    Relating Gene Expression Evolution with CpG Content Changes

    Background: Previous studies have shown that CpG dinucleotides are enriched in a subset of promoters and the CpG content of promoters is positively correlated with gene expression levels. But the relationship between divergence of CpG content and gene expression evolution has not been investigated. Here we calculate the normalized CpG (nCpG) content in DNA regions around transcription start site (TSS) and transcription terminal site (TTS) of genes in nine organisms, and relate them with expression levels measured by RNA-seq. Results: The nCpG content of TSS shows a bimodal distribution in all organisms except platypus, whereas the nCpG content of TTS only has a single peak. When the nCpG contents are compared between different organisms, we observe a different evolution pattern between TSS and TTS: compared with TTS, TSS exhibits a faster divergence rate between closely related species but are more conserved between distant species. More importantly, we demonstrate the link between gene expression evolution and nCpG content changes: up-/down- regulation of genes in an organism is accompanied by the nCpG content increase/decrease in their TSS and TTS proximal regions. Conclusions: Our results suggest that gene expression changes between different organisms are correlated with the alterations in normalized CpG contents of promoters. Our analyses provide evidences for the impact of nCpG content on gene expression evolution

    Visualizing the impact of Covid-19 vaccine passports on pedestrian access to metro stations in Hong Kong

    Pedestrian infrastructures in Hong Kong enable multilevel city life in a vertical metropolis plagued by land scarcity. Public spaces integrated into pedestrian networks play an indispensable role in neighbourhood accessibility. We visualize the impact of the Covid-19 vaccine passport (VP) restrictions on the use of public space on pedestrian accessibility to all 97 metro stations in Hong Kong. Pedestrians without a vaccine passport (PwoVP) need to walk significantly longer alternative routes. Specifically, VP-related access restrictions to indoor walkways have doubled the shortest travel time for PwoVP and a 50% reduction in accessibility of two-thirds of stations

    Exploring public attention and sentiment toward carbon neutrality: evidence from Chinese social media Sina Weibo

    IntroductionExploring the public’s cognition toward carbon neutrality is conducive to improving the quality and effectiveness of policymaking, and promoting the realization of carbon neutrality goals. This study aims to explore the public’s attention and sentiment toward carbon neutrality from the perspective of social psychology.MethodsUsing posts on carbon neutrality from the Chinese social media platform Sina Weibo as the data source, this study uses statistical analysis, the Mann-Kendall method, keyword analysis, the BERT model, and the LDA model to explore public attention and sentiment.ResultsThe results show that: (1) men, people living east of the Hu line (economically developed regions), and the public in the energy finance market are more concerned about carbon neutrality; (2) high public attention and great dynamic changes in public attention toward carbon neutrality could be trigged by highly credible government or international governmental organizations’ information; (3) public sentiment toward carbon neutrality is mostly positive; however, specific topics affect public sentiment differently.DiscussionThe research results contribute to policymakers’ better understanding of the trend of public attention and sentiment toward carbon neutrality, and support improvements in the quality and impact of policymaking

    SECC: A Novel Search Engine Interface with Live Chat Channel

    Traditional information retrieval systems rank documents according to their relevance to users' input queries. State of the art commercial search engines (SEs) train ranking models and suggest query refinements by exploiting collective intelligence implicitly using global users' query logs. However, they do not provide an explicit channel for users to communicate with each other in the search process. By asking or discussing with other users on the fly, a user could find relevant information more conveniently and gain a better search experience. In this paper, we present a demo of novel Search Engine with a live Chat Channel (SECC). SECC can group users automatically based on their input queries and allow them to communicate with each other in real time through a chat interface
