    Gastrointestinal tract size, total-tract digestibility, and rumen microflora in different dairy cow genotypes

    peer-reviewedThe superior milk production efficiency of Jersey (JE) and Jersey × Holstein-Friesian (JE × HF) cows compared with Holstein-Friesian (HF) has been widely published. The biological differences among dairy cow genotypes, which could contribute to the milk production efficiency differences, have not been as widely studied however. A series of component studies were conducted using cows sourced from a longer-term genotype comparison study (JE, JE × HF, and HF). The objectives were to (1) determine if differences exist among genotypes regarding gastrointestinal tract (GIT) weight, (2) assess and quantify whether the genotypes tested differ in their ability to digest perennial ryegrass, and (3) examine the relative abundance of specific rumen microbial populations potentially relating to feed digestibility. Over 3 yr, the GIT weight was obtained from 33 HF, 35 JE, and 27 JE × HF nonlactating cows postslaughter. During the dry period the cows were offered a perennial ryegrass silage diet at maintenance level. The unadjusted GIT weight was heavier for the HF than for JE and JE × HF. When expressed as a proportion of body weight (BW), JE and JE × HF had a heavier GIT weight than HF. In vivo digestibility was evaluated on 16 each of JE, JE × HF, and HF lactating dairy cows. Cows were individually stalled, allowing for the total collection of feces and were offered freshly cut grass twice daily. During this time, daily milk yield, BW, and dry matter intake (DMI) were greater for HF and JE × HF than for JE; milk fat and protein concentration ranked oppositely. Daily milk solids yield did not differ among the 3 genotypes. Intake capacity, expressed as DMI per BW, tended to be different among treatments, with JE having the greatest DMI per BW, HF the lowest, and JE × HF being intermediate. Production efficiency, expressed as milk solids per DMI, was higher for JE than HF and JE × HF. Digestive efficiency, expressed as digestibility of dry matter, organic matter, N, neutral detergent fiber, and acid detergent fiber, was higher for JE than HF. In grazing cows (n = 15 per genotype) samples of rumen fluid, collected using a transesophageal sampling device, were analyzed to determine the relative abundance of rumen microbial populations of cellulolytic bacteria, protozoa, and fungi. These are critically important for fermentation of feed into short-chain fatty acids. A decrease was observed in the relative abundance of Ruminococcus flavefaciens in the JE rumen compared with HF and JE × HF. We can deduce from this study that the JE genotype has greater digestibility and a different rumen microbial population than HF. Jersey and JE × HF cows had a proportionally greater GIT weight than HF. These differences are likely to contribute to the production efficiency differences among genotypes previously reported

    An investigation into the factors associated with ewe colostrum production

    peer-reviewedThe majority of lamb mortality which occurs during the first 24 h post-partum is preventable through providing the lamb with sufficient quantities of high quality colostrum during this time. Data from seven late gestation nutrition experiments carried out at this institute between 2002 and 2014 were collated into a single data set comprising of 415 twin bearing ewes. Analysis was carried out to investigate the key drivers of ewe colostrum production excluding nutrient intake, namely body reserve mobilisation, ewe breed type, ewe age, gestation length and lamb birth weight. The volume of colostrum produced at 1 and 18 h post-partum was significantly lower than the volume recorded at 10 h post-partum (P = 0.01). Multivariate regression analysis indicated that colostrum volume during the first 18 h post-partum was influenced by lamb birth weight (P = 0.01), ewe age (P = 0.01), breed type (P = 0. 01) and gestation length (P = 0.06). Live weight change (P = 0.05) also had a significant influence on the volume of colostrum produced but BCS change did not affect colostrum production (P = 0.25). Further multivariate regression analysis indicated that IgG yield was influenced ewe breed type (P = 0.01), lamb birth weight (P = 0.02), gestation length (P = 0.05) and BCS change (P = 0.04). Live weight change (P = 0.12) and ewe age (P = 0.62) did not influence the quantity of IgG produced. Leicester ewes produced less colostrum per kg lamb birth weight at 1 h post-partum compared to all other ewe breed types (P = 0.01) and less than Suffolk ewes at 10 h post-partum (P = 0.01). The result of this analysis shows the key factors excluding ewe nutrition that drive colostrum production. Ewe breed type in particular appears to play an important role in the ability of the ewe to produce sufficient quantities of adequate quality colostrum. In conclusion the result of this analysis highlights the important factors associated with ewe colostrum volume and IgG yield excluding nutrition. In particular the overall structure of the flock such as breed type and ewe age is important when considering the ability of the flock to meet colostrum demands and hence reduce lamb mortality

    We assess the international competitiveness of the dairy industries in Argentina and Chile, combining recent market intelligence gathered from field visits with quantitative simulations of global policy reform scenarios. Both countries exhibit strong potential for export growth but face significant internal and external barriers to expanding their dairy industries. Global policy reforms would resolve some of the international obstacles to their expansion. Argentina has great potential, but it is handicapped by its current macroeconomic policies, trade policy distortions, and the uncertainty associated with policy implementation. Chile is more limited in terms of natural capacity for expansion, but it has a positive trade and investment environment

    Grazed grass is the cheapest feed source available for ruminant production systems in temperate climates (Finnernan et al. 2010) accounting for 70% and 90% of the diet of dairy cows in Ireland and New Zealand, respectively. Successful operation of grass-based dairy systems is based on achieving large intakes of high quality grass and efficiently converting it into high value milk solids (Prendiville et al. 2010). Prendiville et al. (2009) identified production efficiency differences between Holstein Friesian (HF) and Jersey (J) cows. They found that J had higher milk solids output per 100 kg bodyweight than HF.     Aurora: A Software Radio for Electromagnetic Vector Sensors in Space

    The AERO (Auroral Emission Radio Observer) and VISTA (Vector Interferometry Space Technology using AERO) missions will advance auroral radio science and radio interferometry technology. AERO is intended to qualify and validate electromagnetic vector sensor technology in space while also answering key scientific questions about the nature and sources of auroral radio emissions. These questions cannot be addressed from the ground due to shielding by the ionosphere. VISTA, together with AERO, will provide the first demonstration of interferometric imaging, beamforming, and nulling using electromagnetic vector sensors at low frequencies (100 kHz – 15 MHz) using Space based sensors. A key component of the AERO-VISTA joint mission is the Aurora software radio system which forms the primary mission payload when combined with an electromagnetic vector sensor antenna (VSA). This radio combines the analog, digital, and signal processing necessary to detect and digitize the signals associated with the radio aurora. We provide a detailed discussion of the radio design, implementation, and performance results from early testing of our engineering model units

    Eligibility rates and representativeness of the General Medical Services scheme population in Ireland 2017-2021: A methodological report [version 1; peer review: 2 approved]

    Background: In Ireland, the means tested General Medical Services (GMS) scheme provides access to a range of healthcare services at no or low cost to approximately one third of the population. Individuals eligible for the GMS scheme are often a focus of research, as a population that account for a large proportion of healthcare services use. The aim of this study is to describe the eligibility rates and representativeness of the GMS scheme population over time, with respect to age group, sex, and geographical area in Ireland. Methods: Population data was obtained from the Central Statistics Office (CSO), using 2016 Census figures and projected population figures for 2017-2021. GMS eligibility figures for 2016-2021 were obtained from the HSE Primary Care Reimbursement Service (PCRS). GMS eligibility rates and relative rates of eligibility were calculated for 2016-2021 by age group and sex. Additionally, 2016 eligibility rates were calculated by geographical area. Results: The crude eligibility rate decreased from 36.4% in 2016 to 31.2% in 2020, with a slight increase to 31.6% in 2021. In the 75+ years age group, 78.2% of the total population were eligible for the GMS scheme in 2021. The age group with the lowest rate of eligible individuals was the 25-34 age group, with 19.5% eligible in 2021. The eligibility rate was higher among females compared to males throughout the study period. The highest eligibility rate was seen in Donegal, with a crude rate of 52.8%. Dublin had the lowest rate, with a crude rate of 29.3%. Conclusions: GMS eligibility varies greatly depending on age, sex, and geographical area, and decreased between 2016 and 2021. This study uses the most up-to-date data available to provide age group, sex and area-based figures for GMS eligibility which may inform planning and conduct of research focusing on GMS-eligible individuals

    Targeting the 19S proteasomal subunit, Rpt4, for the treatment of colon cancer.

    Deregulation of the ubiquitin-proteasome pathway has been frequently observed in a number of malignancies. Using quantitative Western blotting of normal and matched tumour tissue, we here identified a significant increase in the 19S proteasome subunit Rpt4 in response to chemoradiation in locally advanced rectal cancer patients with unfavourable outcome. We therefore explored the potential of Rpt4 reduction as a therapeutic strategy in colorectal cancer (CRC). Utilizing siRNA to down regulate Rpt4 expression, we show that silencing of Rpt4 reduced proteasomal activity and induced endoplasmic reticulum stress. Gene silencing of Rpt4 also inhibited cell proliferation, reduced clonogenic survival and induced apoptosis in HCT-116 colon cancer cells. We next developed a cell penetrating peptide-based nanoparticle delivery system to achieve in vivo gene silencing of Rpt4. Administration of Rpt4 siRNA nanoparticles reduced tumour growth and improved survival in a HCT-116 colon cancer xenograft tumour model in vivo. Collectively, our data suggest that inhibition of Rpt4 represents a novel strategy for the treatment of CRC

    Beef production from feedstuffs conserved using new technologies to reduce negative environmental impacts

    End of project reportMost (ca. 86%) Irish farms make some silage. Besides directly providing feed for livestock, the provision of grass silage within integrated grassland systems makes an important positive contribution to effective grazing management and improved forage utilisation by grazing animals, and to effective feed budgeting by farmers. It can also contribute to maintaining the content of desirable species in pastures, and to livestock not succumbing to parasites at sensitive times of the year. Furthermore, the optimal recycling of nutrients collected from housed livestock can often be best achieved by spreading the manures on the land used for producing the conserved feed. On most Irish farms, grass silage will remain the main conserved forage for feeding to livestock during winter for the foreseeable future. However, on some farms high yields of whole-crop (i.e. grain + straw) cereals such as wheat, barley and triticale, and of forage maize, will be an alternative option provided that losses during harvesting, storage and feedout are minimised and that input costs are restrained. These alternative forages have the potential to reliably support high levels of animal performance while avoiding the production of effluent. Their production and use however will need to advantageously integrate into ruminant production systems. A range of technologies can be employed for crop production and conservation, and for beef production, and the optimal options need to be identified. Beef cattle being finished indoors are offered concentrate feedstuffs at rates that range from modest inputs through to ad libitum access. Such concentrates frequently contain high levels of cereals such as barley or wheat. These cereals are generally between 14% to 18% moisture content and tend to be rolled shortly before being included in coarse rations or are more finely processed prior to pelleting. Farmers thinking of using ‘high-moisture grain’ techniques for preserving and processing cereal grains destined for feeding to beef cattle need to know how the yield, conservation efficiency and feeding value of such grains compares with grains conserved using more conventional techniques. European Union policy strongly encourages a sustainable and multifunctional agriculture. Therefore, in addition to providing European consumers with quality food produced within approved systems, agriculture must also contribute positively to the conservation of natural resources and the upkeep of the rural landscape. Plastics are widely used in agriculture and their post-use fate on farms must not harm the environment - they must be managed to support the enduring sustainability of farming systems. There is an absence of information on the efficacy of some new options for covering and sealing silage with plastic sheeting and tyres, and an absence of an inventory of the use, re-use and post-use fate of plastic film on farms. Irish cattle farmers operate a large number of beef production systems, half of which use dairy bred calves. In the current, continuously changing production and market conditions, new beef systems must be considered. A computer package is required that will allow the rapid, repeatable simulation and assessment of alternate beef production systems using appropriate, standardised procedures. There is thus a need to construct, evaluate and utilise computer models of components of beef production systems and to develop mathematical relationships to link system components into a network that would support their integration into an optimal system model. This will provide a framework to integrate physical and financial on-farm conditions with models for estimating feed supply and animal growth patterns. Cash flow and profit/loss results will be developed. This will help identify optimal systems, indicate the cause of failure of imperfect systems and identify areas where applied research data are currently lacking, or more basic research is required
