    Environmental Fate and Effects of Bacillus thuringiensis (Bt) Proteins from Transgenic Crops:  a Review

    This paper reviews the scientific literature addressing the environmental fate and nontarget effects of the Cry protein toxins from Bacillus thuringiensis (Bt), specifically resulting from their expression in transgenic crops. Published literature on analytical methodologies for the detection and quantification of the Cry proteins in environmental matrices is also reviewed, with discussion of the adequacy of the techniques for determining the persistence and mobility of the Bt proteins. In general, assessment of the nontarget effects of Bt protein toxins indicates that there is a low level of hazard to most groups of nontarget organisms, although some investigations are of limited ecological relevance. Some published reports on the persistence of the proteins in soil show short half-lives, whereas others show low-level residues lasting for many months. Improvements in analytical methods will allow a more complete understanding of the fate and significance of Bt proteins in the environment

    Pyrometamorphism and partial melting of shales during combustion metamorphism: mineralogical, textural, and chemical effects

    Eocene shales metamorphosed by a naturally ignited coal seam in the Powder River Basin, Wyoming record a continuum of mineralogic and textural changes from relatively unaltered shale to melt developed during pyrometamorphism. Samples collected along a section 2 m in length, corresponding to a temperature range of approximately 1300°C, were examined optically and by XRD, SEM, and STEM. The low temperature samples are comprised primarily of silt-sized quartz, K-feldspar, and minor amounts of other detrital minerals in a continuous matrix of illite/smectite (I/S). Delamination of phyllosilicates due to dehydroxylation occurs early in the sequence with curling of individual layers from rim to core. Within one-half meter of melted areas, phyllosilicates have undergone an essentially isochemical reconstitution with nucleation and growth of mullite crystals with maximum diameters of 50 nm, randomly distributed within a non-crystalline phase that replaces I/S. Large detrital grains remain for the most part unaffected except for the inversion of quartz to tridymite/cristobalite. Within 1 mm of the solid/melt interface, the mullitebearing clay mineral matrix is essentially homogeneous in composition with obscure grain boundaries, caused by apparent homogenization of poorly crystalline material. This material is similar in composition to parent clays and acts as a matrix to angular, remnant tridymite/cristobalite grains. Rounded, smaller silica grains have reaction rims with the non-crystalline matrix; K-feldspar is no longer present (apparently reacted with the matrix) and the matrix contains abundant pore space due to shrinkage upon dehydroxylation. As isolated pods of paralava (glass) or fractures are approached, Fe−Ti−Al oxides become abundant. Vesicular glass is separated from clinker by a well-defined interface and contains numerous phenocrysts. XRF analyses and reduced area rastering using EDS imply enrichment of the melt phase in Fe, Ca, Mg and Mn, apparently due to vapor transport from other layers lower in the sedimentary sequence.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/47303/1/410_2004_Article_BF00310784.pd

    Development of the morpholino gene knockdown technique in Fundulus heteroclitus : a tool for studying molecular mechanisms in an established environmental model

The Atlantic killifish (Fundulus heteroclitus) is one of the most studied teleost environmental models, yet few genetic or genomic tools have been developed for use in this species. The advancement of genetic and evolutionary toxicology will require that many of the tools developed in laboratory models be transferred into species more applicable to environmental toxicology. Antisense morpholino oligonucleotide (MO) gene knockdown technology has been widely utilized to study development in zebrafish and has been proven to be a powerful tool in toxicological investigations through direct manipulation of molecular pathways. To expand the utility of killifish as an environmental model, MO gene knockdown technology was adapted for use in Fundulus. Morpholino microinjection methods were altered to overcome the significant differences between these two species. Morpholino efficacy and functional duration were evaluated with molecular and phenotypic methods. A cytochrome P450-1A (CYP1A) MO was used to confirm effectiveness of the methodology. For CYP1A MO-injected embryos, a 70% reduction in CYP1A activity, a 86% reduction in total CYP1A protein, a significant increase in β-naphthoflavone-induced teratogenicity, and estimates of functional duration (50% reduction in activity 10 dpf, and 86% reduction in total protein 12 dpf) conclusively demonstrated that MO technologies can be used effectively in killifish and will likely be just as informative as they have been in zebrafish.This work was funded in part by the National Institute of Environmental Health Sciences through the Duke Superfund Basic Research Center (P42ES010356), the Boston University Superfund Basic Research Program (P42ES007381), and the Duke Integrated Toxicology and Environmental Health Program (ES-T32-0007031). Here, we examined effects of PCB153, dioxin-like (DL) PCB126, or a mixture of both, on Cyp3a56 mRNA in killifish generations removed from the wild, without environmental PCB exposures. PCB126 effects in liver and gills differed between populations, as expected. Gill Cyp3a56 was not affected by either congener in NBH F2 generation fish, but was induced by PCB153 in SC F1 fish, with females showing a greater response. PCB153 did not affect Cyp3a56 in liver of either population. Results suggest a heritable resistance to NDL-PCBs in killifish from NBH, in addition to that reported for DL PCBs.     Optimal Image Reconstruction in Radio Interferometry

    We introduce a method for analyzing radio interferometry data which produces maps which are optimal in the Bayesian sense of maximum posterior probability density, given certain prior assumptions. It is similar to maximum entropy techniques, but with an exact accounting of the multiplicity instead of the usual approximation involving Stirling's formula. It also incorporates an Occam factor, automatically limiting the effective amount of detail in the map to that justified by the data. We use Gibbs sampling to determine, to any desired degree of accuracy, the multi-dimensional posterior density distribution. From this we can construct a mean posterior map and other measures of the posterior density, including confidence limits on any well-defined function of the posterior map.Comment: 41 pages, 11 figures.     Incidence of Malaria and Efficacy of Combination Antimalarial Therapies over 4 Years in an Urban Cohort of Ugandan Children

    Combination therapies are now recommended to treat uncomplicated malaria. We used a longitudinal design to assess the incidence of malaria and compare the efficacies of 3 combination regimens in Kampala, Uganda.Children aged 1-10 years were enrolled from randomly selected households in 2004-05 and 2007, and were followed at least monthly through 2008. Insecticide-treated bednets (ITNs) were provided in 2006. Children were randomized upon their first episode, and then treated for all episodes of uncomplicated malaria with amodiaquine/sulfadoxine-pyrimethamine (AQ/SP), artesunate/amodiaquine (AS/AQ), or artemether/lumefantrine (AL). Risks of parasitological failure were determined for each episode of uncomplicated malaria and clinical parameters were followed. A total of 690 children experienced 1464 episodes of malaria. 96% of these episodes were uncomplicated malaria and treated with study drugs; 94% were due to Plasmodium falciparum. The rank order of treatment efficacy was AL > AS/AQ > AQ/SP. Failure rates increased over time for AQ/SP, but not the artemisinin-based regimens. Over the 4-year course of the study the prevalence of asymptomatic parasitemia decreased from 11.8% to 1.4%, the incidence of malaria decreased from 1.55 to 0.32 per person year, and the prevalence of anemia (hemoglobin <10 gm/dL) decreased from 5.9% to 1.0%. No episodes of severe malaria (based on WHO criteria) and no deaths were seen.With ready access to combination therapies and distribution of ITNs, responses were excellent for artemisinin-containing regimens, severe malaria was not seen, and the incidence of malaria and prevalence of parasitemia and anemia decreased steadily over time.isrctn.org ISRCTN37517549

Accurately measuring shoulder rhythm is difficult. To classify shoulder rhythm and identify pathological movement, static measures have been the preferred method. However, dynamic measures are also used and can be less burdensome to obtain. The purpose of this paper was to determine how closely dynamic measures represent static measures using the same acromion marker cluster scapular tracking technique. Five shoulder angles were assessed for 24 participants using dynamic and static tracking techniques during humeral elevation in three planes (frontal, scapular, sagittal). ANOVAs were used to identify where significant differences existed for the factors of plane, elevation angle, and tracking technique (static, dynamic raising, dynamic lowering). All factors were significantly different for all shoulder angles (p<0.001), except for elevation plane in scapulothoracic protraction/retraction (p=0.955). Tracking techniques were influential (p<0.001), but the grouped mean differences fell below a clinically relevant 5° benchmark. There was large variation in mean differences of the techniques across individuals. While population averages are similar, individual static and dynamic shoulder assessments may be different. Caution should be taken when dynamic shoulder assessments are performed on individuals, as they may not reflect those obtained in static scapular motion tracking.Natural Sciences and Engineering Research Council || Canada Foundation for Innovatio
