    Hemijske modifikacije Ī²-laktoglobulina hinonima

    The avarone/avarol quinone/hydroquinone couple. as well as their derivatives show considerable antitumor activity. In this work, covalent modifications of beta-lactogglobulin. isolated from cow milk by avarone, its model compound 2-tert-butyl-1,4-benzoquinone. and several of their alkylthio derivatives were studied. The techniques applied for as-saying the modifications were: UV/VIS spectrophotometry, SDS PAGE and isoelectrofocusing. The results of the SDS PAGE suggest that polymerisation of the protein occurs. The shift of the pI of the protein upon modification toward lower values indicates that lysine amino groups are the principal site of die reaction of beta-lactoglobulin with the quinones.Hinonsko/hidrohinonski par avaron/avarol i njihovi derivati pokazuju značajnu antitumorsku aktivnost. U ovom radu proučavane su kovalentne modifikacije Ī²-laktoglobulina, izolovanog iz kravljeg mleka, avaronom, njegovim model-jedinjenjem 2-tert-butil-1,4-benzohinonom i njihovim alkiltio-derivatima. Za ispitivanje modifikacija koriŔćene su UV/VIS spektrofotometrija, SDS PAGE i izoelektrofokusiranje. Rezultat SDS PAGE ukazuje da se protein polimerizuje. Pomeranje pI vrednosti proteina nakon modifikacije ka nižim vrednostima pokazuje da su amino grupe lizina glavna mesta reakcije Ī²-laktoglobulina sa hinonima

    The avarone/avarol quinone/hydroquinone couple shows considerable antitumor activity. In this work, covalent modification of beta-lactoglobulin by avarone and its derivatives as well as by the synthetic steroidal quinone 2,5(10)-estradiene-1,4,17-trione and its derivatives were studied. The techniques for studying chemical modification of beta-lactoglobulin by quinones were: UV/Vis spectrophotometry, SDS PAGE and isoelectrofocusing. SDS PAGE results suggest that polymerization of the protein Occurs. It Could be seen that the protein of 18 kD gives the bands of 20 kD, 36 kD, 40 kD, 45 kD, 64 kD and 128 kD depending on modification agent. The shift of the pl of the protein (5.4) upon modification toward lower values (from pl 5.0 to 5.3) indicated that lysine amino groups are the principal site of the reaction of beta-lactoglobulin with the quinones.Hinonsko/hidrohinonski par avaron/avarol pokazuje značajnu antitumorsku aktivnost. U ovom radu proučavane su kovalentne modifikacije Ī²-laktoglobulina avaronom, sintetičkim steroidnim hinonom 2,5(10)-estradien-1,4,17-trionom i njihovim derivatima. Tehnike za praćenje hemijske modifikacije bile su: UV/Vis spektrofotometrija, SDS PAGE i izoelektrofokusiranje. Rezultati SDS PAGE ukazuju da se deÅ”ava polimerizacija proteina.Može se videti da protein od 18 kD daje trake od 20 kD, 36 kD, 40 kD, 45 kD, 64 kD i 128 kD u zavisnosti od agensa za modifikaciju. Pomeranje pI vrednosti proteina (5,4) nakon modifikacije ka nižim vrednostima (od pI 5,0 do 5,3) pokazuje da su amino-grupe lizina glavna mesta reakcije Ī²-laktoglobulina sa hinonima

    Regioselektivnost konjugovane adicijena monoalkil-1,4-benzohinone

    The regioselectivity of the reaction of conjugate addition of thiols, amines, methanol and hydrogen chloride with the monoalkyl-1,4-benzoquinones avarone and 2-tert-butyl-1,4-benzoquinone was investigated. It was shown that the regioselectivity of the reaction is influenced by the electrophilicity of position 5 in unprotonated 2-alkylquinones, the increased electrophilicity of position 6 in acidic medium, and by the acidity of the intermediate hydroquinones.Proučavana je regioselektivnost konjugovane adicije tiola, amina, metanola i hlorovodonika na monoalkil-1,4-benzohinone avaron i 2-tert-butil-1,4-benzohinon. Pokazano je da na regioselektivnost reakcije utiču elektrofilnost položaja 5 neprotonovanih 2-alkil-hinona i povećana elektrofilnost položaja 6 u kiseloj sredini, kao i kiselost intermedijernih hidrohinona

    Bone Morphogenetic Proteins (BMPs) are growth and differentiation factors within the TGFĪ² superfam- ily of proteins. They induce ectopic and orthotopic endochondral bone formation and are involved in the regulation of cell proliferation, differentiation, apoptosis and mesenchymal-epithelial interactions in critical morphogenetic processes of tissues beyond bone. BMP2 and BMP7 osteogenic devices have been approved for enhancing healing in patients with long bone defects and anterior spinal fusion proce- dures. However, due to a high price and various serious adverse events including heterotopic ossifica- tion, retrograde ejaculation and pain their clinical use have been limited. In this review we discuss the BMP discovery, biology and their use in clinical studies with particular reference to the newly developed BMP6 based autologous bone graft substitute (ABGS). A novel ABGS consisting of an autologous bone coagulum (ABC) carrier with dispersed BMP6 to initiate the differentiation of mesenchymal cells into endochondral bone. The ABC met the conditions for an optimal delivery system for BMP6 due to han- dling simplicity, without an immunogenic and inflammatory response at the implantation site. Addition of allograft or synthetic ceramics to ABGS demonstrated in animal models significantly increased volume and better microarchitecture of the newly formed bone. The first clinical study was conducted in patients with distal radial fractures (Phase I study) and the second in patients undergoing high tibial osteotomy (Phase I/II study) and no serious adverse events have been observed. Finally, in the ongoing OSTEO- proSPINE study ABGS enforced with allograft bone is evaluated in patients with chronic back pain due to degenerative disc diseases. The novel ABGS bone mimetic is a major breakthrough and contribution to bone biology and regenerative medicine of skeletal repair.KoÅ”tani morfogenetski proteini (BMP) čine grupu čimbenika rasta i diferencijacije unutar TGFĪ² nado- bitelji. Oni induciraju stvaranje ektopične i ortotopične endohondralne kosti te su uključeni u regulaciju stanične proliferacije, diferencijacije, apoptoze i mezenhimalno-epitelne interakcije u važnim tkivnim morfogenetskim procesima izvan koÅ”tanog sustava. KoÅ”tane naprave koje sadrže BMP2 i BMP7 pro- tein odobrene su za poboljÅ”anje koÅ”tanog cijeljenja kod pacijenata s defektima dugih cjevastih kostiju i kod prednje spinalne fuzije kralježnice. Međutim, zbog visoke cijene i mnogobrojnih nuspojava koje su uključivale pojavu heterotopičnih osifikacija, retrogradnu ejakulaciju i bol, njihova je klinička prim- jena ograničena. U ovom smo preglednom radu raspravili otkriće BMP molekula, njihovu biologiju i primjenu u kliničkim studijama s posebnim osvrtom na nedavno otkrivenu novu autolognu koÅ”tanu napravu (ABGS) koja sadrži BMP6. Novi ABGS sastoji se od nosača autolognog koaguluma (ABC) s otopljenim BMP6 koji je ključan za pokretanje diferencijacije mezenhimalnih stanica u smjeru stvaranja endohondralne kosti. ABC je ispunio sve potrebne uvjete za formulaciju optimalnog nosača za BMP6 isključivo zbog jednostavnosti priprave i primjene te odsustva imunogenog i upalnog odgovora na mjestu implantacije. Uz dodatak alografta ili sintetičke keramike Å”to je potvrđeno na životinjskim modelima doÅ”lo je do značajnog povećanja volumena te poboljÅ”anja mikroarhitekture novonastale kosti. Prvo kliničko ispitivanje provedeno je na pacijentima s distalnim prijelomima radijusa (faza I studije), a drugo na pacijentima koji su podvrgnuti visokoj osteotomiji tibije (faza I/II studije) bez uočenih ozbiljnih nuspojava. Trenutno je u tijeku studija OSTEOproSPINE u kojoj se testira učinkovitost ABGS u kom- binaciji s koÅ”tanim alograftom u bolesnika s kroničnim bolovima u leđima uzrokovanim degenerativnim promjenama intervertebralnog diska. Nova ABGS koÅ”tana naprava značajna je prekretnica i napredak u području koÅ”tane biologije te regenerativne medicine koÅ”tanog sustava

    In this work, inhibition of sulfhydryl containing glycolytic enzyme hexokinase by avarone and its derivatives was studied. The enzyme contains lysine and sulfhydryl residues, which can be modified by the quinone moiety. Our results show that avarone and its derivatives are capable of covalent modification of hexokinase
