered a classical LBV outburst in or about 1980 that is still in progress, thus enlightening us about the minimum state of such objects. R71 is currently the most luminous star in the LMC and continues to provide surprises, such as the appearance of [Ca ii] emission lines, as its spectral type becomes unprecedentedly late. Most recently, R71 has developed inverse P Cyg profiles in many metal lines. The other objects are as follows: HDE 269582, now a "second R127" that has been followed from Ofpe/WN9 to A type in its current outburst; HDE 269216, which changed from late B in 2014 to AF in 2016, its first observed outburst; and R143 in the 30 Doradus outskirts. The light curves and spectroscopic transformations are correlated in remarkable detail and their extreme reproducibility is emphasized, both for a given object and among all of them. It is now believed that some LBVs proceed directly to core collapse. One of these unstable LMC objects may thus oblige in the near future, teaching us even more about the final stages of massive stellar evolution.Facultad de Ciencias Astronómicas y GeofísicasInstituto de Astrofísica de La Plat

Abstract

Background

HPV type distribution by cytological status represents useful information to predict the impact of mass vaccination on screening programs.

Methods

women aged from 25 to 64 who attended cervical cancer screening in five different Italian regions were tested for HPV infection with Hybrid Capture II (HCII) low and high risk probes. Women repeating Pap-test upon unsatisfactory or positive results, or as a post-treatment and post-colposcopy follow-up analysis, were excluded from our study. High risk (HR) HPV positive samples were typed using GP5+/GP6+ primed PCR, followed by Reverse Line Blot for 18 high/intermediate risk HPV types, while low risk (LR) HPV positive samples were tested with type specific primers for HPV6 and HPV11.

Results

3410 women had a valid HCII and Pap-test. The prevalence of HR and LR infections was 7.0% and 3.6%, 29.1% and 13.7%, 68.1% and 31.9%, 60.0% and 0.0%, 65.0% and 12.0%, for negative, ASC-US, L-SIL, ASC-H and H-SIL cytology, respectively. The fraction of ASC-US+ cytology due to HPV 16 and 18 ranged from 11.2 (HPV 16/18 alone) to 15.4% (including HPV 16/18 in co-infection with other virus strains), and that due to HPV 6 and 11 ranged from 0.2% (HPV 6/11 alone) to 0.7% (including HPV 6/11 in co-infection with other LR virus strains).

Conclusions

mass vaccination with bivalent or quadrivalent HPV vaccine would modestly impact on prevalence of abnormal Pap-test in screening.</p

VISTA Variables in the Vía Láctea (VVV): Halfway Status and Results

    Get PDF
The VISTA Variables in the Vía Láctea (VVV) survey is one of six near-infrared ESO public surveys, and is now in its fourth year of observing. Although far from being complete, the VVV survey has already delivered many results, some directly connected to the intended science goals (detection of variable stars, microlensing events, new star clusters), others concerning more exotic objects, e.g., novae. Now, at the end of the fourth observing period, and comprising roughly 50% of the proposed observations, the status of the survey, as well some of results based on the VVV data, are presented.Facultad de Ciencias Astronómicas y Geofísica

Design of a Hydraulic System for Liquid Packaging

    No full text
The paper analyzes the dynamic behaviour of an industrial system devoted to the automated hydraulic packaging of beverages. More in detail, a lumped and distributed numerical approach is used to model both the filling system and the multi-actuators hydraulic circuit needed to shape and separate the packages. The model reliability and accuracy are addressed by means of a numerical vs. experimental comparison of the main hydraulic and mechanical quantities for an actual production rate. Afterwards, the system architecture is redesigned in order to obtain higher production rates, and the effects of the hydraulic behaviour variation on the hydraulic efficiency are highlighted. Finally, a sensitivity analysis with respect to the main design parameters is carried out, in order to determine the circuit layout that maximizes the system efficiency in the whole production-rate range
