    Impact-Oriented Contextual Scholar Profiling using Self-Citation Graphs

    Quantitatively profiling a scholar's scientific impact is important to modern research society. Current practices with bibliometric indicators (e.g., h-index), lists, and networks perform well at scholar ranking, but do not provide structured context for scholar-centric, analytical tasks such as profile reasoning and understanding. This work presents GeneticFlow (GF), a suite of novel graph-based scholar profiles that fulfill three essential requirements: structured-context, scholar-centric, and evolution-rich. We propose a framework to compute GF over large-scale academic data sources with millions of scholars. The framework encompasses a new unsupervised advisor-advisee detection algorithm, a well-engineered citation type classifier using interpretable features, and a fine-tuned graph neural network (GNN) model. Evaluations are conducted on the real-world task of scientific award inference. Experiment outcomes show that the F1 score of best GF profile significantly outperforms alternative methods of impact indicators and bibliometric networks in all the 6 computer science fields considered. Moreover, the core GF profiles, with 63.6%-66.5% nodes and 12.5%-29.9% edges of the full profile, still significantly outrun existing methods in 5 out of 6 fields studied. Visualization of GF profiling result also reveals human explainable patterns for high-impact scholars

    Bacterial Microbiota Profiling in Gastritis without Helicobacter pylori Infection or Non-Steroidal Anti-Inflammatory Drug Use

    Recent 16S ribosomal RNA gene (rRNA) molecular profiling of the stomach mucosa revealed a surprising complexity of microbiota. Helicobacter pylori infection and non-steroidal anti-inflammatory drug (NSAID) use are two main contributors to gastritis and peptic ulcer. However, little is known about the association between other members of the stomach microbiota and gastric diseases. In this study, cloning and sequencing of the 16S rRNA was used to profile the stomach microbiota from normal and gastritis patients. One hundred and thirty three phylotypes from eight bacterial phyla were identified. The stomach microbiota was found to be closely adhered to the mucosa. Eleven Streptococcus phylotypes were successfully cultivated from the biopsies. One to two genera represented a majority of clones within any of the identified phyla. We further developed two real-time quantitative PCR assays to quantify the relative abundance of the Firmicutes phylum and the Streptococcus genus. Significantly higher abundance of the Firmicutes phylum and the Streptococcus genus within the Firmicutes phylum was observed in patients with antral gastritis, compared with normal controls. This study suggests that the genus taxon level can largely represent much higher taxa such as the phylum. The clinical relevance and the mechanism underlying the altered microbiota composition in gastritis require further functional studies

    Screening Acute HIV Infections among Chinese Men Who Have Sex with Men from Voluntary Counseling & Testing Centers

    Recent studies have shown the public health importance of identifying acute HIV infection (AHI) in the men who have sex with men (MSM) of China, which has a much higher risk of HIV transmission. However, cost-utility analyses to guide policy around AHI screening are lacking.An open prospective cohort was recruited among MSM living in Liaoning Province, Northeast China. Blood samples and epidemiological information were collected every 10 weeks. Third-generation ELISA and rapid test were used for HIV antibody screening, western blot assay (WB) served for assay validation. Antibody negative specimens were tested with 24 mini-pool nucleic acid amplification testing (NAAT). Specimens with positive ELISA but negative or indeterminate WB results were tested with NAAT individually without mixing. A cost-utility analysis of NAAT screening was assessed. Among the 5,344 follow-up visits of 1,765 MSM in 22 months, HIV antibody tests detected 114 HIV chronic infections, 24 seroconverters and 21 antibody indeterminate cases. 29 acute HIV infections were detected with NAAT from 21 antibody indeterminate and 1,606 antibody negative cases. The HIV-1 prevalence and incidence density were 6.6% (95% CI: 5.5–7.9) and 7.1 (95% CI: 5.4–9.2)/100 person-years, respectively. With pooled NAAT and individual NAAT strategy, the cost of an HIV transmission averted was 1,480.TheadditionofNAATafterHIVantibodytestshadacostutilityratioof1,480. The addition of NAAT after HIV antibody tests had a cost-utility ratio of 3,366 per gained quality-adjusted life year (QALY). The input-output ratio of NAAT was about 1∶16.9.The HIV infections among MSM continue to rise at alarming rates. Despite the rising cost, adding pooled NAAT to the HIV antibody screening significantly increases the identification of acute HIV infections in MSM. Early treatment and target-oriented publicity and education programs can be strengthened to decrease the risk of HIV transmission and to save medical resources in the long run

    Dietary Pectin Increases Intestinal Crypt Stem Cell Survival following Radiation Injury

    This research was performed as a project of the Intestinal Stem Cell Consortium, a collaborative research project funded by the National Institute of Diabetes and Digestive and Kidney Diseases (NIH U01 DK-085508 to CWH), and a grant from Oklahoma Center for the Advancement of Science and Technology to CWH.Gastrointestinal (GI) mucosal damage is a devastating adverse effect of radiation therapy. We have recently reported that expression of Dclk1, a Tuft cell and tumor stem cell (TSC) marker, 24h after high dose total-body gamma-IR (TBI) can be used as a surrogate marker for crypt survival. Dietary pectin has been demonstrated to possess chemopreventive properties, whereas its radioprotective property has not been studied. The aim of this study was to determine the effects of dietary pectin on ionizing radiation (IR)-induced intestinal stem cell (ISC) deletion, crypt and overall survival following lethal TBI. C57BL/6 mice received a 6% pectin diet and 0.5% pectin drinking water (pre-IR mice received pectin one week before TBI until death; post-IR mice received pectin after TBI until death). Animals were exposed to TBI (14 Gy) and euthanized at 24 and 84h post-IR to assess ISC deletion and crypt survival respectively. Animals were also subjected to overall survival studies following TBI. In pre-IR treatment group, we observed a three-fold increase in ISC/crypt survival, a two-fold increase in Dclk1+ stem cells, increased overall survival (median 10d vs. 7d), and increased expression of Dclk1, Msi1, Lgr5, Bmi1, and Notch1 (in small intestine) post-TBI in pectin treated mice compared to controls. We also observed increased survival of mice treated with pectin (post-IR) compared to controls. Dietary pectin is a radioprotective agent; prevents IR-induced deletion of potential reserve ISCs; facilitates crypt regeneration; and ultimately promotes overall survival. Given the anti-cancer activity of pectin, our data support a potential role for dietary pectin as an agent that can be administered to patients receiving radiation therapy to protect against radiation-induces mucositis.Yeshttp://www.plosone.org/static/editorial#pee

    Stability Analysis of an Advanced Persistent Distributed Denial-of-Service Attack Dynamical Model

    The advanced persistent distributed denial-of-service (APDDoS) attack is a fairly significant threat to cybersecurity. Formulating a mathematical model for accurate prediction of APDDoS attack is important. However, the dynamical model of APDDoS attack has barely been reported. This paper first proposes a novel dynamical model of APDDoS attack to understand the mechanisms of APDDoS attack. Then, the attacked threshold of this model is calculated. The global stability of attack-free and attacked equilibrium are both proved. The influences of the model’s parameters on attacked equilibrium are discussed. Eventually, the main conclusions of the theoretical analysis are examined through computer simulations

    Robustness Property of Robust-BD Wald-Type Test for Varying-Dimensional General Linear Models

    An important issue for robust inference is to examine the stability of the asymptotic level and power of the test statistic in the presence of contaminated data. Most existing results are derived in finite-dimensional settings with some particular choices of loss functions. This paper re-examines this issue by allowing for a diverging number of parameters combined with a broader array of robust error measures, called “robust- BD ”, for the class of “general linear models”. Under regularity conditions, we derive the influence function of the robust- BD parameter estimator and demonstrate that the robust- BD Wald-type test enjoys the robustness of validity and efficiency asymptotically. Specifically, the asymptotic level of the test is stable under a small amount of contamination of the null hypothesis, whereas the asymptotic power is large enough under a contaminated distribution in a neighborhood of the contiguous alternatives, thus lending supports to the utility of the proposed robust- BD Wald-type test

    An Advanced Persistent Distributed Denial-of-Service Attacked Dynamical Model on Networks

    The advanced persistent distributed denial-of-service (APDDoS) attack does a serious harm to cyber security. Establishing a mathematical model to accurately predict APDDoS attack on networks is still an important problem that needs to be solved. Therefore, to help us understand the attack mechanisms of APDDoS on networks, this paper first puts forward a novel dynamical model of APDDoS attack on networks. A systematic analysis of this new model shows that the maximum eigenvalue of the networks is a vital factor that determines the success or failure of the attack. What is more, a new sufficient condition for the global stability of attack-free equilibrium is obtained. The global attractivity of attacked equilibrium has also been proved. Eventually, this paper gives some numerical simulations to show the main results
