    Adhesive L1CAM-Robo Signaling Aligns Growth Cone F-Actin Dynamics to Promote Axon-Dendrite Fasciculation in C. elegans

    Neurite fasciculation through contact-dependent signaling is important for the wiring and function of the neuronal circuits. Here, we describe a type of axon-dendrite fasciculation in C. elegans, where proximal dendrites of the nociceptor PVD adhere to the axon of the ALA interneuron. This axon-dendrite fasciculation is mediated by a previously uncharacterized adhesive signaling by the ALA membrane signal SAX-7/L1CAM and the PVD receptor SAX-3/Robo but independent of Slit. L1CAM physically interacts with Robo and instructs dendrite adhesion in a Robo-dependent manner. Fasciculation mediated by L1CAM-Robo signaling aligns F-actin dynamics in the dendrite growth cone and facilitates dynamic growth cone behaviors for efficient dendrite guidance. Disruption of PVD dendrite fasciculation impairs nociceptive mechanosensation and rhythmicity in body curvature, suggesting that dendrite fasciculation governs the functions of mechanosensory circuits. Our work elucidates the molecular mechanisms by which adhesive axon-dendrite signaling shapes the construction and function of sensory neuronal circuits

    The polarity protein VANG-1 antagonizes Wnt signaling by facilitating Frizzled endocytosis

    Signaling that instructs the migration of neurons needs to be tightly regulated to ensure precise positioning of neurons and subsequent wiring of the neuronal circuits. Wnt-Frizzled signaling controls neuronal migration in metazoans, in addition to many other aspects of neural development. We show that Caenorhabditis elegans VANG-1, a membrane protein that acts in the planar cell polarity (PCP) pathway, antagonizes Wnt signaling by facilitating endocytosis of the Frizzled receptors. Mutations of vang-1 suppress migration defects of multiple classes of neurons in the Frizzled mutants, and overexpression of vang-1 causes neuronal migration defects similar to those of the Frizzled mutants. Our genetic experiments suggest that VANG-1 facilitates Frizzled endocytosis through β-arrestin2. Co-immunoprecipitation experiments indicate that Frizzled proteins and VANG-1 form a complex, and this physical interaction requires the Frizzled cysteine-rich domain. Our work reveals a novel mechanism mediated by the PCP protein VANG-1 that downregulates Wnt signaling through Frizzled endocytosis

    Towards Socially Intelligent Agents with Mental State Transition and Human Utility

    Building a socially intelligent agent involves many challenges, one of which is to track the agent's mental state transition and teach the agent to make rational decisions guided by its utility like a human. Towards this end, we propose to incorporate a mental state parser and utility model into dialogue agents. The hybrid mental state parser extracts information from both the dialogue and event observations and maintains a graphical representation of the agent's mind; Meanwhile, the utility model is a ranking model that learns human preferences from a crowd-sourced social commonsense dataset, Social IQA. Empirical results show that the proposed model attains state-of-the-art performance on the dialogue/action/emotion prediction task in the fantasy text-adventure game dataset, LIGHT. We also show example cases to demonstrate: (\textit{i}) how the proposed mental state parser can assist agent's decision by grounding on the context like locations and objects, and (\textit{ii}) how the utility model can help the agent make reasonable decisions in a dilemma. To the best of our knowledge, we are the first work that builds a socially intelligent agent by incorporating a hybrid mental state parser for both discrete events and continuous dialogues parsing and human-like utility modeling

    Shoupa: An AI System for Early Diagnosis of Parkinson's Disease

    Parkinson's Disease (PD) is a progressive nervous system disorder that has affected more than 5.8 million people, especially the elderly. Due to the complexity of its symptoms and its similarity to other neurological disorders, early detection requires neurologists or PD specialists to be involved, which is not accessible to most old people. Therefore, we integrate smart mobile devices with AI technologies. In this paper, we introduce the framework of our developed PD early detection system which combines different tasks evaluating both motor and non-motor symptoms. With the developed model, we help users detect PD punctually in non-clinical settings and figure out their most severe symptoms.     Late Cretaceous (~81Ma) high-temperature metamorphism in the southeastern Lhasa terrane: implication for the Neo-Tethys ocean ridge subduction.

    An integrated study of U-Pb zircon dating, geochemical and Sr-Nd-Hf isotopic compositions of garnet-bearing granulite and marble from the granulite-facies domain of the Nyingchi Complex (eastern Himalayan syntaxis) has provided insights into the tectonic evolution of the southern Lhasa terrane. The peak metamorphism of the garnet-bearing granulite is marked by a mineral assemblage of garnet + orthopyroxene + high-Ti amphibole + plagioclase + quartz + rutile. Abundant exsolved rutile needles are observed within amphibole, garnet and quartz. The peak metamorphic temperatures are estimated at 803–924 °C. Geochemical data from the garnet-bearing granulites provide evidence for a basaltic protolith that formed in a continental-margin arc setting. Sr-Nd-Hf isotopic compositions indicate that this protolith was sourced from partial melting of a depleted mantle. LA-ICP MS U-Pb zircon dating shows that the protolith age and metamorphic age of the garnet-bearing granulites are 89.3 ± 0.6 Ma and 81.1 ± 0.8 Ma, respectively. The detrital magmatic zircons from the marble yield ages from 86.3 to 167 Ma. The age distribution and Hf isotopic composition (εHf(t) = + 5.9 to + 17.5) of the detrital magmatic zircon in the marble are consistent with the isotopic data of zircons from the Jurassic–Cretaceous Gangdese batholiths, suggesting that the clastic sediments were partially derived from these intrusives or associated volcanic rocks, and deposited in the fore-arc basin of the Gangdese arc. The metamorphic zircons in the marble yield a metamorphic age of 81.4 ± 0.5 Ma. These results show that both the arc magmatic rocks and forearc sedimentary rocks underwent high-temperature (HT) granulite-facies metamorphism at ~ 81 Ma, indicating anomalously high heat input in the forearc region. A range of tectonic observations, including a coeval hiatus in arc magmatism and a period of regional uplift, indicate that HT metamorphism resulted from the subduction of the Neo-Tethys ocean ridge beneath the southern Lhasa terrane during the Late Cretaceous

    SocAoG: Incremental Graph Parsing for Social Relation Inference in Dialogues

    Inferring social relations from dialogues is vital for building emotionally intelligent robots to interpret human language better and act accordingly. We model the social network as an And-or Graph, named SocAoG, for the consistency of relations among a group and leveraging attributes as inference cues. Moreover, we formulate a sequential structure prediction task, and propose an α\alpha-β\beta-γ\gamma strategy to incrementally parse SocAoG for the dynamic inference upon any incoming utterance: (i) an α\alpha process predicting attributes and relations conditioned on the semantics of dialogues, (ii) a β\beta process updating the social relations based on related attributes, and (iii) a γ\gamma process updating individual's attributes based on interpersonal social relations. Empirical results on DialogRE and MovieGraph show that our model infers social relations more accurately than the state-of-the-art methods. Moreover, the ablation study shows the three processes complement each other, and the case study demonstrates the dynamic relational inference.Comment: Long paper (oral) accepted by ACL-IJCNLP 202
