    Cell patterning on photolithographically defined parylene-C:SiO2 substrates

    Cell patterning platforms support broad research goals, such as construction of predefined in vitro neuronal networks and the exploration of certain central aspects of cellular physiology. To easily combine cell patterning with Multi-Electrode Arrays (MEAs) and silicon-based ‘lab on a chip’ technologies, a microfabrication-compatible protocol is required. We describe a method that utilizes deposition of the polymer parylene-C on SiO(2 )wafers. Photolithography enables accurate and reliable patterning of parylene-C at micron-level resolution. Subsequent activation by immersion in fetal bovine serum (or another specific activation solution) results in a substrate in which cultured cells adhere to, or are repulsed by, parylene or SiO(2) regions respectively. This technique has allowed patterning of a broad range of cell types (including primary murine hippocampal cells, HEK 293 cell line, human neuron-like teratocarcinoma cell line, primary murine cerebellar granule cells, and primary human glioma-derived stem-like cells). Interestingly, however, the platform is not universal; reflecting the importance of cell-specific adhesion molecules. This cell patterning process is cost effective, reliable, and importantly can be incorporated into standard microfabrication (chip manufacturing) protocols, paving the way for integration of microelectronic technology

    Defining the chromatin signature of inducible genes in T cells

    BACKGROUND Specific chromatin characteristics, especially the modification status of the core histone proteins, are associated with active and inactive genes. There is growing evidence that genes that respond to environmental or developmental signals may possess distinct chromatin marks. Using a T cell model and both genome-wide and gene-focused approaches, we examined the chromatin characteristics of genes that respond to T cell activation. RESULTS To facilitate comparison of genes with similar basal expression levels, we used expression-profiling data to bin genes according to their basal expression levels. We found that inducible genes in the lower basal expression bins, especially rapidly induced primary response genes, were more likely than their non-responsive counterparts to display the histone modifications of active genes, have RNA polymerase II (Pol II) at their promoters and show evidence of ongoing basal elongation. There was little or no evidence for the presence of active chromatin marks in the absence of promoter Pol II on these inducible genes. In addition, we identified a subgroup of genes with active promoter chromatin marks and promoter Pol II but no evidence of elongation. Following T cell activation, we find little evidence for a major shift in the active chromatin signature around inducible gene promoters but many genes recruit more Pol II and show increased evidence of elongation. CONCLUSIONS These results suggest that the majority of inducible genes are primed for activation by having an active chromatin signature and promoter Pol II with or without ongoing elongation

    Geology of part of the central Damara belt around the Tumas River, South West Africa

    The investigation covering 1500 square kilometres within the central granite zone of the Damara belt, South West Africa, revealed Pre - Damara (Abbabis) basement unconformably overlain by metasediments of the Damara Supergroup. The term Leeukop member is proposed for basal metaconglomerates of the Nosib Group that immediately overlie the basement augen-gneisses. Augen-gneiss clasts are present within the Leeukop metaconglomerates. The Damara orogeny has only partly affected the Abbabis rocks of the Tumas River Inlier but further to the west the Husab suite of red granites and granite-gneisses, as field and geochemical evidence suggest, were derived syntectonically during the Damara orogeny by reactivation of the pre- Damara basement. RĂ”ssing alaskitic granites represent late stage melts, that were also derived from Pre- Damara basement rocks during orogenesis, which accumulated post-tectonically in structural traps at the base of the Khomas Subgroup. Salem granitoids are present in synclinal structures associated with metasediments of the Khomas Subgroup and syntectonic derivation by anatexis during the Damara orogeny is suggested. In the east the differentiated Gawib granitoid stock was emplaced post- tectonically through basement rocks into the Damara metasediments. A deep seated origin is indicated by high crystallisation temperatures (>8500°) obtained from quarternary Qz-An-Ab-Or- H₂0 plots. The metamorphic grade increases westwards from medium grade to high grade. In the east, the metapelites contain andalusite, and coexisting muscovite and quartz. This indicates that temperatures of 6000° at 3,5- 4 kb pressure were attained. In the west, coexisting wollastonite and anorthite in the Khan gneisses indicate pressure- temperature conditions of 720° at 4,5-5 kb. Two tectonic events were responsible for the regional structure. An early F₁ episode produced east- west oriented overturned folds and was followed by an intense F₂ episode of isoclinal folding which is responsible for the dominant northeast - southwest regional fabric. The interference of these folds in the proximity of the underlying basement produced the complex dome and basin structures seen in the central and western parts of the area. An F₃ episode of minor importance was also recognized. The presence of continental basement rocks in this central part of the Damara belt is evidence for formation of the orogen by in-situ deformation rather than continental collision

    Structures for Environmental Action

    This article develops a typology of what we term “structures for action”—strategies, mechanisms, and means—used by local environmental groups to facilitate actions such as lifestyle shifts, civic protest, and environmental preservation. Based on data from nineteen groups in several states, we distinguish between internal structures that facilitate action for members of the groups and external structures that facilitate action among nonmembers and other groups. Within both internal and external structures, we identify three dimensions: knowledge, meaning, and praxis. Our typology of structures for action is designed to stimulate further research and to be useful for environmental groups, as well as for other social issue-oriented local groups that seek to be more effective

    Using existing infrastructure as ground control points to support citizen science coastal UAS monitoring programs

    Recent publications have described the ability of citizen scientists to conduct unoccupied aerial system (UAS) flights to collect data for coastal management. Ground control points (GCPs) can be collected to georeference these data, however collecting ground control points require expensive surveying equipment not accessible to citizen scientists. Instead, existing infrastructure can be used as naturally occurring ground control points (NGCPs), although availably of naturally occurring ground control point placement on such infrastructure differs from published best practices of ground control point placement. This study therefore evaluates the achievable accuracy of sites georeferenced with naturally occurring ground control points through an analysis of 20 diverse coastal sites. At most sites naturally occurring ground control points produced horizontal and vertical root mean square errors (RMSE) less than 0.060 m which are similar to those obtained using traditional ground control points. To support future unoccupied aerial system citizen science coastal monitoring programs, an assessment to determine the optimal naturally occurring ground control point quantity and distribution was conducted for six coastal sites. Results revealed that generally at least seven naturally occurring ground control points collected in the broadest distribution across the site will result in a horizontal and vertical root mean square errors less than 0.030 m and 0.075 m respectively. However, the relationship between these placement characteristics and root mean square errors was poor, indicating that georeferencing accuracy using naturally occurring ground control points cannot be optimized solely through ideal quantity and distribution. The results of these studies highlight the value of naturally occurring ground control points to support unoccupied aerial system citizen science coastal monitoring programs, however they also indicate a need for an initial accuracy assessment of sites surveyed with naturally occurring ground control points at the onset of such programs
