Expression von Antikoerperderivaten in eukaryonten Zellen Gewinnung chimaerer Antikoerper. Schlussbericht

Antibodies directed against the Interleukin 2 receptor are potentially valuable aspects for immuno-suppressive treatment of patients (after organ transplantation, with autoimmune disease, etc.). In order to provide antibodies with reduced immunogenic potential we have chimerized a murine monoclonal antibody directed against the Alpha-chain of the Interleukin 2 receptor by gene targeting. For this purpose, recombination vectors encoding the constant domain of the light and heavy chains of human antibodies were transfected into the murine hybridoma cell line and stable transfectants isolated by standard methods. Chimeric antibody was isolated from the supernatant of transfectomas and functionally characterized. Alternative methods for chimerization (PCR technology) are competitive to this approach. (orig.)Antikoerper gegen den Interleukin 2-Rezeptor haben moeglicherweise Bedeutung zur immunsuppressiven Behandlung von Patienten nach Organtransplantationen, mit Autoimmunkrankheiten, etc. Um Antikoerper mit reduziertem immunogenen Potential herzustellen, haben wir mittels ''gene targeting'' einen murinen monoklonalen Antikoerper chimaerisiert, welcher gegen die Alpha-Kette des Interleukin 2-Rezeptors gerichtet ist. Hierzu wurden Rekombinationsvektoren, welche die konstanten Domaenen der leichten und schweren Kette humaner Antikoerper kodieren, in die murine Hybridomazellinie transfiziert und stabile Transfektanten mit Standardmethoden isoliert. Aus dem Ueberstand der Transfektomas wurde chimaerer Antikoerper isoliert und funktionelle charakterisiert. Alternativ zu diesem Verfahren wurden inzwischen Methoden beschrieben, die auf der PCR-Technologie beruhen. Several severe diseases were actually treated with therapeutic polypeptides. To identify novel regulatory peptides for the therapy of bone diseases, neuronal diseases and of hematopoetic disorders, we started a comprehensive screening project. This program included the development of cellular assay systems as well as the establishment of a peptide library from thousands of liters of human blood filtrate. As a major result, we isolated two new osteoanabolic peptides and additionally a neuroprotective peptide. These substances are now in preclinical trials and have a promising therapeutic potential.
