    Protein Profiling of Human Nonpigmented Ciliary Epithelium Cell Secretome: The Differentiation Factors Characterization for Retinal Ganglion Cell line

    The purpose of this paper was to characterize proteins secreted from the human nonpigmented ciliary epithelial (HNPE) cells, which have differentiated a rat retinal ganglion cell line, RGC-5. Undifferentiated RGC-5 cells have been shown to express several marker proteins characteristic of retinal ganglion cells. However, RGC-5 cells do not respond to N-methyl-D aspartate (NMDA), or glutamate. HNPE cells have been shown to secrete numbers of neuropeptides or neuroproteins also found in the aqueous humor, many of which have the ability to influence the activity of neuronal cells. This paper details the profile of HNPE cell-secreted proteins by proteomic approaches. The experimental results revealed the identification of 132 unique proteins from the HNPE cell-conditioned SF-medium. The biological functions of a portion of these identified proteins are involved in cell differentiation. We hypothesized that a differentiation system of HNPE cell-conditioned SF-medium with RGC-5 cells can induce a differentiated phenotype in RGC-5 cells, with functional characteristics that more closely resemble primary cultures of rat retinal ganglion cells. These proteins may replace harsh chemicals, which are currently used to induce cell differentiation

    The association between subjective memory complaint and objective cognitive function in older people with previous major depression

    The goal of this study is to investigate associations between subjective memory complaint and objective cognitive performance in older people with previous major depression-a high-risk sample for cognitive impairment and later dementia. A cross-sectional study was carried out in people aged 60 or over with previous major depression but not fulfilling current major depression criteria according to DSM-IV-TR. People with dementia or Mini-Mental State Examination score less than 17 were excluded. Subjective memory complaint was defined on the basis of a score ≧4 on the subscale of Geriatric Mental State schedule, a maximum score of 8. Older people aged equal or over 60 without any psychiatric diagnosis were enrolled as healthy controls. Cognitive function was evaluated using a series of cognitive tests assessing verbal memory, attention/speed, visuospatial function, verbal fluency, and cognitive flexibility in all participants. One hundred and thirteen older people with previous major depression and forty-six healthy controls were enrolled. Subjective memory complaint was present in more than half of the participants with depression history (55.8%). Among those with major depression history, subjective memory complaint was associated with lower total immediate recall and delayed verbal recall scores after adjustment. The associations between subjective memory complaint and worse memory performance were stronger in participants with lower depressive symptoms (Hamilton Depression Rating Scale score<7). The results suggest subjective memory complaint may be a valid appraisal of memory performance in older people with previous major depression and consideration should be given to more proactive assessment and follow-up in these clinical samples

    A 3D Model of the Membrane Protein Complex Formed by the White Spot Syndrome Virus Structural Proteins

    Outbreaks of white spot disease have had a large negative economic impact on cultured shrimp worldwide. However, the pathogenesis of the causative virus, WSSV (whit spot syndrome virus), is not yet well understood. WSSV is a large enveloped virus. The WSSV virion has three structural layers surrounding its core DNA: an outer envelope, a tegument and a nucleocapsid. In this study, we investigated the protein-protein interactions of the major WSSV structural proteins, including several envelope and tegument proteins that are known to be involved in the infection process.In the present report, we used coimmunoprecipitation and yeast two-hybrid assays to elucidate and/or confirm all the interactions that occur among the WSSV structural (envelope and tegument) proteins VP51A, VP19, VP24, VP26 and VP28. We found that VP51A interacted directly not only with VP26 but also with VP19 and VP24. VP51A, VP19 and VP24 were also shown to have an affinity for self-interaction. Chemical cross-linking assays showed that these three self-interacting proteins could occur as dimers.From our present results in conjunction with other previously established interactions we construct a 3D model in which VP24 acts as a core protein that directly associates with VP26, VP28, VP38A, VP51A and WSV010 to form a membrane-associated protein complex. VP19 and VP37 are attached to this complex via association with VP51A and VP28, respectively. Through the VP26-VP51C interaction this envelope complex is anchored to the nucleocapsid, which is made of layers of rings formed by VP664.     Toxicity of Terahertz-Based Functional Mineral Water (Plant-Derived) to Immature Stages of Mosquito Vectors

    Functional mineral water and related products are popular in some Asian countries as health drinks and, recently, have been employed in agricultural crop production as well as pest control. This study aimed to investigate the survival of mosquito vectors exposed to plant-derived functional mineral water produced by terahertz technology. The terahertz-based functional mineral water used in the current study not only decreased the hatching of Culex quinquefasciatus (Say) larvae but also showed concentration-dependent toxicity to the 3rd instar larvae and pupae of the three mosquito species tested. Aedes albopictus (Skuse) and Cx. quinquefasciatus pupae were more susceptible to terahertz-based functional mineral water than the larval stage, as indicated by their lower LC50. Lower concentrations (Aedes aegypti (Linnaeus) hatching, it could potentially be used for vector control at the larvae and pupae stages. The larvicidal and pupicidal activity of diluted terahertz-based functional mineral water gradually diminished after 24 h, indicating that it is a biodegradable and eco-friendly bioinsecticide. However, as the terahertz-based functional mineral water is also toxic to larvivorous predatory-copepods, it should not be utilized in aquatic environments where predatory-based mosquito control programs are employed

The relevant experiments have mostly been conducted in cold and temperate ecosystems, but rarely in subtropical ecosystems.AimsIn this study, we aimed to construct a unified radial growth model for Cinnamomum kanehirae Hayata and to identify its sensitivity to temperature.MethodsThe wood radial increments were quantified for 3 years by either pinning or microcoring. The radial wood growth curves were modelled integratively by semiparametric regression and individually by curve fitting. The effects of tree social class, interannual and environmental factors on radial growth were analysed quantitatively.ResultsA unified S-shaped growth model for C. kanehirae was successfully constructed. By including the social class effect, the model was significantly improved. The maximum radial increment (A) was significantly correlated with the maximum growth rate (μ); both A and μ were significantly higher in dominant than in codominant trees. The time-varying radial growth rate was more sensitive to air temperature in dominant than in codominant trees.ConclusionSemiparametric models revealed an S-shaped growth curve of C. kanehirae and confirmed the higher temperature sensitivity of dominant trees compared to codominant trees in humid subtropical areas

    Neck Circumference as a Predictive Indicator of CKD for High Cardiovascular Risk Patients

    Background. Neck circumference (NC) is an anthropometric measure of obesity for upper subcutaneous adipose tissue distribution which is associated with cardiometabolic risk. This study investigated whether NC is associated with indicators of chronic kidney disease (CKD) for high cardiometabolic risk patients. Methods. A total of 177 consecutive patients who underwent the outpatient departments of cardiology were prospectively enrolled in the study. The patients were aged >20 years with normal renal function or with stages 1–4 CKD. A linear regression was performed using the Enter method to present an unadjusted R2, standardized coefficients, and standard error, and the Durbin-Watson test was used to assess residual independence. Results. Most anthropometric measurements from patients aged ≧65 were lower than those from patients aged <65, except for women’s waist circumference (WC) and waist hip ratio. Female NC obtained the highest R2 values for 24 hr CCR, uric acid, microalbuminuria, hsCRP, triglycerides, and HDL compared to BMI, WC, and hip circumference. The significances of female NC with 24 hr CCR and uric acid were improved after adjusted age and serum creatinine. Conclusions. NC is associated with indicators of CKD for high cardiometabolic risk patients and can be routinely measured as easy as WC in the future
