Technology demonstrator of a four-stroke naturally aspirated spark-ignition engine
Diplomová práce se zabývá konstrukčním návrhem jednoválcového motoru, který slouží jako technický demonstrátor čtyřválcového nepřeplňovaného zážehového motoru Mazda Skyactiv-G. Úvod práce se zabývá konstrukcí nepřeplňovaných zážehových motorů a srovnáním jejich výkonnostních parametrů. Výpočtová část obsahuje silové působení v klikovém mechanismu. Výpočty z této kapitoly jsou poté využity při MKP výpočtu ojnice ve dvou kritických stavech.Master’s thesis deals with design of single cylinder engine which is technology demonstrator of four cylinder naturally aspirated spark ignition engine Mazda Skyactiv-G. Outset of this thesis deals with construction of modern naturally aspirated engines and with comparison of their parameters. Computational part consists of forces in crank mechanism. These calculations are later used in FEM analysis of connecting rod.

A novel method for detection of covered conductor faults in medium voltage overhead line systems
Although there aremany reasons whymedium voltage (MV) overhead lines with covered conductors (CCs) are used instead of aluminum-conductor steel-reinforced cables, the operation of CCs takes into account a difficult detection of faults, e.g., the contact of a tree branch with a CC. In this paper, we continue with the previous work on an electric voltage-based method enabling the detection of the CC faults; we introduce a new indicator of the faults based on the frequency of peaks in the signal of partial discharges. We mainly demonstrate the fault detection in a real MV distribution system, including a number of fault detectors; a fault is detected in 99.2% of occurrences within a long-term testing, although the cost of the detector is minimized in comparison with other similar fault detectors.Web of Science65155254
MLMD—A Malware-Detecting Antivirus Tool Based on the XGBoost Machine Learning Algorithm
This paper focuses on training machine learning models using the XGBoost and extremely randomized trees algorithms on two datasets obtained using static and dynamic analysis of real malicious and benign samples. We then compare their success rates—both mutually and with other algorithms, such as the random forest, the decision tree, the support vector machine, and the naïve Bayes algorithms, which we compared in our previous work on the same datasets. The best performing classification models, using the XGBoost algorithm, achieved 91.9% detection accuracy and 98.2% sensitivity, 0.853 AUC, and 0.949 F1 score on the static analysis dataset, and 96.4% accuracy and 98.5% sensitivity, 0.940 AUC, and 0.977 F1 score on the dynamic analysis dataset. Then, we exported the best performing machine learning models and used them in our proposed MLMD program, automating the process of static and dynamic analysis and allowing the trained models to be used for classification on new samples
βcatenin is a marker of poor clinical characteristics and suppressed immune infiltration in testicular germ cell tumors
Abstract Background WNT/βcatenin (WNTβ) pathway is activated in early stages of embryonic development. We aimed to evaluate the significance of βcatenin in germ cell tumors (GCTs) and explore associations with the inflamed environment. Methods Surgical specimens from 247 patients were analyzed. Βcatenin expression was detected in the tumor tissue by immunohistochemistry and correlated with clinical characteristics, outcome, PD-L1 expression and systemic immune-inflammation index (SII). The Ingenuity Pathway Analysis (IPA) was used to investigate the immune-cell related effects of βcatenin and PD-L1 encoding genes. Results βcatenin was expressed in 86.2% of GCTs. The expression in seminomas was significantly lower compared to all subtypes of non-seminoma (all P 150) was associated with primary mediastinal non-seminoma (P = 0.035), intermediate/poor risk disease (P = 0.033) and high tumor markers (P = 0.035). We observed a positive correlation with the PD-L1 in tumor and an inverse correlation with the SII. IPA uncovered relationships of CTNNB (βcatenin) and CD274 (PD-L1) genes and their effects on differentiation, proliferation and activation of lymphocyte subtypes. Conclusion Herein, we showed that βcatenin is associated with male adult GCT characteristics as well as supressed immune environment
