    Developing an Unnatural Amino Acid-Specific Aminoacyl tRNA Synthetase

    Unnatural Amino Acids (UAAs), amino acids not present in the human genetic code, have been synthesized to have a broad range of useful properties, in this case, as metal-binders which could have drug delivery applications. In order for the cell to place a UAA into the protein, two components, a unique aminoacyl tRNA synthetase and a corresponding tRNA must be present. If an amino acid is successfully charged to the tRNA, a stop codon is suppressed and a functional protein is built with the UAA at the mutation site. Such a tRNA molecule has previously been developed, as well as many synthetases specific to UAAs. In this work, the range of UAAs which can be incorporated into proteins using the E. coli’s own machinery is expanded by the development of a novel aminoacyl tRNA synthetase. By making a library of synthetase-coding plasmid variants and performing positive and negative screenings, the binding pocket of the synthetase can be modified for specificity to a UAA while not allowing the tRNA to be charged with a natural amino acid. In this work, we are attempting to evolve new tRNA synthetases for the incorporation of metal-binding amino acids by developing the plasmid library and a screening system to find synthetase variants meeting these criteria

    Although female magicians have existed since the rise of entertainment magic, women have faced difficulty in entering the “fraternity” of the magic community. As an art form largely based around persuasion, it is useful to study the performance of magic as a text. It is additionally useful to study female magicians within this context of rhetoric. Not only will examining the rhetoric of female magicians provide insights on the rhetoric of women in this unique arena, but also of women in a historically gendered and underrepresented field. Research into this area may disclose other details regarding the communicative differences between women and men and how communication is adapted within a gendered communication paradigm

    Exploring the Abilities of 3D Printing and its Viability for Consumption in the Fashion Industry

    Abstract With the ever-evolving state of today’s technology, designers and retailers in the apparel industry are seeking out new technological methods that have the capacity to revolutionize and individualize their brand, as well as meet consumer needs and preferences. An emerging technology is 3D printing, which utilizes computer-aided technology and a variety of filaments to construct an object. Though 3D printing technology offers the ability for rapid prototyping, a condensed supply chain, and a sustainable additive manufacturing process, there is question as to whether or not consumers are ready for 3D printed clothing to enter their wardrobes. In this creative study, the authors designed a 3D printed garment in order to test whether 3D printers could be used to make wearable clothing of similar characteristics to clothing typically made of fabric. A survey was then conducted on the University of Arkansas campus to measure consumer response to the project garment. Three primary factors were measured: prior exposure and interest in 3D printing, general fashion interest, and aesthetic appeal of the project 3D printed garment. Overall perceptions of the project garment as well as further use of 3D printing for the apparel industry were positive. The ability of this study to create a fully 3D printed garment as well as understand consumer response to 3D printed clothing provides insight into this emerging technology. The results warrant further research into its capabilities for fashion and that the fashion industry could move towards adopting this technology on a wider scale in coming years. The results indicate that a major transformation in ready-to-wear style is feasible and beneficial to the apparel industry because of 3D printing.     Ultrasonic wave propagation in heterogenous media

    The non-destructive testing of austenitic steel welds using ultrasound is of vital importance for assessing safety critical structures such as those found in the nuclear industry. The internal geometry of these welds is heterogeneous and highly scattering and this makes it dicult to detect and characterise any defects within them. To help overcome these diculties the use of ultrasound transducer arrays and the associated Full Matrix Capture is becoming more widespread. There is a need therefore to develop post-processing algorithms that best utilise the data from such devices. This paper considers the use of a time-frequency domain method known as the Decomposition of the Time Reversal Operator (DORT) method. To develop this method and to demonstrate its ecacy in tackling this problem a series of simulated data sets are used. The simulated data is generated using a finite element method (PZFLEX) with the heterogeneous internal microstructure of the weld being given by previous Electron Backscatter Diraction measurements. A range of artificial flaws are then inserted into this geometry. By varying the flaw size and type a comparison is conducted between the DORT method and the Total Focusing Method (TFM) and their relative ability to perform flaw detection assessed. Importantly, however, the DORT method relies on a Singular Value Decomposition in time and frequency space and this spectral information contains information about the flaw size and shape
