    MDCK cells express serotonin-regulable 11β-hydroxysteroid dehydrogenase type 2

    Prior to this work, we found that adrenal as well as extra-adrenal factors activate the response of renal l 11β-hydroxysteroid dehydrogenase 2 to stressful situations. These results -showing ways through which the organism hinders the pathological occupation of mineralocorticoid receptors by glucocorticoids leading to sodium retention and hypertension- prompted the present study on the nature of the above-mentioned extra-adrenal factors. Serotonin was chosen because of its properties as a widely distributed neurohormone, known to interact with glucocorticoids at many sites, also exhibiting increased levels and effects under stressful situations. We studied serotonin effects on 11β-hydroxysteroid dehydrogenase 2 activity in a cell line derived from distal nephron polarized-epithelium, employing 3H-corticosterone as substrate. The end-product, 3H-11- dehydrocorticosterone was separated from the substrate by HPLC and quantified. Serotonin stimulated 11β-hydroxysteroid dehydrogenase 2 activity only at 2nM and 25pM, the magnitude of the response depending also on substrate concentration. The stimulation was blocked by the specific inhibitors methiothepin and ketanserin. We postulate that the organism partially prevents renal mineralocorticoid receptor occupancy by glucocorticoids, circulating at enhanced levels under stressful situations, through serotonin-mediated catabolic regulation of the 11β-hydroxysteroid dehydrogenase 2 activity.     Effects of benzene and several pharmaceuticals on the growth and microcystin production in Microcystis ruginosa PCC 7806

    Currently, the presence of several pharmaceuticals and other organic compounds called ‘emerging’ contaminants has increased in water bodies. These compounds do not need to persist in the environment to cause negative effects because they are continuously introduced. Hence, assessing the effects of these compounds on aquatic ecosystems is essential. Therefore, the aim of the present study was to analyse the effects of several emerging contaminants (ibuprofen, atenolol, diclofenac and paracetamol) and benzene on Microcystis aeruginosa PCC 7806 growth and toxicity. For this purpose, the growth and intracellular microcystin concentration in M. aeruginosa PCC 7806 were measured in presence of the four emerging contaminants and benzene. The growth rate was estimated by chlorophyll a concentration, and no relevant changes were found. Changes in the expression of the operon mcy were examined by semi-quantitative RT-PCR. Intracellular microcystin production was determined using a MicroCystest¿R kit, and no changes were observed compared to control cells. Thus, the tested concentrations of the contaminants analysed in this study do not have a significant effect on microcystin production in this strain under laboratory conditions

    Development and validation of a computational model for steak double-sided pan cooking

    The objective of this study was to develop and validate a numerical model to adequately simulate the double-sided pan cooking of beef in a domestic environment. The proposed model takes into account the heat flow from the pan to the meat and the moisture transfer, simultaneously with the meat deformation. The model considers the swelling pressure gradient caused by the shrinkage of the meat fibers and connective tissue due to the denaturation of proteins and the loss of the water holding capacity during cooking. The model results were successfully verified with experimental data of the central temperature and weight loss recorded during cooking for three degrees of doneness. The measured experimental temperatures at the center of the meat were 30 ± 3 °C (very rare), 44 ± 3 °C (rare) and 57 ± 2 °C (done) for a 19 mm steak thickness. Meanwhile, their water losses were 4 ± 2 %, 8 ± 1 % and 11 ± 2 %, respectively. The root mean squared errors of the model predictions were 2.16 °C (very rare), 3.56 °C (rare) and 4.57 °C (done) for the central temperature and 1.48 %, 2.08 % and 2.40 %, respectively for the water loss. The model also correctly predicts cooking times for steaks of different thicknesses, taking weight loss as a reference to set this time. The proposed model is postulated as a useful cooking assistance tool to estimate the optimal cooking time according to consumer preferences

En Afrique subsaharienne, 17 espèces de tiques ont été reconnues en tant que vecteur de la maladie, dont 12 sont présents en Sénégambie et Mauritanie. Ont été étudiées les cinq plus importantes espèces dans la transmission du virus CCHF au Sénégal : #Amblyomma variegatum, #Hyalomma impeltatum, #Hyalomma marginatum rufipes, #Hyalomma truncatum$, et #Rhipicephalus guilhoni

    Nasal spray formulations based on combined hyalurosomes and glycerosomes loading zingiber officinalis extract as green and natural strategy for the treatment of rhinitis and rhinosinusitis

    A total green nanotechnological nasal spray has been manufactured and proposed as an alternative treatment of rhinitis and rhinosinusitis. It was obtained by combining the strengthening effect of liposomes on barrier function, the hydrating and lubricating properties of sodium hyaluro-nan and the anti-inflammatory and antioxidant activities of the extract of Zingiber officinalis. To this purpose, the extract was loaded in special phospholipid vesicles immobilized with hyaluronic acid (hyalurosomes), which were further enriched with glycerol in the water phase. Liposomes and glycerosomes were prepared as well and used as reference. Vesicles were oligolamellar and multi-compartment, as confirmed by cryogenic transmission electron microscopy (cryo-TEM) observation, small in size (~140 nm) and negatively charged (~-23 mV). Spray characteristics were evaluated by using the Spraytec® and instant images, from which the plume angle was measured. The range of the droplet size distribution and the narrow spray angle obtained suggest a good nebulization and a possible local deposition in the nasal cavity. In vitro studies performed by using human keratinocytes confirmed the high biocompatibility of vesicles and their ability to effectively counteract oxidative damage on cells induced by hydrogen peroxide.     Tunable Lyapunov exponent in inverse magnetic billiards

    The stability properties of the classical trajectories of charged particles are investigated in a two dimensional stadium-shaped inverse magnetic domain, where the magnetic field is zero inside the stadium domain and constant outside. In the case of infinite magnetic field the dynamics of the system is the same as in the Bunimovich billiard, i.e., ergodic and mixing. However, for weaker magnetic fields the phase space becomes mixed and the chaotic part gradually shrinks. The numerical measurements of the Lyapunov exponent (performed with a novel method) and the integrable/chaotic phase space volume ratio show that both quantities can be smoothly tuned by varying the external magnetic field. A possible experimental realization of the arrangement is also discussed.Comment: 4 pages, 6 figure

    Combining support vector machines and segmentation algorithms for efficient anomaly detection: a petroleum industry application

Similarly, when patterns are numerically distant from the rest of sample, anomalies are indicated as outliers. Anomaly detection had recently attracted the attention of the research community for real-world applications. The petroleum industry is one of the application contexts where these problems are present. The correct detection of such types of unusual information empowers the decision maker with the capacity to act on the system in order to correctly avoid, correct, or react to the situations associated with them. In that sense, heavy extraction machines for pumping and generation operations like turbomachines are intensively monitored by hundreds of sensors each that send measurements with a high frequency for damage prevention. For dealing with this and with the lack of labeled data, in this paper we propose a combination of a fast and high quality segmentation algorithm with a one-class support vector machine approach for efficient anomaly detection in turbomachines.     Repurposing Disulfiram as an Antimicrobial Agent in Topical Infections

    Antimicrobial drugs applied topically offer several advantages. However, the widespread use of antibiotics has led to increasing antimicrobial resistance. One interesting approach in the drug discovery process is drug repurposing. Disulfiram, which was originally approved as an anti-alcoholism drug, offers an attractive alternative to treat topical multidrug resistance bacteria in skin human infections. This study aimed to evaluate the biopharmaceutical characteristics of the drug and the effects arising from its topical application in detail. Microdilution susceptibility testing showed antibacterial activity against Gram-positive bacteria Staphylococcus aureus and Streptococcus pyogenes. Dermal absorption revealed no permeation in pig skin. The quantification of the drug retained in pig skin demonstrated concentrations in the stratum corneum and epidermis, enough to treat skin infections. Moreover, in vitro cytotoxicity and micro-array analyses were performed to better understand the mechanism of action and revealed the importance of the drug as a metal ion chelator. Together, our findings suggest that disulfiram has the potential to be repurposed as an effective antibiotic to treat superficial human skin infections
