    Accumulation of Flavonols over Hydroxycinnamic Acids Favors Oxidative Damage Protection under Abiotic Stress

    Efficient detoxification of reactive oxygen species (ROS) is thought to play a key role in enhancing the tolerance of plants to abiotic stresses. Although multiple pathways, enzymes, and antioxidants are present in plants, their exact roles during different stress responses remain unclear. Here, we report on the characterization of the different antioxidant mechanisms of tomato plants subjected to heat stress, salinity stress, or a combination of both stresses. All the treatments applied induced an increase of oxidative stress, with the salinity treatment being the most aggressive, resulting in plants with the lowest biomass, and the highest levels of H2O2 accumulation, lipid peroxidation, and protein oxidation. However, the results obtained from the transcript expression study and enzymatic activities related to the ascorbate-glutathione pathway did not fully explain the differences in the oxidative damage observed between salinity and the combination of salinity and heat. An exhaustive metabolomics study revealed the differential accumulation of phenolic compounds depending on the type of abiotic stress applied. An analysis at gene and enzyme levels of the phenylpropanoid metabolism concluded that under conditions where flavonols accumulated to a greater degree as compared to hydroxycinnamic acids, the oxidative damage was lower, highlighting the importance of flavonols as powerful antioxidants, and their role in abiotic stress tolerance.This work was supported by the Ministry of Economy and Competitiveness from Spain (GrantNo. AGL2015-66033-R), and Seneca Foundation from Region of Murcia, Spain (Grant no.15288/ PI/10).Peer reviewedPeer Reviewe

    Activation of innate and specific immune responses in hemolytic uremic syndrome (HUS)- patients

    La función primaria del sistema inmunológico es preservar al individuo sano frente a infinidad de agentes microbianos patógenos o injuriantes. Sin embargo, en determinadas circunstancias los mecanismos agresores normalmente montados contra un agente invasor, pueden tornarse altamente injuriantes para el propio individuo. Hay importantes evidencias tanto clínicas como experimentales, de que la reacción inflamatoria inducida por los distintos componentes de las bacterias Escherichia coli productoras de toxina Shiga (Stx) (STEC), fundamentalmente la Stx y los lipopolisacáridos (LPS), contribuye decisivamente en la evolución a la forma completa de SUH Así los pacientes al ser diagnosticados de SUH, presentan evidencias de haber sufrido un proceso de activación del sistema inmune innato, o reacción inflamatoria muy aguda y temprana en la evolución de la enfermedad. Algunas de estas evidencias pueden resumirse como: una neutrofilia marcada, leucocitos neutrófilos (PMN) circulantes que se encuentran “agotados o exhaustos”, los monocitos diferenciados hacia un fenotipo inflamatorio (menor expresión de CD14 y aumento de CD16), y se encuentra un significativo descenso en los leucocitos que presentan el receptor para la quimioquina Fractalquina (FKN, CX3CL1)) (CX3CR1): los. monocitos clásicos y células Natural Killer (NK). Estas células tienen un alto potencial citotóxico. La FKN se expresa en endotelio y epitelio renal y ha sido involucrada en los mecanismos patogénicos en distintas nefropatías. Llamativamente, encontramos una correlación significativa entre la severidad del cuadro renal y las alteraciones mencionadas. Por último se discute el papel protector que la respuesta inmune específica podría ejercer, fundamentalmente a través de la producción de anticuerpos neutralizantes de la Stx.The central role of the immune system is the preservation of the health against several pathogenic microbes and injury agents. However, on special conditions defensive mechanisms triggered towards the foreign agent can damage the host. Clinical and experimental evidence indicate that inflammatory reaction triggered by the main components of Shiga toxin (Stx)- producing Escherichia coli (STEC), participate in the evolution to the complete form of HUS. When children are diagnosed of HUS, they present evidence that have suffered a very strong and early inflammatory response. These features include: the presence of a marked neutrophilia, the polymorfonuclear leucocytes (PMN) are “deactivated or exhausted” and the monocytes are differentiated towards an inflammatory phenotype (CD14-reduced and CD16-enhanced membrane expression). In addition, HUS-patients show a marked reduction in the absolute and relative number of leucocytes carrying the receptor (CX3CR1) for the chemokine “Fractalkine” (FKN, CX3 CL1), which are the classic monocytes and Natural Killer cells (NK). All these cells express a high cytotoxic potencial. The chemokine FKN is expressed in endothelial and epithelial renal cells, and is involved in the pathogenic mechanism of different nephropathies. Noteworthy, we found a significant correlation between the severity of the renal damage (as days of anuria) and the alterations described above. Finally, the protective role of specific immune response, mainly through the antibody production with Stx-neutralizing capacity, is discussed.Fil: Palermo, Marina Sandra.     Estudo clínico duplo cego comparando praziquantel com oxamniquine

    Com objetivo de se compararem a tolerabilidade e eficácia do praziquantel e oxamniquine, procedeu-se a um estudo prospectivo duplo-cego envolvendo 120 pacientes com esquistossomose intestinal ou hepatintestinal. Os pacientes foram randomizados em dois grupos. Um foi tratado com praziquantel, na dose de 55 mg/kg de peso, o outro com oxamniquine, 15 mg/kg de peso, sempre administrados em dose única por via oral. O diagnóstico e seguimento parasitológicos basearam-se ho exame de fazes peio método de Kate Katz. Em 73 de 77 casos negativos após tratamento, executaram-se biópsias retais. Efeitos colaterais, principalmente tontura, sonolência, dores abdominais, cefaléia, náuseas e diarréia foram observados em 87% dos casos. Sua incidência, intensidade e duração foram semelhantes em ambos os grupos, mas a dor abdominal foi significativamente mais freqüente após praziquantel, havendo maior tendência para tontura intensa após oxamniquine. Observou-se aumento significante de alamina-aminotransferase e gama-glutamiltransferase após oxamniquine e de bilirrubina total após praziquantel. Um total de 48 pacientes tratados com praziquantel e 46 com oxamniquine completaram os exames de controle até o sexto mês. As percentages de cura foram de 79,2% e de 84,8% respectivamente, diferença não significativa. Os pacientes não curados mostraram redução média do número de ovos de 93,5% e de 84,1%, diferença não significativa. Cinco pacientes retratados com praziquantel curaram-se, mas somente um de três retratados com oxamniquine. Estes resultados mostram, que ambas as drogas-apesar de diferentes propriedades farmacológicas provocam reações colaterais semelhantes e apresentam eficácia terapêutica comparável.A double-blind clinical trial involving 120 patients with chronic schistosomiasis was carried out to compare the tolerability and efficacy of praziquantel and oxamniquine. The patients were randomly allocated into two groups. One was treated with praziquantel, 55 mg/kg of body weight CBWT), and the other one with oxamniquine, 15mg/kg bwt, administered in a single oral dose. The diagnosis and the parasitological follow-up was based on stool examinations by quantitative Kato-Katz method and on rectal biopsies. Side-effects mainly dizziness, sleepness, abdominal distress, headache, nausea and diarrhea were observed in 87% of the cases. Their incidence, intensity and duration were similar for both drugs but abdominal pain was significantly more frequent after praziquantel intake and severe dizziness was more commonly reported after oxamniquine. A significant increase of alanine-aminotransferase and y-glutamyltransferase was found with the latter drug and of total bilirubin with the former one. A total of 48 patients treated with praziquantel and 46 with oxamniquine completed with negative findings the required three post-treatment parasitological controls three slides of each stool sample on the first, third and sixth month. The achieved cure rates were 79.2% and 84.8%, respectively, a difference without statistical significance. The non-cured cases showed a mean reduction in the number of eggs per gram of feces of 93.5% after praziquantel and of 84.1% after oxamniquine. This diference also was not significant. Five patients retreated with praziquantel were cured but only one out of three treated a second time with oxamniquine. These findings show that both drugs despite their different chemical structures, pharmacological properties and mechanisms-of-action induce similar side-effects as well as a comparable therapeutical efficacy, in agreement with the results reported from analogous investigations

    Increasing the Rate of Magnesium Intercalation Underneath Epitaxial Graphene on 6H-SiC(0001)

    Magnesium intercalated 'quasi-freestanding' bilayer graphene on 6H-SiC(0001) (Mg-QFSBLG) has many favorable properties (e.g., highly n-type doped, relatively stable in ambient conditions). However, intercalation of Mg underneath monolayer graphene is challenging, requiring multiple intercalation steps. Here, we overcome these challenges and subsequently increase the rate of Mg intercalation by laser patterning (ablating) the graphene to form micron-sized discontinuities. We then use low energy electron diffraction to verify Mg-intercalation and conversion to Mg-QFSBLG, and X-ray photoelectron spectroscopy to determine the Mg intercalation rate for patterned and non-patterned samples. By modeling Mg intercalation with the Verhulst equation, we find that the intercalation rate increase for the patterned sample is 4.5±\pm1.7. Since the edge length of the patterned sample is \approx5.2 times that of the non-patterned sample, the model implies that the increased intercalation rate is proportional to the increase in edge length. Moreover, Mg intercalation likely begins at graphene discontinuities in pristine samples (not step edges or flat terraces), where the 2D-like crystal growth of Mg-silicide proceeds. Our laser patterning technique may enable the rapid intercalation of other atomic or molecular species, thereby expanding upon the library of intercalants used to modify the characteristics of graphene, or other 2D materials and heterostructures.Comment: 24 pages, 4 figure

    The influence of olive tree fertilization on the phenols in virgin olive oils. A review

    The total phenols in virgin olive oil are highly dependent on cultivar, but also on ripening stage and other agronomic factors. The focus of most studies on agronomic factors has been irrigation, while fertilization has received less attention. Most of the fertilization works find that nitrogen over-fertilization leads to a decrease in phenol contents in virgin olive oil (VOO) and extra virgin olive oil (EVOO), under rain-fed or irrigation management. Ortho-diphenols also decrease with high doses of nitrogen, with no effect on secoiridoids. Phosphorous has a minor effect on irrigated trees; while the role of potassium is controversial, with a lack of trials with calcium and micro-nutrients. Due to the great impact of the fertilization on the phenol content and quality of VOO, new research is necessary with focus aimed at different cultivars and agronomic factors.info:eu-repo/semantics/publishedVersio
