    Expression and Clinical Relevance of uPA and ET-1 in Non-small Cell Lung Cancer

    Background and objective uPA and ET-1 proteins have been reported to be up-regulated in some of human cancers. The aim of this study is to investigate the alteration and clinical relevance of uPA and ET-1 protein levels in non-small cell lung cancer (NSCLC). Methods Expressions of uPA and ET-1 protein were detected in 155 cases of NSCLC with tissue microarrays and immunohistochemistry (TMA-IHC) technique. The correlations between the alteration of the two proteins and clinicopathological parameters were analyzed. Results Negative/weak, moderate and high expression of uPA were observed in 12.3%, 64.4% and 23.3% of squamous cell carcinomas, in 12.2%, 53.7% and 34.1% of adenocarcinomas, and in 12.3%, 58.7% and 29.0% of all cases. ET-1 presented negative/weak, moderate and high expression in 2.7%, 42.5% and 54.8% of squamous cell carcinomas, in 11.0%, 30.5% and 58.5% of adenocarcinomas, and in 7.1%, 36.1% and 56.8% of all cases. Simultaneously high expression of uPA and ET-1 were found in adenocarcinomas without lymph node metastasis (P=0.017). Adenocarcinoma patients with high expression of uPA or with high expression of both ET-1 and uPA had the longer survival time (P=0.007 and 0.016). Conclusion Detection of uPA and ET-1 protein levels might contribute to the prognosis evaluation of NSCLC

    ImageManip: Image-based Robotic Manipulation with Affordance-guided Next View Selection

    In the realm of future home-assistant robots, 3D articulated object manipulation is essential for enabling robots to interact with their environment. Many existing studies make use of 3D point clouds as the primary input for manipulation policies. However, this approach encounters challenges due to data sparsity and the significant cost associated with acquiring point cloud data, which can limit its practicality. In contrast, RGB images offer high-resolution observations using cost effective devices but lack spatial 3D geometric information. To overcome these limitations, we present a novel image-based robotic manipulation framework. This framework is designed to capture multiple perspectives of the target object and infer depth information to complement its geometry. Initially, the system employs an eye-on-hand RGB camera to capture an overall view of the target object. It predicts the initial depth map and a coarse affordance map. The affordance map indicates actionable areas on the object and serves as a constraint for selecting subsequent viewpoints. Based on the global visual prior, we adaptively identify the optimal next viewpoint for a detailed observation of the potential manipulation success area. We leverage geometric consistency to fuse the views, resulting in a refined depth map and a more precise affordance map for robot manipulation decisions. By comparing with prior works that adopt point clouds or RGB images as inputs, we demonstrate the effectiveness and practicality of our method.     Genome-wide characterization of endogenous retroviruses in snub-nosed monkeys

    Background Endogenous retroviruses (ERV) are remnants of former exogenous retroviruses that had previously invaded the germ line of the host that can be vertically transmitted across generations. While the majority of ERVs lack infectious capacity due to the accumulation of deleterious mutations, some ERVs remain active and produce potentially infectious viral particles. ERV sequences have been reported in all mammals; however, the distribution and diversity of ERVs in several primate taxa remains unclear. The aim of this study was to identify and classify the ERV sequences in the genomes of the golden snub-nosed monkey (Rhinopithecus roxellana) and the black and white snub-nosed monkey (Rhinopithecus bieti), two endangered primate species that exploit high altitude (2,500–4,500 m) temperate forests in southern and central China. Methods We used a TBLASTN program to search the ERV sequences of golden snub-nosed monkey genome and the black and white snub-nosed monkey genome. We retrieved all complete accession sequences from the homology search and then used the program, RetroTector, to check and identify the ERV sequences. Results We identified 284 and 263 endogenous retrovirus sequences in R. roxellana and R. bieti respectively. The proportion of full-length sequences of all ERV was 30% in R. roxellana and 21% in R. bieti and they were described as class I and class II or gamma-retrovirus and beta-retrovirus genera. The truncation pattern distribution in the two species was virtually identical. By analyzing and comparing ERV orthologues among 6 primate species, we identified the co-evolution of ERVs with their host. We also examined ERV-like sequences and found 48 such genes in R. roxellana and 63 in R. bieti. Some of those genes are associated with diseases, suggesting that ERVs might have involved the abnormal expression of certain genes that have contributed to deleterious consequences for the host. Conclusions Our results indicate that ERV sequences are widely distributed in snub-nosed monkeys, and their phylogenetic history can mirror that of their hosts over long evolutionary time scales. In addition, ERV sequences appear to have an important influence on the evolution of host pathology

    Nutlin-3 overcomes arsenic trioxide resistance and tumor metastasis mediated by mutant p53 in Hepatocellular Carcinoma

    Background: Arsenic trioxide has been demonstrated as an effective anti-cancer drug against leukemia and solid tumors both in vitro and in vivo. However, recent phase II trials demonstrated that single agent arsenic trioxide was poorly effective against hepatocellular carcinoma (HCC), which might be due to drug resistance. Methods: Mutation detection of p53 gene in arsenic trioxide resistant HCC cell lines was performed. The therapeutic effects of arsenic trioxide and Nutlin-3 on HCC were evaluated both in vitro and in vivo. A series of experiments including MTT, apoptosis assays, co-Immunoprecipitation, siRNA transfection, lentiviral infection, cell migration, invasion, and epithelial-mesenchy-mal transition (EMT) assays were performed to investigate the underlying mechanisms. Results: The acquisition of p53 mutation contributed to arsenic trioxide resistance and enhanced metastatic potential of HCC cells. Mutant p53 (Mutp53) silence could re-sensitize HCC resistant cells to arsenic trioxide and inhibit the metastatic activities, while mutp53 overexpression showed the opposite effects. Neither arsenic trioxide nor Nutlin-3 could exhibit obvious effects against arsenic trioxide resistant HCC cells, while combination of them showed significant effects. Nutlin-3 can not only increase the intracellular arsenicals through inhibition of p-gp but also promote the p73 activation and mutp53 degradation mediated by arsenic trioxide. In vivo experiments indicated that Nutlin-3 can potentiate the antitumor activities of arsenic trioxide in an orthotopic hepatic tumor model and inhibit the metastasis to lung. Conclusions: Acquisitions of p53 mutations contributed to the resistance of HCC to arsenic trioxide. Nutlin-3 could overcome arsenic trioxide resistance and inhibit tumor metastasis through p73 activation and promoting mutant p53 degradation mediated by arsenic trioxide

    Association of dietary patterns and sarcopenia in the elderly population: a cross-sectional study

    Background: Sarcopenia, defined as the loss of muscle mass and strength, has been associated with increased hospitalization and mortality. Dietary pattern analysis is a whole diet approach which in this study was used to investigate the relationship between diet and sarcopenia. This study aims to estimate the prevalence of sarcopenia and explore possible factors associated with it among a large population in Beijing, China.Methods: A cross-sectional study with 1,059 participants aged more than 50 years was performed. Sarcopenia was defined based on the guidelines of the Asian Working Group for Sarcopenia. The total score of the MNA-SF questionnaire was used to analyse nutrition status. The baseline demographic information, diet structure and eating habits were collected by clinicians trained in questionnaire data collection and anthropometric and bioimpedance measurements.Results: The overall prevalence of sarcopenia was 8.8% and increased with age: 5%, 5.8%, 10.3% and 26.2% in the 50–59, 60–69, 70–79, and ≥80 years groups, respectively. Marital status (with or without a spouse) was not an independent factor associated with sarcopenia adjusted by age and sex. However, nutritional risk or malnutrition, vegetable diet, advanced age and spicy eating habits were risk factors for sarcopenia. Meanwhile, daily fruit, dairy and nut consumption were protective factors against sarcopenia adjusted by age, sex, income status and spouse status.Conclusion: Although further studies are required to explore the association between healthy dietary patterns and the risk of sarcopenia, the present study provides basic data for identifying correlates of sarcopenia in elderly Chinese individual

    Whole-genome sequencing of the snub-nosed monkey provides insights into folivory and evolutionary history

    Colobines are a unique group of Old World monkeys that principally eat leaves and seeds rather than fruits and insects. We report the sequencing at 146× coverage, de novo assembly and analyses of the genome of a male golden snub-nosed monkey (Rhinopithecus roxellana) and resequencing at 30× coverage of three related species (Rhinopithecus bieti, Rhinopithecus brelichi and Rhinopithecus strykeri). Comparative analyses showed that Asian colobines have an enhanced ability to derive energy from fatty acids and to degrade xenobiotics. We found evidence for functional evolution in the colobine RNASE1 gene, encoding a key secretory RNase that digests the high concentrations of bacterial RNA derived from symbiotic microflora. Demographic reconstructions indicated that the profile of ancient effective population sizes for R. roxellana more closely resembles that of giant panda rather than its congeners. These findings offer new insights into the dietary adaptations and evolutionary history of colobine primates

    High-Range and High-Linearity 2D Angle Measurement System for a Fast Steering Mirror

    In order to solve the problem of the insufficient range of the traditional fast mirror (FSM) angle measurement system in practical applications, a 2D large-angle FSM photoelectric angle measurement system based on the principle of diffuse reflection is proposed. A mathematical model of the angle measurement system is established by combining the physical properties of the diffuse reflecting plate, such as the rotation angle, rotation center, rotation radius, reflection coefficient and the radius of the diffuse reflecting surface. This paper proposes a method that optimizes the degree of nonlinearity based on this mathematical model. The system is designed and tested. The experimental results show that changing the diffuse reflection surface area can improve the nonlinearity of the angle measurement system effectively. When the radius of the diffuse reflection surface is 3.3 mm, the range is ±20°, the non-linearity is 0.74%, and the resolution can reach up to 2.3″. The system’s body is simple and compact. It is also capable of measuring a wider range of angles while linearity is guaranteed

    Research on Attribute Analysis and Modeling of Color Harmony Based on Objective and Subjective Factors

    Color harmony is the hotspot of many researchers in the fields of art and design which is widely used in various artworks and design activities. As for psychology, color harmony is closely related to human’s perception and cognition, although, the research of its generation process has not been fully focused on. With experimental psychology with artificial intelligence technology combined together, our aim is to investigate the factors affecting color harmony on the basis of the generation process from perception to cognition, and construct a mathematical model to predict its degree, so as to realize the quantitatively analysis and measurement of color harmony. We classify the factors affecting color harmony into objective and subjective ones. Among them, the objective factors are constructed based on the three principle elements of color with a total of 21-dimensional color-pair objective physical features extracted. And the subjective factors are divided into direct psychological effects and indirect psychological effects. 180 two-color combinations were evaluated by 30 subjects based on color harmony and its subjective factors on discrete scales ranging from −2 to 2. Correlation analysis reveals that color harmony is not only affected by objective factors, but also by subjective factors which has a stronger correlation than that of the objective ones. Finally, machine learning algorithms were adopted to construct a mathematical model, which has been proved that the linear relationship can well explain the generation mechanism of color harmony

    Comprehensive evaluation of nutritional status before and after hematopoietic stem cell transplantation in 170 patients with hematological diseases

    Objective: To investigate the nutritional status of patients before and after hematopoietic stem cell transplantation (HSCT), and explore optimal methods for assessing nutritional status in patients with hematological diseases. Methods: This cohort study enrolled 170 patients who were diagnosed with hematological diseases and underwent allogeneic HSCT in the Department of Hematology, Peking University People's Hospital between May 2011 and April 2013. We used fixed-point continuous sampling and four nutritional screening tools, Nutritional Risk Screening 2002 (NRS-2002), Mini Nutritional Assessment (MNA), Subjective Global Assessment (SGA) and Malnutrition Universal Screening Tools (MUST), in combination with body measurements, to extensively screen and evaluate nutritional risks and status in patients receiving HSCT before entering and after leaving laminar air flow rooms. Results: After HSCT, patients had significant reduction in weight, hip circumference, waist-hip ratio, calf circumference, mid-upper arm circumference, and suprailiac skinfold thickness compared with pre-HSCT measurements. Before HSCT, NRS-2002 identified that 21.2% of patients were at nutritional risks, compared with 100% after HSCT. MUST indicated that before HSCT, 11.77% of patients were at high nutritional risk, compared with 59.63% after HSCT. MNA assessed that 0.06% of patients were malnourished before HSCT, compared with 19.27% after HSCT. SGA identified that before HSCT, 1.76% of patients had mild to severe malnutrition, which increased to 83.3% after HSCT. There is a significant increase in the nutritional risk and malnutrition in patients who received HSCT. Conclusions: Before HSCT, some patients already had nutritional risk or nutritional deficiencies, and prompt and close nutritional screening or assessment should be performed. The nutritional status of patients after HSCT was generally deteriorated compared with that before transplantation. Body measurements should be taken more frequently during the subsequent treatment window in the laminar air flow rooms. After HSCT, it is recommended to combine MNA and SGA to fully evaluate the nutritional status, and thus provide timely and reasonable nutritional support.SCI(E)中国科技核心期刊(ISTIC)中国科学引文数据库(CSCD)ARTICLE6626-6332
