Estimating the demand for union-led learning in Scotland
The evidence contained here is the first comprehensive and formal statistical analysis of demand for union-led learning in Scotland or the UK. A high degree of consistency of outcome resulted from each of the data sources used. This provides reliable evidence that there is considerable current demand and latent demand for union-led learning in Scotland, and that increased union activity in this area is likely to further stimulate demand. Any expansion of union-led learning would, of course, place additional learning demands on ULRs and would highlight further the need for them to be appropriately supported by unions and employers
Walking and chewing gum at the same time: Australia’s free trade area strategy
The pursuit of free trade area agreements (FTAs), according to some, does not impede the multilateral trade negotiations process. It is argued to the contrary in the present paper that the FTA approach does impede the multilateral one. Comments are offered on common approaches to the analysis of the impact of FTAs, on the reasons why, despite concerns evident in these comments, the FTA strategy has become so popular, and on ways out of a dilemma of FTA proliferation, or at least ways to avoid the risks associated with it.International Relations/Trade,
Institutionalizing health impact assessment in London as a public health tool for increasing synergy between policies in other areas
Objectives: To describe the background to the inclusion of health impact assessment (HIA) in the development process for the London mayoral strategies, the HIA processes developed, how these evolved, and the role of HIA in identifying synergies between and conflicting priorities of different strategies.Study design: Case series.Methods: Early HIAs had just a few weeks for the whole HIA process. A rapid appraisal approach was developed. Stages included: scoping, reviewing published evidence, a stakeholder workshop, drafting a report, review of the report by the London Health Commission, and submission of the final report to the Mayor. The process evolved as more assessments were conducted. More recently, an integrated impact assessment (IIA) method has been developed that fuses the key aspects of this HIA method with sustainability assessment, strategic environmental assessment and equalities assessment.Results: Whilst some of the early strategy drafts encompassed some elements of health, health was not a priority. Conducting HIAs was important both to ensure that the strategies reflected health concerns and to raise awareness about health and its determinants within the Greater London Authority (GLA). HIA recommendations were useful for identifying synergies and conflicts between strategies. HIA can be successfully integrated into other impact assessment processes.Conclusions: The HIAs ensured that health became more integral to the strategies and increased understanding of determinants of health and how the GLA impacts on health and health inequalities. Inclusion of HIA within IIA ensures that health and health inequalities impacts are considered robustly within statutory impact assessments. (C) 2010 The Royal Society for Public Health. DevA, a GntR-like transcriptional regulator required for development in streptomyces coelicolor
The gram-positive filamentous bacterium Streptomyces coelicolor has a complex developmental cycle with three distinct phases: growth of the substrate mycelium, development of reproductive structures called aerial hyphae, and differentiation of these aerial filaments into long chains of exospores. During a transposon mutagenesis screen, we identified a novel gene (devA) required for proper development. The devA mutant produced only rare aerial hyphae, and those that were produced developed aberrant spore chains that were much shorter than wild-type chains and had misplaced septa. devA encodes a member of the GntR superfamily, a class of transcriptional regulators that typically respond to metabolite effector molecules. devA forms an operon with the downstream gene devB, which encodes a putative hydrolase that is also required for aerial mycelium formation on R5 medium. S1 nuclease protection analysis showed that transcription from the single devA promoter was temporally associated with vegetative growth, and enhanced green fluorescent protein transcriptional fusions showed that transcription was spatially confined to the substrate hyphae in the wild type. In contrast, devAB transcript levels were dramatically upregulated in a devA mutant and the devA promoter was also active in aerial hyphae and spores in this background, suggesting that DevA might negatively regulate its own production. This suggestion was confirmed by gel mobility shift assays that showed that DevA binds its own promoter region in vitro
The effect of Cichorium intybus and Lotus corniculatus on nematode burdens and production in grazed lambs
This report was presented at the UK Organic Research 2002 Conference. The study was designed to examine the hypothesis that chicory (Cichorium intybus) and Lotus sp. (Lotus corniculatus) have the potential to affect the naturally acquired nematode burden in grazed lambs. Organic male castrate lambs (48) with a naturally acquired parasite burden grazed replicate combination plots (0.6 ha) of chicory, Lotus corniculatus, perennial ryegrass (Lolium perenne) and white clover (Trifolium repens). Lamb performance was determined by weekly weight gain and condition score assessments. Nematode burden was assessed by individual lamb faecal egg count (FEC) before and after drenching (levamisole). The range of parasitic helminths present was assessed by faecal culture and by total worm counts performed on a proportion of the lambs at slaughter. Weekly pasture larval counts (PLCs) were conducted on the trial plots. A concurrent small plot study (6 x 1m2 replicates) of each of the forages used in the grazing trial was run to assess the potential effect of forage type on the development and survival of Teladorsagia circumcincta assessed by weekly PLCs. Preliminary data suggest that lambs grazing chicory or a combination of lotus and chicory had lower FECs than those grazing PRG/WC, however there was no significant difference in the total worm counts

The melanocortin 4 receptor: Oligomer formation, interaction sites and functional significance
This study involves the structural and functional properties of the recombinant melanocortin 4 receptor
(MC4R) expressed in the HEK-293 cell line. Using co-immuno-purification approaches, the receptor appears
to be an oligomer, which can be crosslinked through disulphide bonds involving a native cysteine residue
(84) to give a dimeric species. This position is located near the cytosolic region of transmembrane segment
2 and it is suggested that this is an interacting interface between MC4R monomers. Using co-expression of
the native protein and a C84A mutant, it appears that the receptor also forms higher order oligomers via alternative
interfaces. Interestingly, disulphide crosslink formation does not occur if the receptor is uncoupled
from its G-protein, even though the oligomeric state is preserved. This suggests that the conformational
changes, which occur on activation, affect the TM2 interface. The pharmacology of the agonist, NDP-MSH, indicates
that the MC4R retains high affinity for the ligand in the absence of the G-protein but occupancy for the
ligand is increased. The data can be interpreted to suggest that the G-protein exerts a negative allosteric effect
on the receptor. Co-expression of one receptor lacking the ability to signal with another, which cannot bind
the agonist, restored ligand-dependent activation of the G-protein to situations in which neither receptor on
its own could activate the G-protein. Such transactivation suggests meaningful cross talk between the receptor
subunits in the oligomeric complex. These studies demonstrate further unique features of the MC4R
