    Quercetin protects human thyroid cells against cadmium toxicity

    Various natural compounds have been successfully tested for preventing or counteracting the toxic effects of exposure to heavy metals. In this study, we analyzed the effects of cadmium chloride (CdCl2) on immortalized, non\u2010tumorigenic thyroid cells Nthy\u2010ori\u20103\u20101. We investigated the molecular mechanism underlying its toxic action as well as the potential protective effect of quercetin against CdCl2\u2010induced damage. CdCl2 suppressed cell growth in a dose\u2010 and time\u2010dependent manner (IC50 value ~10 \u3bcM) associated with a decrease in levels of phospho\u2010ERK. In addition, CdCl2 elicited an increase in reactive oxygen species (ROS) production and lipid peroxidation. A significant increase in GRP78, an endoplasmic reticulum (ER) stress\u2010related protein, was also observed. Supplementation of quercetin counteracted the growth\u2010inhibiting action of CdCl2 by recovering ERK protein phosphorylation levels, attenuating ROS overproduction, decreasing MDA content and reducing the expression of GRP78 in cells exposed to CdCl2. Thus, in addition to revealing the molecular effects involved in cadmium\u2010induced toxicity, the present study demonstrated, for the first time, a protective effect of quercetin against cadmium\u2010induced damages to normal thyroid cells

    Raman Microspectroscopy Analysis in the Treatment of Acanthamoeba Keratitis

    Acanthamoeba keratitis is a rare but serious corneal disease, often observed in contact lens wearers. Clinical treatment of infected patients frequently involves the use of polyhexamethylene biguanide (PHMB), a polymer used as a disinfectant and antiseptic, which is toxic also for the epithelial cells of the cornea. Prompt and effective diagnostic tools are hence highly desiderable for both starting early therapy and timely suspension of the treatment. In this work we use Raman microspectroscopy to analyse in vitro a single Acanthamoeba cell in cystic phase. In particular, we investigate the effect of PHMB at the single-cell level, providing useful information on both the underlying biochemical mechanism and the time frame for Acanthamoeba eradication in ocular infections. Furthermore, we demonstrate that Raman spectroscopy, in conjunction with standard multivariate analysis methods, allows discriminating between live and dead Acanthamoebas, which is fundamental to optimizing patients' treatment

    RUOLO DELLA RM-DWI INTEGRATA DAL PARAMETRO ADC NELLO STUDIO DEL COLESTEATOMA

    : La Risonanza Magnetica pesata in diffusione (RM-DWI) distingue qualitativamente il colesteatoma dal tessuto di granulazione nei pazienti con otite media cronica. Il calcolo del coefficiente di diffusione apparente (ADC) consente un'analisi quantitativa identificando un cut-off numerico che differenzia le due tipologie tissutali. MATERIALI E METODI: lo studio retrospettivo ha riguardato 100 pazienti sottoposti a Timpanoplastica per colesteatoma primario o residuo/recidivante. Tutti i pazienti sono stati sottoposti a RM-DWI preoperatoria tra Aprile 2015 e Marzo 2019, con calcolo di ADC (R-version 3.2.5). La presenza di colesteatoma è stata accertata intraoperatoriamente e confermata istologicamente. RISULTATI: i reperti chirurgici di 100 pazienti sono stati confrontati con i valori medi ADC ed analizzata la distribuzione di quest’ultimi per colesteatoma/tessuto di granulazione, evidenziando due gruppi. La differenza nel valore medio di ADC tra colesteatoma (0,86 × 10-3mm2/s) e tessuto di granulazione (2.22×10-3mm2/s) ha rilevato una differenza statisticamente significativa di ADC per i pazienti con/senza colesteatoma (p <2.2 × 10-16). Il modello binomiale di regressione logistica prevede la probabilità dello stato patologico, mediante ADC. Il potere predittivo di questo modello è stato misurato mediante curva ROC e applicando la statistica Youden J sulla curva abbiamo stabilito un valore di probabilità di ~ 0,6 per definire un cut-off (specificità = 1.0 sensibilità = 0.9861111) pari a ADC=1,71×10-3mm2/s. CONCLUSIONI: lo studio ha evidenziato che è possibile stabilire un cut-off di ADC per identificare pre-operatoriamente con elevata significatività statistica la presenza di matrice colesteatomatosa. In tal modo la RM-DWI diviene una “tool diagnostico” utile ed affidabile in quei casi in cui il colesteatoma non è certamente evidenziabile in otomicroscopia e/o nel follow-up di pazienti già sottoposti a primo tempo chirurgico

    A novel helitron from the genome of the icefish species Chionodraco hamatus, belonging to the Channichthyidae, the most derived Notothenioids family, was isolated, characterized and designated as HeliNoto (8.9 kb). Its ORF was compared to homologous sequences from different species in a comprehensive phylogenetic analysis. For the first time the putative functional domains of a helitron were subjected to a well accurate structural analysis including chromosomal localization. Finally, the distribution of HeliNoto among Notothenioids was investigated

    Differential DMRT1 expression in the gonads of Podarcis sicula (Reptilia: Lacertidae)

    DMRT genes encode a large family of transcription factors which share an unusual cysteine-rich DNA-binding motif, the DM domain. DM family members have been studied in the context of sexual development; in particular, the DMRT1 gene appeared to be the one most directly involved in sex determination, but its activity is largely unexplored and possible downstream targets of this factor have yet to be identified. DMRT1 of the lacertid lizard Podarcis sicula (PsDMRT1) was isolated as a model to study differential gene expression during the seasonal reproductive cycle of an ectothermal species. The adult testis of P. sicula exhibits full activity in spring, complete regression in summer and a slow autumnal recrudescence without spermiation. We cloned a 591-bp partial ORF of the PsDMRT1 fragment, whose putative amino acid sequence contains the conserved DM domain. Northern blot analysis of mRNA from different tissues of P. sicula individuals captured in spring demonstrated DMRT1 transcripts only in testis. Semi-quantitative RT-PCR and in situ hybridization experiments showed peak PsDMRT1 expression in spring, lower expression in autumn and no expression during the period of gonad regression. A possible correlation between androgen level variations and PsDMRT1 transcripts is hypothesized and discussed. Finally, data showed that PsDMRT1 is expressed only in spermatogenic cells, before the second meiotic division, suggesting that its role is confined to the proliferation and maintenance of spermatogonia and spermatocytes

    A chromosome study was carried out on a number of European and Central Asiatic diploid green toad populations by means of standard and various other chromosome banding and staining methods (Ag-NOR-, Q-, CMA3-, late replicating [LR] banding pattern, C-and sequential C-banding + CMA3 + DAPI). This study revealed the remarkable karyological uniformity of specimens from all populations, with the only exception being specimens from a Moldavian population, where one chromosome pair was heteromorphic. Though similar in shape, size and with an identical heterochromatin distribution,the difference in the heteromorphic pair was due to a large inverted segment on its long arms. This heteromorphism was restricted to females, suggesting a female heterogametic sex chromosome system of ZZ/ZW type at a very early step of differentiation
