    Assessing the Integrity of Spillway Foundations

    The erosion under a spillway can be a long-term issue that threatens the structural integrity of a water reservoir. The spillway under investigation was suspected to be defective after it had been commissioned in 1987 (Figure 1). Potholes and subsurface cavities were confirmed in the safety assessment using various NDT techniques including ground penetrating radar and impact echo. The GPR inspection was able to differentiate the intact region from the cavities under concrete slabs (Figure 2). The impact echo results and associated analyses provided further evidence of inferior condition in the soil under the concrete slabs. The engineering team designed and executed the repair project based on the conclusion of the integrity assessment. Subsequent GPR inspection has been performed so as to monitor the integrity of the spillway in a period of 18 months following the repair

    A novel regulatory event-based gene set analysis method for exploring global functional changes in heterogeneous genomic data sets

    <p>Abstract</p> <p>Background</p> <p>Analyzing gene expression data by assessing the significance of pre-defined gene sets, rather than individual genes, has become a main approach in microarray data analysis and this has promisingly derive new biological interpretations of microarray data. However, the detection power of conventional gene list or gene set-based approaches is limited on highly heterogeneous samples, such as tumors.</p> <p>Results</p> <p>We developed a novel method, the regulatory <b>e</b>vent-based <b>G</b>ene <b>S</b>et <b>A</b>nalysis (eGSA), which considers not only the consistently changed genes but also every gene regulation (event) of each sample to overcome the detection limit. In comparison with conventional methods, eGSA can detect functional changes in heterogeneous samples more precisely and robustly. Furthermore, by utilizing eGSA, we successfully revealed novel functional characteristics and potential mechanisms of very early hepatocellular carcinoma (HCC).</p> <p>Conclusion</p> <p>Our study creates a novel scheme to directly target the major cellular functional changes in heterogeneous samples. All potential regulatory routines of a functional change can be further analyzed by the regulatory event frequency. We also provide a case study on early HCCs and reveal a novel insight at the initial stage of hepatocarcinogenesis. eGSA therefore accelerates and refines the interpretation of heterogeneous genomic data sets in the absence of gene-phenotype correlations.</p

    Plasma levels in sepsis patients of annexin A1, lipoxin A4, macrophage inflammatory protein-3a, and neutrophil gelatinase-associated lipocalin

    AbstractBackgroundThe relationship between the various cytokine responses that occur during sepsis remains controversial. Emerging evidence indicates that the proinflammatory and anti-inflammatory responses are regulated simultaneously from the beginning of sepsis. However, the roles of the novel anti-inflammatory mediators annexin (Anx)A1 and lipoxin (LX)A4 and the proinflammatory cytokines neutrophil gelatinase-associated lipocalin (NGAL) and macrophage inflammatory protein (MIP)-3a have been studied.MethodsIn this study, the plasma levels of AnxA1, LXA4, NGAL, MIP-3a, interleukin (IL)-8 and IL-6 in patients with sepsis were determined on admission to the intensive care unit. The patients were classified into survivors and non-survivors based on their outcome on day 28.ResultsAnxA1 and LXA4 levels were decreased in sepsis patients compared with control patients, whereas the levels of the proinflammatory cytokines MIP-3a, NGAL, IL-8, and IL-6 were elevated. Furthermore, a significantly higher level of MIP-3a was detected in nonsurviving patients compared with surviving patients (p < 0.05), whereas there were no significant differences between these two groups for the levels of the other mediators. Correlation analysis demonstrated that only NGAL level was closely correlated with the level of IL-6. Univariate analysis indicated that the levels of MIP-3a and IL-8 were independent factors associated with patient survival, but this was not confirmed by the multivariate analysis.ConclusionAnxA1 and LXA4 plasma levels were found to be decreased in sepsis patients, whereas the levels of MIP-3a and NGAL were found to be elevated. This warrants further study in order to determine the clinical implications of these changes

    Bioprocess intelligent for the improvements and prediction on fed-batch cell culture in bioreactor

    With advances in biotechnology, the antibody productivity is not the only issue in biopharmaceutical manufacturing; moreover, how to control the quality and quantity of antibody production in bioprocess has become prominent. In typical fed-batch cell culture, it is not easy to control the dynamic cultivation and feeding conditions. The study is to present an intelligent bioprocess make use of design of experiments (DOE) and multivariate data analysis (MVDA). In the culture medium optimization, we performed the medium screening with the DOE method in shake spin tubes and shake flasks. DOE provides a cost-effective methodology for medium development and optimization, and furthermore we utilized multivariate data analysis methodology to build up the fed-batch intelligence bioprocess in 5 L bioreactor. We analyzed mass data transferred from 5 L bioreactor process and established the fed-batch culture model with SIMCA software. Combination of the batch process and big data sets, we can be easy to do batch-to-batch comparison, cell culture profile prediction, and key parameter finding to improve the process performance with more steady product quality and less error. In this fed-batch culture model, we achieved more than 5 g/L cell productivity consistently and predictably in 5 L bioreactor cultivation with CRISPR-mediated targeted gene site-specific integration CHO cell line
