    Moho topography beneath the Corinth Rift area (Greece) from inversion of gravity data

    International audienceOur aim is to understand better the rifting process by imaging the Moho depth variation beneath Corinth and Evvia. We present here the results of a gravity inversion analysis in the region of the Corinth and Evvia rift system, and compare them to those obtained independently from teleseismic tomography and receiver function analyses. The results of these different studies appear to be consistent and show (1) a 10 km crustal thickening in the western part of the area beneath the Hellenides mountains, (2) NW–SE-trending periodic crustal thinning, and (3) a maximum crustal thinning north of the Gulf of Corinth. This 4 km thinning is unlikely to be the result of the rifting alone, which seems to have been reactivated since only 1 Ma. We propose here a geodynamical scenario in two major steps to explain the evolution of Corinth area. Aegean Miocene extension involving boudinage resulted in periodic crustal thinning, consistent with observations. These lithospheric instabilities could have favoured rupture initiation in particular areas, especially near the city of Corinth. Then, the reactivation of the Corinth Rift extension, 1 Myr ago, led to westward rift propagation. The offset observed between the maximum crustal thinning and the Gulf of Corinth could be accommodated by a low-angle normal fault at about 10–15 km depth. The Corinth Rift is thus asymmetrical and was initiated in places of crustal weakness due to Miocene lithospheric instabilities

    Joint inversion of teleseismic and GOCE gravity data: application to the Himalayas

    Our knowledge and understanding of the 3-D lithospheric structure of the Himalayas and the Tibetan Plateau is still challenging although numerous geophysical studies have been performed in the region. The GOCE satellite mission has the ambitious goal of mapping Earth's gravity field with unprecedented precision (i.e. an accuracy of 1-2 mGal for a spatial resolution of 100 km) to observe the lithosphere and upper-mantle structure. Consequently, it gives new insights in the lithospheric structure beneath the Himalayas and the Tibetan Plateau. Indeed, the GOCE gravity data now allow us to develop a new strategy for joint gravimetry-seismology inversion. Combined with teleseismic data over a large region in a joint inversion scheme, they will lead to lithospheric velocity-density models constrained in two complementary ways. We apply this joint inversion scheme to the Hi-CLIMB (Himalayan-Tibetan Continental Lithosphere during Mountain Building) seismological network which was deployed in South Tibet and the Himalayas for a 3-yr period. The large size of the network, the high quality of the seismological data and the new GOCE gravity data set allow us to image the entire lithosphere of this active area in an innovative way. We image 3-D low velocity and density structures in the middle crust that fit the location of discontinuous low S-velocity zones revealed by receiver functions in previous geophysical studies. In the deeper parts of our velocity model we image a positive anomaly interpreted to be the heterogenous Indian lithosphere vertically descending beneath the centre of the Tibetan Platea

    Reduction of atherosclerotic lesions in rabbits treated with etoposide associated with cholesterol-rich nanoemulsions

Association of paclitaxel with LDE markedly reduces the lesions. In previous studies, treatment of refractory cancer patients with etoposide associated with LDE had been shown devoid of toxicity. In this study, the ability of etoposide to reduce lesions and inflammatory factors in atherosclerotic rabbits was investigated.Methods: Eighteen New Zealand rabbits were fed a 1% cholesterol diet for 60 days. Starting from day 30, nine animals were treated with four weekly intravenous injections of etoposide oleate (6 mg/kg) associated with LDE, and nine control animals were treated with saline solution injections.Results: LDE-etoposide reduced the lesion areas of cholesterol-fed animals by 85% and intima width by 50% and impaired macrophage and smooth muscle cell invasion of the intima. Treatment also markedly reduced the protein expression of lipoprotein receptors (LDL receptor, LDL-related protein-1, cluster of differentiation 36, and scavenger receptor class B member 1), inflammatory cytokines (interleukin-1β and tumor necrosis factor-α), matrix metallopeptidase-9, and cell proliferation markers (topoisomerase IIα and tubulin).Conclusion: The ability of LDE-etoposide to strongly reduce the lesion area and the inflammatory process warrants the great therapeutic potential of this novel preparation to target the inflammatory-proliferative basic mechanisms of the disease.Keywords: atherosclerosis treatment, drug delivery, LDL-receptor

    Deep structure of the Baikal rift zone revealed by joint inversion of gravity and seismology

    International audience[1] The question of plate boundary forces and deep versus shallow asthenospheric uplift has long been debated in intracontinental rift areas, particularly in the Baikal rift zone, Asia, which is colder than other continental rifts. As previous gravity and teleseismic studies support the dominance of opposing mechanisms in the Baikal rift, we reconsidered both data sets and jointly inverted them. This more effective approach brings insight into location of the perturbing bodies related to the extension in this region. Our new joint inversion method allows for inverting the velocity-density relationship with independent model parametrization. We obtain velocity and density models that consistently show (1) crustal heterogeneities that coincide with the main tectonic features at the surface, (2) a faster and denser cratonic mantle NW of Lake Baikal that we relate to the thermal contrast between old and depleted Archean (Siberian platform) and Paleozoic orogenic belt (Sayan-Baikal belt), (3) three-dimensional topographic variations of the crust-mantle boundary with well-located upwarpings, and (4) the lithosphere-asthenosphere boundary uplift up to 70 km depth with a NW dip. Our resulting velocity and density models support the idea of a combined influence of lithospheric extension and inherited lithospheric heterogeneities for the origin of the Baikal rift zone.     Simultaneous transfer of cholesterol, triglycerides, and phospholipids to high-density lipoprotein in aging subjects with or without coronary artery disease

    OBJECTIVE: To verify whether the capacity of high-density lipoprotein (HDL) to simultaneously receive nonesterified cholesterol, triglycerides, cholesteryl esters, and phospholipids changes with aging and the presence of coronary artery disease. DESIGN: Cross-sectional study with biochemical analyses. SUBJECTS: Eleven elderly patients with coronary artery disease (74±5 years) were compared with the following groups of non-coronary artery disease subjects (referred to as "healthy"): 25 young (25±5 years), 25 middle-aged (42± years), and 25 elderly subjects (75±8 years). METHODS: Plasma samples were incubated with a nanoemulsion labeled with radioactive lipids; the transfer of the lipids from the nanoemulsion to the HDL was measured in chemically precipitated HDL. HDL size and paraoxonase-1 activity were also determined. RESULTS: The transfer of cholesteryl esters and phospholipids to high-density lipoprotein was significantly greater (p<0.001) in healthy elderly subjects than in the middle-aged and younger subjects. Non-esterified cholesterol and triglyceride transfer was not different among these three groups. The HDL size was significantly greater (p<0.001) in healthy elderly subjects than in the middle-aged and younger subjects. The paraoxonase-1 activity was similar among the groups. Compared with healthy elderly subjects, coronary artery disease elderly subjects had significantly less (p<0.05) transfer of non-esterified cholesterol, triglycerides, and cholesteryl esters to the HDL and a significantly smaller (p<0.05) HDL size. CONCLUSION: Because lipid transfer is enhanced in healthy elderly subjects but not in those with coronary artery disease, increasing lipid transfer to HDL may be a protective mechanism against the disease

    Do health behaviours change after colonoscopy? A prospective cohort study on diet, alcohol, physical activity and smoking among patients and their partners

    Objectives: To describe diet, alcohol, physical activity and tobacco use prospectively, that is, before and 10 months after colonoscopy for patients and their partners. Design: Prospective cohort study of health behaviour change in patients and partners. Comparison groups are patients receiving a normal result notification (NRN) versus patients receiving an abnormal result notification (ARN). Patients and partners (controls) are also compared. Setting: 5 Scottish hospitals. Participants: Of 5798 colonoscopy registrations, 2577 (44%) patients met the eligibility criteria of whom 565 (22%) were recruited; 460 partners were also recruited. Measures: International Physical Activity Questionnaire, Scottish Collaborative Group Food Frequency Questionnaire (includes alcohol), smoking status, sociodemographic characteristics, body mass index, medical conditions, colonoscopy result, Multidimensional Health Locus of Control Scale, behaviour-specific self-efficacy scales. Results: 57% of patients were men, with a mean age of 60.8 years (SE 0.5) and 43% were from more affluent areas. 72% (n=387) of patients received an ARN and 28% (n=149) received an NRN. Response rate of the second questionnaire was 68.9%. Overall, 27% of patients consumed < 5 measures of fruit and vegetables/day, 20% exceeded alcohol limits, 50% had low levels of physical activity and 21% were obese. At 10-month follow-up, a 5% reduction in excessive alcohol consumption and an 8% increase in low levels of physical activity were observed among patients; no significant changes occurred in partners. Baseline high alcohol consumption and low physical activity were the strongest predictors of these behaviours at follow-up. Low alcohol self-efficacy and increasing age were associated with poorer health-related behaviours at follow-up for alcohol consumption and physical activity, respectively. Conclusions: Colonoscopy is associated with marginal beneficial changes in some behaviours but not others. Further work is needed to explore how services can optimise increases in beneficial behaviours and mitigate increases in harmful ones

    Paternal UPD15: Further Genetic and Clinical Studies in Four Angelman Syndrome Patients

    Among 25 patients diagnosed with Angelman syndrome, we detected 21 with deletion and 4 with paternal uniparental disomy (UPD), 2 isodisomies originating by postzygotic error, and 1 MII nondisjunction event. The diagnosis was obtained by molecular techniques, including methylation pattern analysis of exon 1 of SNRPN and microsatellite analysis of loci within and outside the 15q11-q13 region. Most manifestations present in deletion patients are those previously reported. Comparing the clinical data from our and published UPD patients with those with deletions we observed the following: the age of diagnosis is higher in UPD group (average 7 3 ⁄12 years), microcephaly is more frequent among deletion patients, UPD children start walking earlier (average age 2 9 ⁄12 years), whereas in deletion patients the average is 4 1 ⁄2 years, epilepsy started later in UPD patients (average 5 10 ⁄12 years) than in deletion patients (average 1 11 ⁄12 years), weight above the 75th centile is reported mainly in UPD patients, complete absence of speech is more common in the deleted (88.9%) than in the UPD patients because half of the children are able to say few words. Thus, besides the abnormalities already described, the UPD patients have somewhat better verbal development, a weight above the 75th centile, and OFC in the upper normal range. Am. J. Med.
