Effectiveness of specific RNA-mediated interference through ingested double-stranded RNA in Caenorhabditis elegans.
BACKGROUND: In Caenorhabditis elegans, injection of double-stranded RNA (dsRNA) results in the specific inactivation of genes containing homologous sequences, a technique termed RNA-mediated interference (RNAi). It has previously been shown that RNAi can also be achieved by feeding worms Escherichia coli expressing dsRNA corresponding to a specific gene; this mode of dsRNA introduction is conventionally considered to be less efficient than direct injection, however, and has therefore seen limited use, even though it is considerably less labor-intensive. RESULTS: Here we present an optimized feeding method that results in phenotypes at least as strong as those produced by direct injection of dsRNA for embryonic lethal genes, and stronger for genes with post-embryonic phenotypes. In addition, the interference effect generated by feeding can be titrated to uncover a series of hypomorphic phenotypes informative about the functions of a given gene. Using this method, we screened 86 random genes on consecutive cosmids and identified functions for 13 new genes. These included two genes producing an uncoordinated phenotype (a previously uncharacterized POU homeodomain gene, ceh-6, and a gene encoding a MADS-box protein) and one gene encoding a novel protein that results in a high-incidence-of-males phenotype. CONCLUSIONS: RNAi by feeding can provide significant information about the functions of an individual gene beyond that provided by injection. Moreover, it can be used for special applications for which injection or the use of mutants is sometimes impracticable (for example, titration, biochemistry and large-scale screening). Refractory Coats’ Disease of Adult Onset
Purpose: We present the case of an 18-year-old Caucasian male with a unilateral macular star and retinal vascular anomalies compatible with adult onset Coats’ disease. Methods: Diagnosis was based on fundoscopic, fluorescein angiography and optical coherence tomography findings. Results: The patient presented to our emergency department with complaints of low vision in his left eye (LE) detected 10 days before. The best-corrected visual acuity in the LE was 20/50. Fundoscopy of the LE evidenced a complete macular star. Optical coherence tomography showed increased retinal thickness, infiltration of the retinal wall, and detachment of the neuroepithelium. Angiography revealed no appreciable diffusion in the macula. Above the superior temporal (ST) arcade, anomalies in the retinal vasculature were found, with interruption of the peripheral vessels and vessels which were ‘sausage’-like. After 1 month, the LE vision evolved to hand movements. Laser photocoagulation was performed in the ST quadrant. Intravitreal injection of bevacizumab 1.25 mg/0.05 ml and photodynamic therapy were performed without any significant changes, progression of ST serous detachment of the neuroepithelium, and finally progression to macular fibrosis. Discussion: Coats’ disease is usually diagnosed in childhood, but rare cases may occur in adults. Those cases usually have a more indolent course which was not observed in our patient. When there is macular involvement, prognosis is more guarded, despite treatment
Facile Synthesis of High Quality Graphene Nanoribbons
Graphene nanoribbons have attracted attention for their novel electronic and
spin transport properties1-6, and because nanoribbons less than 10 nm wide have
a band gap that can be used to make field effect transistors. However,
producing nanoribbons of very high quality, or in high volumes, remains a
challenge. Here, we show that pristine few-layer nanoribbons can be produced by
unzipping mildly gas-phase oxidized multiwalled carbon nanotube using
mechanical sonication in an organic solvent. The nanoribbons exhibit very high
quality, with smooth edges (as seen by high-resolution transmission electron
microscopy), low ratios of disorder to graphitic Raman bands, and the highest
electrical conductance and mobility reported to date (up to 5e2/h and 1500
cm2/Vs for ribbons 10-20 nm in width). Further, at low temperature, the
nanoribbons exhibit phase coherent transport and Fabry-Perot interference,
suggesting minimal defects and edge roughness. The yield of nanoribbons was ~2%
of the starting raw nanotube soot material, which was significantly higher than
previous methods capable of producing high quality narrow nanoribbons1. The
relatively high yield synthesis of pristine graphene nanoribbons will make
these materials easily accessible for a wide range of fundamental and practical

Research highlights from the 2018 European Respiratory Society International Congress: airway disease.
The annual European Respiratory Society (ERS) International Congress (held in Paris in 2018) was once again a platform for discussion of the highest-quality scientific research, cutting-edge techniques and innovative new therapies within the respiratory field. This article discusses only some of the high-quality research studies presented at this year's Congress, with a particular focus on airway diseases including asthma, chronic obstructive pulmonary disease (COPD), bronchiectasis and cough, as presented through Assembly 5 of the ERS (Airway Diseases: Asthma and COPD). The authors establish the key take-home messages of these studies, compare their findings and place them in the context of current understanding

Este documento de consenso fornece recomendações para a atualização e uniformização dos códigos empregues na RMC. São igualmente feitas recomendações quanto às técnicas e códigos a utilizar nas indicações clínicas mais frequentes
Lepton Number Violation from Colored States at the LHC
The possibility to search for lepton number violating signals at the Large
Hadron Collider (LHC) in the colored seesaw scenario is investigated. In this
context the fields that generate neutrino masses at the one-loop level are
scalar and Majorana fermionic color-octets of SU(3). Due to the QCD strong
interaction these states may be produced at the LHC with a favorable rate. We
study the production mechanisms and decays relevant to search for lepton number
violation signals in the channels with same-sign dileptons. In the simplest
case when the two fermionic color-octets are degenerate in mass, one could use
their decays to distinguish between the neutrino spectra. We find that for
fermionic octets with mass up to about 1 TeV the number of same-sign dilepton
events is larger than the standard model background indicating a promising
signal for new physics.Comment: minor corrections, added reference

In this consensus document, recommendations are made for updating and standardizing CMR codes in Portugal. Guidance on which techniques and codes should be used in the most common clinical scenarios is also provided
