Modality-Independent Teachers Meet Weakly-Supervised Audio-Visual Event Parser

Audio-visual learning has been a major pillar of multi-modal machine
learning, where the community mostly focused on its modality-aligned setting,
i.e., the audio and visual modality are both assumed to signal the prediction
target. With the Look, Listen, and Parse dataset (LLP), we investigate the
under-explored unaligned setting, where the goal is to recognize audio and
visual events in a video with only weak labels observed. Such weak video-level
labels only tell what events happen without knowing the modality they are
perceived (audio, visual, or both). To enhance learning in this challenging
setting, we incorporate large-scale contrastively pre-trained models as the
modality teachers. A simple, effective, and generic method, termed Visual-Audio
Label Elaboration (VALOR), is innovated to harvest modality labels for the
training events. Empirical studies show that the harvested labels significantly
improve an attentional baseline by 8.0 in average F-score (Type@AV).
Surprisingly, we found that modality-independent teachers outperform their
modality-fused counterparts since they are noise-proof from the other
potentially unaligned modality. Moreover, our best model achieves the new
state-of-the-art on all metrics of LLP by a substantial margin (+5.4 F-score
for Type@AV). VALOR is further generalized to Audio-Visual Event Localization
and achieves the new state-of-the-art as well. Code is available at:

High-Reliability Trigate Poly-Si Channel Flash Memory Cell With Si-Nanocrystal Embedded Charge-Trapping Layer

Abstract-This letter introduces a polycrystalline-silicon nanowire (NW) thin-film nonvolatile memory (NVM) with a self-assembled silicon-nanocrystal (Si-NC) embedded chargetrapping (CT) layer. This process is simple and compatible with conventional CMOS processes. Experimental results indicate that this NW NVM exhibits high reliability due to a deep-quantum-well structure and immunity of enhanced electric field underneath a disk-shaped Si-NC. After 10 000 P/E cycles, the memory window loss of the NVM with a Si-NC embedded CT layer is less than 12% until 10 4 s at 150 • C. Accordingly, a poly-Si thin-film transistor with a Si-NC embedded CT layer is highly promising for NVM applications. Index Terms-Nanocrystal (NC), nonvolatile memory (NVM), thin-film transistor (TFT)
Alternative Complement Pathway Is Activated and Associated with Galactose-Deficient IgA1 Antibody in IgA Nephropathy Patients

BackgroundGalactose-deficient IgA1 (Gd-IgA1) and alternative complement pathway activation are considered to be involved in the pathogenesis of IgA nephropathy (IgAN). Nevertheless, the relationships between alternative pathway activation and disease activity or Gd-IgA1 level remains unclear.MethodsNinety-eight biopsy-diagnosed IgAN, twenty-five primary focal segmental sclerosis (FSGS) patients and forty-two healthy individuals were recruited in this study. Among them, fifty IgAN patients received immunosuppression. Follow-up blood samples at 1 and 3~6 months after immunosuppression were collected. Plasma levels of complement C5a, factor Ba and Gd-IgA1 were measured and analyzed. Immunostaining for complement was performed in twenty-five IgAN and FSGS patients.ResultsAt baseline, IgAN patients had higher levels of plasma C5a, factor Ba and Gd-IgA1 than control subjects. Gd-IgA1 levels positively correlated with plasma C5a and factor Ba. In addition, levels of factor Ba and Gd-IgA1 were positively associated with proteinuria and negatively associated with renal function. Immunostaining revealed positive staining for factor Bb and C3c in glomeruli in IgAN patients, but not in FSGS patients. At baseline, patients receiving immunosuppression had more severe proteinuria and higher factor Ba. After 6 months, eGFR declined and proteinuria persisted in patients without immunosuppression. In contrast, patients who received immunosuppression exhibited decreased plasma levels of C5a, factor Ba, and Gd-IgA1 as early as 1 month after treatment. Proteinuria decreased and renal function also remained stable 6 months after immunosuppression.ConclusionsOur results indicate a close relationship between alternative complement pathway activation, Gd-IgA1 concentration and clinical severity of IgAN. Level of complement factor B may be a potential marker for disease activity and therapeutic target in IgAN patients

BACKGROUND: Enterovirus 71 (EV71) has caused several epidemics of hand, foot and mouth diseases (HFMD) in Asia and now is being recognized as an important neurotropic virus. Effective medications and prophylactic vaccine against EV71 infection are urgently needed. Based on the success of inactivated poliovirus vaccine, a prototype chemically inactivated EV71 vaccine candidate has been developed and currently in human phase 1 clinical trial. PRINCIPAL FINDING: In this report, we present the development of a serum-free cell-based EV71 vaccine. The optimization at each step of the manufacturing process was investigated, characterized and quantified. In the up-stream process development, different commercially available cell culture media either containing serum or serum-free was screened for cell growth and virus yield using the roller-bottle technology. VP-SFM serum-free medium was selected based on the Vero cell growth profile and EV71 virus production. After the up-stream processes (virus harvest, diafiltration and concentration), a combination of gel-filtration liquid chromatography and/or sucrose-gradient ultracentrifugation down-stream purification processes were investigated at a pilot scale of 40 liters each. Although the combination of chromatography and sucrose-gradient ultracentrifugation produced extremely pure EV71 infectious virus particles, the overall yield of vaccine was 7-10% as determined by a VP2-based quantitative ELISA. Using chromatography as the downstream purification, the virus yield was 30-43%. To retain the integrity of virus neutralization epitopes and the stability of the vaccine product, the best virus inactivation was found to be 0.025% formalin-treatment at 37 °C for 3 to 6 days. Furthermore, the formalin-inactivated virion vaccine candidate was found to be stable for >18 months at 4 °C and a microgram of viral proteins formulated with alum adjuvant could induce strong virus-neutralizing antibody responses in mice, rats, rabbits, and non-human primates. CONCLUSION: These results provide valuable information supporting the current cell-based serum-free EV71 vaccine candidate going into human Phase I clinical trials

In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to provide realistic and reasonable critiques of reports that are focused on these processes. These guidelines are not meant to be a dogmatic set of rules, because the appropriateness of any assay largely depends on the question being asked and the system being used. Moreover, no individual assay is perfect for every situation, calling for the use of multiple techniques to properly monitor autophagy in each experimental setting. Finally, several core components of the autophagy machinery have been implicated in distinct autophagic processes (canonical and noncanonical autophagy), implying that genetic approaches to block autophagy should rely on targeting two or more autophagy-related genes that ideally participate in distinct steps of the pathway. Along similar lines, because multiple proteins involved in autophagy also regulate other cellular pathways including apoptosis, not all of them can be used as a specific marker for bona fide autophagic responses. Here, we critically discuss current methods of assessing autophagy and the information they can, or cannot, provide. Our ultimate goal is to encourage intellectual and technical innovation in the field

Expression profile of metastasis-related genes in invasive oral cancers

We used suppression subtractive
hybridization (SSH) and oligonucleotide microarray to
differentiate expression profiles of metastasis-related
genes and to evaluate their clinical significance in
patients with invasive oral cancer (OCa).
Overexpression of the specific genes was confirmed by
reverse transcription-PCR (RT-PCR). Cells expressing
the gene were identified by immunohistochemistry in
pathology specimens. Clinical correlation and
significance were analyzed statistically. Using these
methods, we detected increased expressions of MMP-1,
-3, -7, -9, -10 and interleukin (IL)-8 in invasive OCa.
Moreover, our data showed that overexpressions of
MMP-1, -3, -7, -10 and IL-8 were associated with
reduced survival
