Visualization of plasmid delivery to keratinocytes in mouse and human epidermis
The accessibility of skin makes it an ideal target organ for nucleic acid-based therapeutics; however, effective patient-friendly delivery remains a major obstacle to clinical utility. A variety of limited and inefficient methods of delivering nucleic acids to keratinocytes have been demonstrated; further advances will require well-characterized reagents, rapid noninvasive assays of delivery, and well-developed skin model systems. Using intravital fluorescence and bioluminescence imaging and a standard set of reporter plasmids we demonstrate transfection of cells in mouse and human xenograft skin using intradermal injection and two microneedle array delivery systems. Reporter gene expression could be detected in individual keratinocytes, in real-time, in both mouse skin as well as human skin xenografts. These studies revealed that non-invasive intravital imaging can be used as a guide for developing gene delivery tools, establishing a benchmark for comparative testing of nucleic acid skin delivery technologies

A preliminary study: saltiness and sodium content of aqueous extracts from plants and marine animal shells
To develop a salt substitute with low sodium
content, the water-soluble components of seaweed, kelp,
clamshell, oyster shell, semen cassiae, cuttlefish bone, inula
flower, Arabia cowry shell, and sanna leaf were extracted
with water. The aqueous extracts of nine species of plants
and marine animal shells were obtained after drying the
plants and shells at 105 °C until achieving a constant
weight. The hedonic scale test revealed that the clamshell
and cuttlefish bone aqueous extracts tasted distinctly salty.
The result of the degree of difference test showed that the
1 % clamshell extract solution (m/v) and 0.6 % cuttlefish
bone extract solution (m/v) both had equivalent saltiness of
0.6 % NaCl (m/v). In contrast, the sodium content in the
cuttlefish bone extract solution was 27 % less than that in
a NaCl solution of the same degree of saltiness. Therefore,
a novel salt substitute will be developed in future studies in
accordance with the principles of bionics and a deep understanding
of the salty taste interactions among key salty
components in the cuttlefish bone extract
Reducing inequities among adult female migrants at higher risk for drowning in Australia: The value of swimming and water safety programs
Issue addressed Drowning is a global public health challenge with a need to ensure equity to drowning prevention information and interventions. In Australia, people born overseas are identified as being at greater risk of drowning. This paper presents findings from a community-based qualitative evaluation of swimming and water safety (SWS) programs delivered to adults from migrant backgrounds in Sydney, Australia. Methods A qualitative study was conducted in November-December 2019 among 35 female participants of SWS programs targeted to adult migrants. While offered to all SWS program participants, no males took part in the study. Focus groups and interviews were recorded, transcribed and thematically analysed using a deductive approach. The domains of enquiry were guided by the health belief model and the theory of planned behaviour. Results Study participants were >= 25 years, first generation and most had lived in Australia for >= 10 years. Most were nonswimmers and were fearful of water prior to the program. Key themes were: direct SWS program outcomes, health and well-being; enablers and barriers to participation including: motivation, a program coordinator, fear and settlement priorities. Conclusion Findings suggest that in order to increase SWS participation among migrant communities, the broader determinants of health need to be considered. Culturally appropriate strategies are required to enable both men and women equal opportunities to access SWS programs. So what SWS programs provide multiple benefits for adult migrants; however, the impact on reducing inequities is limited, with broader multi-strategic health promotion approaches and policies required for inclusion and sustainability
