Molecular epidemiology and genetic diversity of human astrovirus in South Korea from 2002 to 2007
AbstractThe present study was conducted to survey the prevalence and genotypic distribution of human astrovirus (HAstV) circulating in South Korea. Of 160,027 patients with acute gastroenteritis, 2,057 (1.3%) were positive for HAstV antigen. We determined the genotypes of 187 HAstV strains collected from laboratories across the country. Genetic analysis revealed genotype 1 to be the most prevalent, accounting for 72.19% of the strains, followed by genotypes 8 (9.63%), 6 (6.95%), 4 (6.42%), 2 (3.21%) and 3 (1.60%). Our findings indicate that HAstV is less common but, even so, a potentially important viral agent of gastroenteritis in South Korea, with significant genetic diversity among circulating HAstV strains

Synaptic processes and immune-related pathways implicated in Tourette syndrome.
Tourette syndrome (TS) is a neuropsychiatric disorder of complex genetic architecture involving multiple interacting genes. Here, we sought to elucidate the pathways that underlie the neurobiology of the disorder through genome-wide analysis. We analyzed genome-wide genotypic data of 3581 individuals with TS and 7682 ancestry-matched controls and investigated associations of TS with sets of genes that are expressed in particular cell types and operate in specific neuronal and glial functions. We employed a self-contained, set-based association method (SBA) as well as a competitive gene set method (MAGMA) using individual-level genotype data to perform a comprehensive investigation of the biological background of TS. Our SBA analysis identified three significant gene sets after Bonferroni correction, implicating ligand-gated ion channel signaling, lymphocytic, and cell adhesion and transsynaptic signaling processes. MAGMA analysis further supported the involvement of the cell adhesion and trans-synaptic signaling gene set. The lymphocytic gene set was driven by variants in FLT3, raising an intriguing hypothesis for the involvement of a neuroinflammatory element in TS pathogenesis. The indications of involvement of ligand-gated ion channel signaling reinforce the role of GABA in TS, while the association of cell adhesion and trans-synaptic signaling gene set provides additional support for the role of adhesion molecules in neuropsychiatric disorders. This study reinforces previous findings but also provides new insights into the neurobiology of TS
The surface detector array of the Telescope Array experiment
The Telescope Array (TA) experiment, located in the western desert of
Utah,USA, is designed for observation of extensive air showers from extremely
high energy cosmic rays. The experiment has a surface detector array surrounded
by three fluorescence detectors to enable simultaneous detection of shower
particles at ground level and fluorescence photons along the shower track. The
TA surface detectors and fluorescence detectors started full hybrid observation
in March, 2008. In this article we describe the design and technical features
of the TA surface detector.Comment: 32 pages, 17 figure
New air fluorescence detectors employed in the Telescope Array experiment
Since 2007, the Telescope Array (TA) experiment, based in Utah, USA, has been
observing ultra high energy cosmic rays to understand their origins. The
experiment involves a surface detector (SD) array and three fluorescence
detector (FD) stations. FD stations, installed surrounding the SD array,
measure the air fluorescence light emitted from extensive air showers (EASs)
for precise determination of their energies and species. The detectors employed
at one of the three FD stations were relocated from the High Resolution Fly's
Eye experiment. At the other two stations, newly designed detectors were
constructed for the TA experiment. An FD consists of a primary mirror and a
camera equipped with photomultiplier tubes. To obtain the EAS parameters with
high accuracies, understanding the FD optical characteristics is important. In
this paper, we report the characteristics and installation of new FDs and the
performances of the FD components. The results of the monitored mirror
reflectance during the observation time are also described in this report.Comment: 44 pages, 23 figures, submitted to NIM-
Genome-wide bidirectional CRISPR screens identify mucins as host factors modulating SARS-CoV-2 infection
Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) causes a range of symptoms in infected individuals, from mild respiratory illness to acute respiratory distress syndrome. A systematic understanding of host factors influencing viral infection is critical to elucidate SARS-CoV-2âhost interactions and the progression of Coronavirus disease 2019 (COVID-19). Here, we conducted genome-wide CRISPR knockout and activation screens in human lung epithelial cells with endogenous expression of the SARS-CoV-2 entry factors ACE2 and TMPRSS2. We uncovered proviral and antiviral factors across highly interconnected host pathways, including clathrin transport, inflammatory signaling, cell-cycle regulation, and transcriptional and epigenetic regulation. We further identified mucins, a family of high molecular weight glycoproteins, as a prominent viral restriction network that inhibits SARS-CoV-2 infection in vitro and in murine models. These mucins also inhibit infection of diverse respiratory viruses. This functional landscape of SARS-CoV-2 host factors provides a physiologically relevant starting point for new host-directed therapeutics and highlights airway mucins as a host defense mechanism
Highly-parallelized simulation of a pixelated LArTPC on a GPU
The rapid development of general-purpose computing on graphics processing units (GPGPU) is allowing the implementation of highly-parallelized Monte Carlo simulation chains for particle physics experiments. This technique is particularly suitable for the simulation of a pixelated charge readout for time projection chambers, given the large number of channels that this technology employs. Here we present the first implementation of a full microphysical simulator of a liquid argon time projection chamber (LArTPC) equipped with light readout and pixelated charge readout, developed for the DUNE Near Detector. The software is implemented with an end-to-end set of GPU-optimized algorithms. The algorithms have been written in Python and translated into CUDA kernels using Numba, a just-in-time compiler for a subset of Python and NumPy instructions. The GPU implementation achieves a speed up of four orders of magnitude compared with the equivalent CPU version. The simulation of the current induced on 10^3 pixels takes around 1 ms on the GPU, compared with approximately 10 s on the CPU. The results of the simulation are compared against data from a pixel-readout LArTPC prototype
Factors affecting the measurement accuracy of ITER neutron activation system
International audienceOne of the main purposes of the ITER(2) neutron activation system (NAS) is to evaluate the total neutron production rate from all over the plasma. The measurement accuracy depends on the position and profile of the plasma and the material in front of the irradiation end. It is required to minimize the amount of material and its density variation across the field of view between the plasma and the irradiation end. Due to the radiation and thermal environment of the ITER in-vessel, however, the measurement from ITER NAS cannot avoid the strong influence from in-vessel materials such as the diagnostic first wall, blanket modules, and divertor cassettes, those are located near the irradiation ends. In order to improve the reliability of the measurement in such environment, special cutouts in the diagnostic first wall are introduced near the irradiation end structures located in the port plugs. The effect of the materials and the position and profile of the neutron source in the plasma are evaluated for these irradiation locations, as well as the ones under the divertor cassettes and between blanket modules, by the neutron transport calculation. Calculation results show that simultaneous measurements at upper port and divertor location can provide highly accurate results even without a position or profile correction from other diagnostics
