    Beyond A/B Testing: Sequential Randomization for Developing Interventions in Scaled Digital Learning Environments

    Randomized experiments ensure robust causal inference that are critical to effective learning analytics research and practice. However, traditional randomized experiments, like A/B tests, are limiting in large scale digital learning environments. While traditional experiments can accurately compare two treatment options, they are less able to inform how to adapt interventions to continually meet learners' diverse needs. In this work, we introduce a trial design for developing adaptive interventions in scaled digital learning environments -- the sequential randomized trial (SRT). With the goal of improving learner experience and developing interventions that benefit all learners at all times, SRTs inform how to sequence, time, and personalize interventions. In this paper, we provide an overview of SRTs, and we illustrate the advantages they hold compared to traditional experiments. We describe a novel SRT run in a large scale data science MOOC. The trial results contextualize how learner engagement can be addressed through inclusive culturally targeted reminder emails. We also provide practical advice for researchers who aim to run their own SRTs to develop adaptive interventions in scaled digital learning environments

    Strategic Role, Challenges And Opportunities For Small And Medium Enterprises Facing DR-CAFTA: The Case Of Costa Rica

    The development of regional business treaties and the subsequent removal of trade barriers has created new paradigms and challenges for companies in Latin America, especially in fast-growing Central America. This paper examines the role, challenges, and opportunities for small and medium enterprises (SMEs) in Costa Rica relative to the Dominican Republic-Central American Free Trade Agreement (DR-CAFTA, or CAFTA in the US). Opportunities and challenges facing SMEs and the potential impact of DR-CAFTA on industry performances in Costa Rica are discerned. Central American nations and Costa Rica in particular, are facing new challenges, threats and opportunities that arise from trade agreements. Such is the case with DR-CAFTA. These paradigms and competitive pressures will directly impact external trade and sustainable economic development. Opportunities are associated with new laws and regulations, incentives to reinforce and improve business practices especially with regard to human capital, as well as new scenarios for international cooperation, commercial agreements and the growing use of outsourcing. The implications of DR-CAFTA are far-reaching and directly impact the development of small and medium enterprises in Costa Rica

    Efecto de la impregnación al vacío sobre los cambios físicos durante la elaboración de aceitunas de mesa

    Among the benefits which vacuum impregnation (VI) may provide to fruits and vegetables, this study focused on weight and texture changes during the processing of table olives. VI applied to Manzanilla olives led to around 10% weight gain, which was maintained after their packing as black olives. However, this weight gain was only around 4 % for Hojiblanca olives. Likewise, the use of calcium chloride was recommended to maintain the firmness of the olives, in particular those of the softer Manzanilla cultivar. With regard to the Spanish-style, the Hojiblanca cultivar achieved around 4% weight gain during processing but the use of VI for Manzanilla olives was ruled out due to softening of the fruit. In addition. the black and green color of olives and their flavor were not modified by the application of VI. This technology could be very useful to reduce weight loss during table olive processing.Entre los beneficios que la aplicación de la impregnación al vacío puede originar en frutas y verduras, este estudio se ha centrado sobre los cambios en la textura y el peso de las aceitunas de mesa durante su procesamiento. La impregnación al vacío de aceitunas Manzanilla dio lugar a un aumento cercano al 10 % de peso y ello se mantuvo después de su envasado como aceitunas negras, mientras que esta ganancia fue de sólo el 4& para aceitunas Hojiblanca. Asimismo, se vio necesario el empleo de cloruro cálcico para el mantenimiento de la textura de los frutos, en particular para aquellos de la variedad Manzanilla. Con respecto al procesamiento como verdes estilo español, la variedad Hojiblanca consiguió un aumento del 4% en peso debido al empleo de impregnación al vacío y este tratamiento se descartó para la Manzanilla debido al ablandamiento de la aceituna. Además, se debe indicar que el color verde y negro de las aceitunas no se vio afectado por el hecho de emplear impregnación al vacío. Esta tecnología podría ser muy útil para reducir las pérdidas de peso que se producen durante la elaboración de aceitunas de mesa

Currently, inclusion refers to equal opportunities for the 3.0 person and it is not limited to special physical needs or reducing the digital divide.  Faced with this reality, universities, in general must transform themselves. Open universities, in particular, will have to renovate themselves and build the scenarios required by new students.     Costa Rica's economy like that of many developing and developed nations is largely service-based. The information and communication technologies (ICT) sector plays an increasingly key role in strengthening the service aspect of the economy.  A large part of the ICT sector is the software sector. This study is an update of a review of that sector’s contributions conducted in 2008.  In that effort, software producers’ characteristics and needs were investigated. The current article investigates the evolution of the sector and its competitive position in the business sector. The study is national in scope and represents a random sample of firms in the industry. The findings highlight the achievements and remaining challenges for the industry and provide public policy recommendations

    Quantitative proteomics reveals tissue-specific, infection-induced and species-specific neutrophil protein signatures

    Neutrophils are one of the first responders to infection and are a key component of the innate immune system through their ability to phagocytose and kill invading pathogens, secrete antimicrobial molecules and produce extracellular traps. Neutrophils are produced in the bone marrow, circulate within the blood and upon immune challenge migrate to the site of infection. We wanted to understand whether this transition shapes the mouse neutrophil protein landscape, how the mouse neutrophil proteome is impacted by systemic infection and perform a comparative analysis of human and mouse neutrophils. Using quantitative mass spectrometry we reveal tissue-specific, infection-induced and species-specific neutrophil protein signatures. We show a high degree of proteomic conservation between mouse bone marrow, blood and peritoneal neutrophils, but also identify key differences in the molecules that these cells express for sensing and responding to their environment. Systemic infection triggers a change in the bone marrow neutrophil population with considerable impact on the core machinery for protein synthesis and DNA replication along with environmental sensors. We also reveal profound differences in mouse and human blood neutrophils, particularly their granule contents. Our proteomics data provides a valuable resource for understanding neutrophil function and phenotypes across species and model systems.</p

    Optimizing Spectronaut Search Parameters to Improve Data Quality with Minimal Proteome Coverage Reductions in DIA Analyses of Heterogeneous Samples

    Data-independent acquisition has seen breakthroughs that enable comprehensive proteome profiling using short gradients. As the proteome coverage continues to increase, the quality of the data generated becomes much more relevant. Using Spectronaut, we show that the default search parameters can be easily optimized to minimize the occurrence of false positives across different samples. Using an immunological infection model system to demonstrate the impact of adjusting search settings, we analyzed Mus musculus macrophages and compared their proteome to macrophages spiked withCandida albicans. This experimental system enabled the identification of “false positives” as Candida albicans peptides and proteins should not be present in the Mus musculus-only samples. We show that adjusting the search parameters reduced “false positive” identifications by 89% at the peptide and protein level, thereby considerably increasing the quality of the data. We also show that these optimized parameters incurred a moderate cost, only reducing the overall number of “true positive” identifications across each biological replicate by &lt;6.7% at both the peptide and protein level. We believe the value of our updated search parameters extends beyond a two-organism analysis and would be of great value to any DIA experiment analyzing heterogeneous populations of cell types or tissues.</p
