    Analysis of β-globin chromatin micro-environment using a novel 3C variant, 4Cv

    Copyright: © 2010 Pink et al.Higher order chromatin folding is critical to a number of developmental processes, including the regulation of gene expression. Recently developed biochemical techniques such as RNA TRAP and chromosome conformation capture (3C) have provided us with the tools to probe chromosomal structures. These techniques have been applied to the β-globin locus, revealing a complex pattern of interactions with regions along the chromosome that the gene resides on. However, biochemical and microscopy data on the nature of β-globin interactions with other chromosomes is contradictory. Therefore we developed a novel 4C variant, Complete-genome 3C by vectorette amplification (4Cv), which allows an unbiased and quantitative method to examine chromosomal structure. We have used 4Cv to study the microenvironment of the β-globin locus in mice and show that a significant proportion of the interactions of β-globin are inter-chromosomal. Furthermore, our data show that in the liver, where the gene is active, β-globin is more likely to interact with other chromosomes, compared to the brain where the gene is silent and is more likely to interact with other regions along the same chromosome.     Attachment and Information Seeking Strategy Preference in Romantic Relationships

    Two studies examined how attachment relates to information seeking strategy preference in established romantic relationships using a hypothetical scenario (Study 1) and an experiment (Study 2). In both studies, we tested hypotheses examining 1) if highly anxious individuals prefer to seek information indirectly (vs. directly) in potentially relationship-threatening situations, and 2) if these individuals tend to associate direct information seeking with negative outcomes. Study 1 revealed that as predicted, highly anxious individuals were more likely to endorse indirect information seeking strategies but less likely to endorse a direct approach. The negative association between attachment anxiety and direct strategy endorsement was fully mediated by expected outcomes. In contrast, in Study 2 highly anxious individuals in the threat condition reported greater desire to directly seek information from their partners. These conflicting results suggest that the conditions influencing highly anxious individuals’ strategy preferences may be quite complex and warrant future research

    Relationships between technical efficiency and the quality and costs of health care in Italy†

    Objectives This paper reports the measurement of technical efficiency of Tuscan Local Health Authorities and its relationship with quality and appropriateness of care. Design First, a bias-corrected measure of technical efficiency was developed using the bootstrap technique applied to data envelopment analysis. Then, correlation analysis was used to investigate the relationships among technical efficiency, quality and appropriateness of care. Setting and Participants These analyses have been applied to the Local Health Authorities of Tuscany Region (Italy), which provide not only hospital inpatient services, but also prevention and primary care. All top managers of Tuscan Local Health Authorities were involved in selection of the inputs and outputs for calculating technical efficiency. Main Outcome Measures The main measures used in this study are volume, quality and appropriateness indicators monitored by the multidimensional performance evaluation system developed in the Tuscany Region. Results On average, Tuscan Local Health Authorities experienced 14(%) of bias-corrected inefficiency in 2007. Correlation analyses showed a significant negative correlation between per capita costs and overall performance. No correlation was found in 2007 between technical efficiency and overall performance or between technical efficiency and per capita costs. Conclusions Technical efficiency cannot be considered as an extensive measure of healthcare performance, but evidence shows that Tuscan Local Health Authorities have room for improvement in productivity levels. Indeed, correlation findings suggest that, to pursue financial sustainability, Local Health Authorities mainly have to improve their performance in terms of quality and appropriateness

    Active Microrheology of Networks Composed of Semiflexible Polymers. II. Theory and comparison with simulations

    Building on the results of our computer simulation (ArXiv cond-mat/0503573)we develop a theoretical description of the motion of a bead, embedded in a network of semiflexible polymers, and responding to an applied force. The theory reveals the existence of an osmotic restoring force, generated by the piling up of filaments in front of the moving bead and first deduced through computer simulations. The theory predicts that the bead displacement scales like x ~ t^alfa with time, with alfa=0.5 in an intermediate- and alfa=1 in a long-time regime. It also predicts that the compliance varies with concentration like c^(-4/3) in agreement with experiment.Comment: 18 pages and 2 figure
