    Characterization of carbohydrate fractions and fermentation quality in ensiled alfalfa treated with different additives

    This experiment was carried out to evaluate the effects of adding fast-sile (FS), previous fermented juice (PFJ), sucrose (S) or fast-sile + sucrose (FS + S) on the fermentation characteristics and carbohydrates fractions of alfalfa silages by the Cornell net carbohydrates and proteins systems (CNCPS). Silages quality were well preserved determined by pH, lactic acid (LA), acetic acid (AA), propionic acid (PA), butyric acid (BA) and (NH3-N, % of TN). Except for the silage with no addition of (CK), all other silages were well preserved. FS + S addition showed the lowest pH and contents of AA, PA, BA, and the highest contents of LA. The contents of WSC (Water soluble carbohydrate) in all alfalfa silages decreased with the extension of ensiling time, especially in the former 15 days and decreased sharply in the first 2 days. The content of sucrose in all alfalfa silages in the residual mono and disaccharides was highest, and the content of fructose was the least. The contents of all these sugars decreased sharply in the first 2 days. The content of hemicellulose decreased during ensiling, while no obvious change on content of cellulose. The content of ADL (acid detergent lignin) in alfalfa silages increased during ensiling. The content of starch in silages reduced rapidly in the former days, and then had not obvious change.Key words: Carbohydrate fractions, alfalfa silage, additives, water soluble carbohydrate (WSC)

    Effects of pulsed electric fields (PEF) on vitamin C and its antioxidant properties

    In this study, pulsed electric fields (PEF) treatments and their effects on the structure of vitamin C (VIT-C) were estimated by fluorescence and Fourier transform infrared (FT-IR) spectroscopy, the relative content of VIT-C was measured by HPLC and the antioxidant properties of treated VIT-C by DPPH radical scavenging as well as reducing power tests. The fluorescence intensity of treated VIT-C increased slightly compared to the untreated VIT-C. Moreover, the effect of PEF on the structure of VIT-C was observed using the FT-IR spectra. These phenomena indicated that the PEF affected the conformation of VIT-C, which promoted the VIT-C isomer transformed enol-form into keto-form. In addition, the PEF treatments did not suffer the damage to VIT-C and could slow down the oxidation process in involving of experimental conditions by HPLC. The antioxidant properties of the treated VIT-C were enhanced, which was proved by radical scavenging and also the reducing power tests

    Shotgun Lipidomics Identifies a Paired Rule for the Presence of Isomeric Ether Phospholipid Molecular Species

    Ether phospholipids are abundant membrane constituents present in electrically active tissues (e.g., heart and the brain) that play important roles in cellular function. Alterations of ether phospholipid molecular species contents are associated with a number of genetic disorders and human diseases.Herein, the power of shotgun lipidomics, in combination with high mass accuracy/high resolution mass spectrometry, was explored to identify a paired rule for the presence of isomeric ether phospholipid molecular species in cellular lipidomes. The rule predicts that if an ether phospholipid A'-B is present in a lipidome, its isomeric counterpart B'-A is also present (where the ' represents an ether linkage). The biochemical basis of this rule results from the fact that the enzymes which participate in either the sequential oxidation of aliphatic alcohols to fatty acids, or the reduction of long chain fatty acids to aliphatic alcohols (metabolic precursors of ether lipid synthesis), are not entirely selective with respect to acyl chain length or degree of unsaturation. Moreover, the enzymatic selectivity for the incorporation of different aliphatic chains into the obligatory precursor of ether lipids (i.e., 1-O-alkyl-glycero-3-phosphate) is also limited.This intrinsic amplification of the number of lipid molecular species present in biological membranes predicted by this rule and demonstrated in this study greatly expands the number of ether lipid molecular species present in cellular lipidomes. Application of this rule to mass spectrometric analyses provides predictive clues to the presence of specific molecular species and greatly expands the number of identifiable and quantifiable ether lipid species present in biological samples. Through appropriate alterations in the database, use of the paired rule increases the number of identifiable metabolites in metabolic networks, thereby facilitating identification of biomarkers presaging disease states

    Male water striders attract predators to intimidate females into copulation

    Despite recent advances in our understanding of sexual conflict and antagonistic coevolution between sexes, the role of interspecific interactions, such as predation, in these evolutionary processes remains unclear. In this paper, we present a new male mating strategy whereby a male water strider Gerris gracilicornis intimidates a female by directly attracting predators as long as she does not accept the male's coercive copulation attempt. We argue that this male strategy is a counteradaptation to the evolution of the female morphological shield protecting her genitalia from coercive intromission by water strider males. The G. gracilicornis mating system clearly represents an effect expected from models of the coevolutionary arms race between sexes, whereby one sex causes a decrease in the fitness component of the other sex. Moreover, our study demonstrates a crucial role that interspecific interactions such as predation can have in the antagonistic coevolution between sexes

    Estimating genomic breeding values and detecting QTL using univariate and bivariate models

    Background Genomic selection is particularly beneficial for difficult or expensive to measure traits. Since multi-trait selection is an important tool to deal with such cases, an important question is what the added value is of multi-trait genomic selection. Methods The simulated dataset, including a quantitative and binary trait, was analyzed with four univariate and bivariate linear models to predict breeding values for juvenile animals. Two models estimated variance components with REML using a numerator (A), or SNP based relationship matrix (G). Two SNP based Bayesian models included one (BayesA) or two distributions (BayesC) for estimated SNP effects. The bivariate BayesC model sampled QTL probabilities for each SNP conditional on both traits. Genotypes were permuted 2,000 times against phenotypes and pedigree, to obtain significance thresholds for posterior QTL probabilities. Genotypes were permuted rather than phenotypes, to retain relationships between pedigree and phenotypes, such that polygenic effects could still be estimated. Results Correlations between estimated breeding values (EBV) of different SNP based models, for juvenile animals, were greater than 0.93 (0.87) for the quantitative (binary) trait. Estimated genetic correlation was 0.71 (0.66) for model G (A). Accuracies of breeding values of SNP based models were for both traits highest for BayesC and lowest for G. Accuracies of breeding values of bivariate models were up to 0.08 higher than for univariate models. The bivariate BayesC model detected 14 out of 32 QTL for the quantitative trait, and 8 out of 22 for the binary trait. Conclusions Accuracy of EBV clearly improved for both traits using bivariate compared to univariate models. BayesC achieved highest accuracies of EBV and was also one of the methods that found most QTL. Permuting genotypes against phenotypes and pedigree in BayesC provided an effective way to derive significance thresholds for posterior QTL probabilitie

    Male germ cell-specific protein Trs4 binds to multiple proteins

    Temperature-related sequence 4 (Trs4) has been identified as a testis-specific gene with expression sensitive to the abdominal temperature changes induced by artificial cryptorchidism. In murine testes, Trs4 mRNA was detected in round spermatids and its protein was localized mainly in the elongating spermatids as well as in the acrosomes and tails of mature spermatozoa. Using a yeast two-hybrid screening system, we identified Rshl-2, Gstmu1, and Ddc8 as putative binding partners of the Trs4 protein in mouse testes. Their interactions were confirmed by in vivo and in vitro binding assays. Further studies demonstrated that Ddc8, a newly identified gene with unknown functions, displayed a similar expression pattern with Trs4 in mouse testes. In particular, Trs4, Ddc8, and Rshl-2 proteins were co-localized to the tails of mature spermatozoa. These results suggested that Trs4 might be involved in diverse processes of spermiogenesis and/or fertilization through interactions with its multiple binding partners. © 2009 Elsevier Inc.postprin
