    Centella asiatica extract protects against amyloid β1–40-induced neurotoxicity in neuronal cells by activating the antioxidative defence system

    AbstractCentella asiatica (雷公根 léi gōng gēn) is a traditional medicinal herb with high antioxidant activity, which decreases amyloid-β (Aβ) deposition in the brain. At the same time, aggregated Aβ-induced oxidative stress is the trigger in the pathogenesis of Alzheimer's disease (AD). Here, we investigated the ability of C. asiatica ethanol extract (CAE) to protect PC12 and IMR32 cells from Aβ1–40-induced production of reactive oxygen species (ROS) and concomitant neurotoxicity. Aggregated Aβ1–40 treatment resulted in reduced cell viability, which can be reversed by cotreatment with 25, 50, and 100 μg/mL CAE. Moreover, CAE eliminated the Aβ1–40-mediated increase in ROS production. Thus, CAE-mediated protection against aggregated Aβ1–40-induced neurotoxicity is attributable to modulation of the antioxidative defense system in cells, including the activities of superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase, and levels of glutathione and glutathione disulfide by CAE. This emphasizes the potential therapeutic and preventive value of CAE in the treatment of AD

    THE EFFECT OF REGULA EXERCISE FOR FUNTIONAL HEALTH OF THE ELDERLY

    The purpose of this study was to investigate the performance difference in walking, stair descent and ascent, sit-to-stand, and stand-to-sit, between elderlies who exercise regularly and those who don’t. Twenty elderlies participated in this study (8 males and 12 females). All tests were assessed using a 10-camera Vicon system. The visual3D software was used to analyse data. A positive correlation exists between movement time of stair descent and anterior-posterior balance ability. The negative correlations exist between movement time of stair descent and ascent and balance ability in lateral-medial. A significant difference in balance ability of stair decent exists between those with regular exercise and those without. According to the ACSM principle, regular exercise can indeed reduce the chance of falling in the elderly

    Spontaneous Arrangement of Two-way Flow in Water Bridge

    By revisiting the century-old problem of water bridge, we demonstrate that it is in fact dynamic and comprises of two coaxial currents that carry different charges and flow in opposite directions. Initially, the inner flow is facilitated by the cone jet that is powered by H+ and flows out of the anode beaker. The negative cone jet from cathode is established later and forced to take the outer route. This spontaneous arrangement of two-way flow is revealed by the use of chemical dyes, e.g., fluorescein and FeCl3, carbon powder, and the Particle Image Velocimetry. These two opposing flows are found to carry non-equal flux that results in a net transport of water to the cathode beaker. By combining the above information and taking into account the counter flow to equate the water level from the connecting pipe, we can estimate the cross section and flow speed of these co-axial flows as a function of time and applied voltage.Comment: 5 pages, 5 figure

    Exploring impacts of project leaders’ written expressions in virtual and fluid projects: The role of personality and emotion

    This paper aims to tackle challenges of managing projects in highly virtual and fluid contexts, characterized by diversity, dispersion, digital dependence, unstable membership, and dynamic coordination and configuration. We investigate project leaders’ personality and emotion expressed in written expression and examine their impacts on collaboration outcomes. IBM Watson Personality Insights and Tone Analyzer were adopted to assess the leader’s personality and emotion. A computation model to classify collaboration patterns into taskwork-related and teamwork-related communication is under development. We report preliminary findings based on 417 weekly meetings between October 2018 and February 2020 in 8 open-source software teams around WordPress. The research results have the potential to inform researchers and practitioners about what personality profiles and emotions should be considered to foster collaboration in virtual and fluid projects. It is possible to extend the boundary condition of the traits school of leadership for project management in the new context

    Cascaded Local Implicit Transformer for Arbitrary-Scale Super-Resolution

    Implicit neural representation has recently shown a promising ability in representing images with arbitrary resolutions. In this paper, we present a Local Implicit Transformer (LIT), which integrates the attention mechanism and frequency encoding technique into a local implicit image function. We design a cross-scale local attention block to effectively aggregate local features. To further improve representative power, we propose a Cascaded LIT (CLIT) that exploits multi-scale features, along with a cumulative training strategy that gradually increases the upsampling scales during training. We have conducted extensive experiments to validate the effectiveness of these components and analyze various training strategies. The qualitative and quantitative results demonstrate that LIT and CLIT achieve favorable results and outperform the prior works in arbitrary super-resolution tasks

    AMP-activated protein kinase activation mediates CCL3-induced cell migration and matrix metalloproteinase-2 expression in human chondrosarcoma

    Chemokine (C-C motif) ligand 3 (CCL3), also known as macrophage inflammatory protein-1α, is a cytokine involved in inflammation and activation of polymorphonuclear leukocytes. CCL3 has been detected in infiltrating cells and tumor cells. Chondrosarcoma is a highly malignant tumor that causes distant metastasis. However, the effect of CCL3 on human chondrosarcoma metastasis is still unknown. Here, we found that CCL3 increased cellular migration and expression of matrix metalloproteinase (MMP)-2 in human chondrosarcoma cells. Pre-treatment of cells with the MMP-2 inhibitor or transfection with MMP-2 specific siRNA abolished CCL3-induced cell migration. CCL3 has been reported to exert its effects through activation of its specific receptor, CC chemokine receptor 5 (CCR5). The CCR5 and AMP-activated protein kinase (AMPK) inhibitor or siRNA also attenuated CCL3-upregulated cell motility and MMP-2 expression. CCL3-induced expression of MMP-2 and migration were also inhibited by specific inhibitors, and inactive mutants of AMPK, p38 mitogen activated protein kinase (p38 or p38-MAPK), and nuclear factor κB (NF-κB) cascades. On the other hand, CCL3 treatment demonstrably activated AMPK, p38, and NF-κB signaling pathways. Furthermore, the expression levels of CCL3, CCR5, and MMP-2 were correlated in human chondrosarcoma specimens. Taken together, our results indicate that CCL3 enhances the migratory ability of human chondrosarcoma cells by increasing MMP-2 expression via the CCR5, AMPK, p38, and NF-κB pathways

    Mixed Sequence Reader: A Program for Analyzing DNA Sequences with Heterozygous Base Calling

    The direct sequencing of PCR products generates heterozygous base-calling fluorescence chromatograms that are useful for identifying single-nucleotide polymorphisms (SNPs), insertion-deletions (indels), short tandem repeats (STRs), and paralogous genes. Indels and STRs can be easily detected using the currently available Indelligent or ShiftDetector programs, which do not search reference sequences. However, the detection of other genomic variants remains a challenge due to the lack of appropriate tools for heterozygous base-calling fluorescence chromatogram data analysis. In this study, we developed a free web-based program, Mixed Sequence Reader (MSR), which can directly analyze heterozygous base-calling fluorescence chromatogram data in .abi file format using comparisons with reference sequences. The heterozygous sequences are identified as two distinct sequences and aligned with reference sequences. Our results showed that MSR may be used to (i) physically locate indel and STR sequences and determine STR copy number by searching NCBI reference sequences; (ii) predict combinations of microsatellite patterns using the Federal Bureau of Investigation Combined DNA Index System (CODIS); (iii) determine human papilloma virus (HPV) genotypes by searching current viral databases in cases of double infections; (iv) estimate the copy number of paralogous genes, such as β-defensin 4 (DEFB4) and its paralog HSPDP3

    Feasibility of Bispectral Index-Guided Propofol Infusion for Flexible Bronchoscopy Sedation: A Randomized Controlled Trial

    There are safety issues associated with propofol use for flexible bronchoscopy (FB). The bispectral index (BIS) correlates well with the level of consciousness. The aim of this study was to show that BIS-guided propofol infusion is safe and may provide better sedation, benefiting the patients and bronchoscopists.After administering alfentanil bolus, 500 patients were randomized to either propofol infusion titrated to a BIS level of 65-75 (study group) or incremental midazolam bolus based on clinical judgment to achieve moderate sedation. The primary endpoint was safety, while the secondary endpoints were recovery time, patient tolerance, and cooperation.The proportion of patients with hypoxemia or hypotensive events were not different in the 2 groups (study vs. control groups: 39.9% vs. 35.7%, p = 0.340; 7.4% vs. 4.4%, p = 0.159, respectively). The mean lowest blood pressure was lower in the study group. Logistic regression revealed male gender, higher American Society of Anesthesiologists physical status, and electrocautery were associated with hypoxemia, whereas lower propofol dose for induction was associated with hypotension in the study group. The study group had better global tolerance (p<0.001), less procedural interference by movement or cough (13.6% vs. 36.1%, p<0.001; 30.0% vs. 44.2%, p = 0.001, respectively), and shorter time to orientation and ambulation (11.7±10.2 min vs. 29.7±26.8 min, p<0.001; 30.0±18.2 min vs. 55.7±40.6 min, p<0.001, respectively) compared to the control group.BIS-guided propofol infusion combined with alfentanil for FB sedation provides excellent patient tolerance, with fast recovery and less procedure interference.ClinicalTrials. gov NCT00789815
