    The effects of man-made drainage systems on the water quality of receiving bodies Is relatively unknown. During the ice-free season of 1979, thirteen drainage ditches and f our rivers were sampled at intervals of three to eight days. With increasing flow, total orthophosphatephosphorous, total Kjeldahl-nitrogen, nitrate-nitrogen and total dissolved solids increased, but not proportionately to f low, with total orthophosphate-phosphorous showing the least proportionality. Variation was observed in each ditch and river when broken down seasonally for each parameter. Generally, when the percentage of flow was greater in one of the three seasons, the percentage of each of the other parameters followed the same trend

    Non-equilibrium sedimentation of colloids on the particle scale

    We investigate sedimentation of model hard sphere-like colloidal dispersions confined in horizontal capillaries using laser scanning confocal microscopy, dynamical density functional theory, and Brownian dynamics computer simulations. For homogenized initial states we obtain quantitative agreement of the results from the respective approaches for the time evolution of the one-body density distribution and the osmotic pressure on the walls. We demonstrate that single particle information can be obtained experimentally in systems that were initialized further out-of-equilibrium such that complex lateral patterns form.Comment: to be published in Phys.     Phase-separation of miscible liquids in a centrifuge

    We show that a liquid mixture in the thermodynamically stable homogeneous phase can undergo a phase-separation transition when rotated at sufficiently high frequency ω\omega. This phase-transition is different from the usual case where two liquids are immiscible or where the slow sedimentation process of one component (e.g. a polymer) is accelerated due to centrifugation. For a binary mixture, the main coupling is due to a term Δρ(ωr)2\propto \Delta\rho(\omega r)^2, where Δρ\Delta\rho is the difference between the two liquid densities and rr the distance from the rotation axis. Below the critical temperature there is a critical rotation frequency ωc\omega_c, below which smooth density gradients occur. When ω>ωc\omega>\omega_c, we find a sharp interface between the low density liquid close to the center of the centrifuge and a high density liquid far from the center. These findings may be relevant to various separation processes and to the control of chemical reactions, in particular their kinetics.Comment: 6 pages, 3 figure

    Evidence for down-regulation of beta-2-adreno-ceptors in cirrhotic patients with severe ascites

    The density and affinity of beta-2-adrenoceptors on mononuclear cells from peripheral blood were studied in fifteen patients with cirrhosis of different severity and in thirteen controls. There was no significant difference between cirrhotic patients and controls in density or affinity of beta-2 binding sites. Within the cirrhotic group, however, the number of binding sites per cell was significantly lower in patients with severe ascites than in patients with mild to moderate or no ascites. This down-regulation of beta-adrenoceptors could influence the haemodynamic response to beta-blockers

    Target cell-dependent T cell-mediated lysis of vaccinia virus-infected cells

    Vaccinia virus specific cytotoxicity against infected target cells was observed in vitro. Spleen lymphocytes from normal and immunized mice of the inbred strains C3H and DBA/2 were incubated with vaccinia virus-infected and non-infected 51Cr-labeled mastocytoma P-815-X2 cells and L-929 fibroblasts, which were used as targets. Cytotoxic lymphocytes could be isolated from the mice as early as 2 days after infection with vaccinia virus. The highest cytotoxic effect was obtained with lymphocytes taken 6 days after infection. The degree of lysi was correlated with the ratio of immune lymphocytes to target cells. Specific blocking of target cell lysis resulted after addition of anti-vaccinia antibody from different sources. The effector cells could be characterized as T cells by elimination of macrophages and B cells. Target cell killing was only possible in a syngeneic system; allogeneic infected target cells were not lysed significantly

    Comparison of Ultrastructural Cytotoxic Effects of Carbon and Carbon/Iron Particulates on Human Monocyte-Derived Macrophages

    In this study, we tested the hypothesis that the presence of iron in carbon particulates enhances ultrastructural perturbation in human monocyte-derived macrophages (MDMs) after phagocytosis. We used 1-μm synthetic carbon-based particulates, designed to simulate environmental particulates of mass median aerodynamic diameter ≤ 2.5 μm (PM(2.5)). Cultures of human MDMs or T-lymphocytes (as a nonphagocytic control) were exposed to carbon or carbon/iron particulates for various time periods and examined by transmission electron microscopy for ultrastructural changes. T-cells failed to internalize either of the particulates and showed no organelle or nuclear changes. Conversely, MDMs avidly phagocytized the particulates. MDMs treated with C particulates exhibited morphologic evidence of macrophage activation but no evidence of lysis of organelles. In contrast, MDMs treated with C/Fe particulates exhibited coalescence of particulate-containing lysosomes. This phenomenon was not observed in the case of C particulates. By 24 hr there was a tendency of the C/Fe particulates to agglomerate into loose or compact clusters. Surrounding the compact C/Fe agglomerates was a uniform zone of nearly total organelle lysis. The lytic changes diminished in proportion to the distance from the agglomerate. In such cells, the nucleus showed loss of chromatin. Although C particles induced no detectable oxidative burst on treated MDMs, C/Fe particles induced a nearly 5-fold increase in the extracellular oxidative burst by treated MDMs compared with untreated controls. Iron bound to C particles catalyzed the decomposition of hydrogen peroxide to generate hydroxyl radicals. Results of these studies suggest that, among particulates of similar size, biologic activity can vary profoundly as a function of particulate physicochemical properties
