    "It is Quietly Chaotic. It Confuses Time"*: Final Report of Excavations at the Bordley-Randall Site in Annapolis, Maryland, 1993-1995 (18AP50)

Crowther's description of the Randall Court pathway (Cowther 1985).Part of the State Circle Site: http://hdl.handle.net/1903/11004During the summers of 1993, 1994, and 1995, the Archaeology in Annapolis project conducted excavations at the Bordley-Randall site (1 8AP50) in Annapolis, Maryland. The site now consists of the central portion of the block formed by North Street, College Avenue, Prince George Street, Maryland Avenue, and State Circle. The excavations were undertaken as part of the University of Maryland, College Park's Field School in Urban Archaeology and were organized to be support for dissertation research being done by Christopher Matthews of Columbia University. This report provides a background, summary, and interpretation of these archaeological investigations of the Bordley-Randall site. The site was tested in five areas: the Front Yard, the Back Yard, the West Wing Yard, the East Wing Yard, and the interior of the East Wing. The excavations revealed significant deposits from several different periods of occupation. These deposits show the progression of the site from the early Settlement Period in Annapolis through the Modem Period (as defined in Weissman 1986). In many areas of the site the excavations discovered deposits dating to the early 1700s when the site was first occupied and built on by Thomas Bordley. These deposits also helped to date the house and the East Wing to before 1748. Later alterations to the site, dating to the third quarter of the 18th century, were associated with the construction of and use of a terrace around the East Wing. The landscape of the front and rear yards were discovered to have been altered in the mid-19th century by the laying in of an extensive kitchen garden in the rear yard and the building of a park-like garden in the front. These alterations were predominantly defined by fill soils and the definition of garden paths. Later alterations made the city block fully modern as the street front lots were sold off and built over with businesses on Maryland Avenue and residences on the other streets beginning in the 1870s. In the interior, around 1895, an oval-shaped path was built in the front yard to which many of the new residences faced forming an enclosed, semi-private, semi-public space, now known as Randall Court. This space has remained essentially in tact since the early years of the 20th century. The appendices to this report include a transcription of several key historic documents related to the site, the report to the Maryland Humanities Council for funding in support of a public program at the site in 1995, the level and feature reports, and the staff qualifications.     Lonely Islands: Culture, Community, and Poverty in Archaeological Perspective

    ABSTRACT The study of poverty in historical archaeology has not yet developed a productive engagement with the complex political economy of impoverishment. A primary concern with culture and representation has instead supported the production of often essentialized subjects who ultimately mirror the problematic foundations of the "culture of poverty" thesis. This paper critiques the processes of constructing impoverished subjects and considers the notion of a "poverty of culture" as a relational position for analysis. Working in close collaboration with members of an impoverished African American community in Setauket, New York, alternative readings of poverty, culture, heritage, and archaeology are discussed. These alternatives serve as the foundations of a community-driven project informed by indigenous meanings and interests in the archaeological past in order to challenge the marginalization of this part of the broader local community

    Use of a Novel Imaging Technology for Remote Autism Diagnosis: A Reflection on Experience of Stakeholders

    AbstractTimely diagnosis and early interventions are critical to improving the long term functioning of a child with ASD. However, a major challenge facing parents is difficulty in obtaining on-time access to appropriate diagnostic services. To address this need, an imaging technology, NODA® (Naturalistic Observation Diagnostic Assessment), has been successfully developed and field-tested. NODA® includes 1) NODA SmartCapture; a smart-phone based recording system for parents to capture and share in-home video evidence of their child behavior and 2) NODA Connect; a HIPPA compliant web-platform for diagnosticians to conduct remote autism diagnostic assessments based on in-home video evidence of behavior, developmental history and their clinical judgment. In the field study, parents captured and shared videos evidence from their homes via NODA SmartCapture and diagnosticians conducted remote diagnostic assessment via NODA Connect. Results show that parents were able to successfully collect video evidence of behavior as per given prescription and diagnosticians were able to complete remote diagnostic assessments. This paper is a reflection on the first hand experience of key stakeholders (parents and diagnosticians) using NODA® in the field

    Inhibitory Input from the Lateral Hypothalamus to the Ventral Tegmental Area Disinhibits Dopamine Neurons and Promotes Behavioral Activation

    Projections from the lateral hypothalamus (LH) to the ventral tegmental area (VTA), containing both GABAergic and glutamatergic components, encode conditioned responses and control compulsive reward-seeking behavior. GABAergic neurons in the LH have been shown to mediate appetitive and feeding-related behaviors. Here we show that the GABAergic component of the LH-VTA pathway supports positive reinforcement and place preference, while the glutamatergic component mediates place avoidance. In addition, our results indicate that photoactivation of these projections modulates other behaviors, such as social interaction and perseverant investigation of a novel object. We provide evidence that photostimulation of the GABAergic LH-VTA component, but not the glutamatergic component, increases dopamine (DA) release in the nucleus accumbens (NAc) via inhibition of local VTA GABAergic neurons. Our study clarifies how GABAergic LH inputs to the VTA can contribute to generalized behavioral activation across multiple contexts, consistent with a role in increasing motivational salience.National Institute of Mental Health (U.S.) (Grant R01-MH102441-01

    Decoding Neural Circuits that Control Compulsive Sucrose Seeking

    SummaryThe lateral hypothalamic (LH) projection to the ventral tegmental area (VTA) has been linked to reward processing, but the computations within the LH-VTA loop that give rise to specific aspects of behavior have been difficult to isolate. We show that LH-VTA neurons encode the learned action of seeking a reward, independent of reward availability. In contrast, LH neurons downstream of VTA encode reward-predictive cues and unexpected reward omission. We show that inhibiting the LH-VTA pathway reduces “compulsive” sucrose seeking but not food consumption in hungry mice. We reveal that the LH sends excitatory and inhibitory input onto VTA dopamine (DA) and GABA neurons, and that the GABAergic projection drives feeding-related behavior. Our study overlays information about the type, function, and connectivity of LH neurons and identifies a neural circuit that selectively controls compulsive sugar consumption, without preventing feeding necessary for survival, providing a potential target for therapeutic interventions for compulsive-overeating disorder

    Direct Molecular Detection and Genotyping of Borrelia burgdorferi from Whole Blood of Patients with Early Lyme Disease

    Direct molecular tests in blood for early Lyme disease can be insensitive due to low amount of circulating Borrelia burgdorferi DNA. To address this challenge, we have developed a sensitive strategy to both detect and genotype B. burgdorferi directly from whole blood collected during the initial patient visit. This strategy improved sensitivity by employing 1.25 mL of whole blood, a novel pre-enrichment of the entire specimen extract for Borrelia DNA prior to a multi-locus PCR and electrospray ionization mass spectrometry detection assay. We evaluated the assay on blood collected at the initial presentation from 21 endemic area patients who had both physician-diagnosed erythema migrans (EM) and positive two-tiered serology either at the initial visit or at a follow-up visit after three weeks of antibiotic therapy. Results of this DNA analysis showed detection of B. burgdorferi in 13 of 21 patients (62%). In most cases the new assay also provided the B. burgdorferi genotype. The combined results of our direct detection assay with initial physician visit serology resulted in the detection of early Lyme disease in 19 of 21 (90%) of patients at the initial visit. In 5 of 21 cases we demonstrate the ability to detect B. burgdorferi in early Lyme disease directly from whole blood specimens prior to seroconversion
