Texto completo: acesso restrito. p.109-115The reaction of nickel(II) cation with hydroxynaphthol blue (HNB) in aqueous media at pH 5.2–6.0 results in a red complex that is stable for at least 2h. Beer's Law is obeyed up to 3.2 μg/ml of nickel(II) with an apparent molar absorptivity of 1.38 × 104l/mol/cm at 563 nm. This paper proposes procedures for nickel determination by ordinary and first-derivative spectrophotometry. The results demonstrate that the linear dynamic range is 0.08–3.20 μg/ml with a limit of detection of 23 ng/ml for ordinary spectrophotometry, compared with 21–800 ng/ml and 6 ng/ml, respectively, for first-derivative spectrophotometry. Calcium(II), magnesium(II), barium(II), strontium(II), cadmium(II), lead(II), manganese(II), bismuth(III) and molybdenum(VI) ions do not interfere for at least 100∶1 mass ratios. The main interferents are cobalt(II), titanium(IV), aluminium(III), mercury(II) and copper(II). The interferences of titanium(IV), aluminium(III), zirconium(IV) and iron(III) can be masked by fluoride and mercury(II) and copper(II) with thiosulfate or thiourea. The derivative method is applied to nickel determination in standard brasses and the results demonstrate that there is no significant difference between the results and certified values at the 95% confidence level

Foram realizados estudos com as partes aéreas de P. niruri L. e P. tenellus Roxb., a fim de fornecer subsídios para o controle de qualidade botânico da matéria prima vegetal, determinando-se parâmetros morfológicos que permitem a diagnose das duas espécies, tanto no estado íntegro como fragmentado.Studies were performed on the aerial parts of P. niruri L. and P. tenellus Roxb. The characteristics of botanical quality control of the vegetal raw material were verified for both species. Development and validation of analytical methodology by agar diffusion assay for determination of Ciclopirox olamine topical solution
A freqüência de infecções fúngicas tem aumentado nas duas últimas décadas em decorrência do prolongamento da vida, em razão do avanço na tecnologia médica. Esta sobrevida ocasionou a manifestação de infecções fúngicas em novas proporções, o que aumentou o interesse pelo estudo dos fármacos antifúngicos. O ciclopirox olamina é um agente antifúngico com amplo espectro de ação, com atividades antiinflamatória e antibacteriana. Neste trabalho foi desenvolvido e validado método por difusão em ágar para quantificação de solução tópica de ciclopirox olamina, empregando Candida albicans ATCC 10231 como microrganismo teste. O método apresentou linearidade, precisão e exatidão adequadas.Frequency of fungal infections has risen during the last decades due to the increasing human lifespan, because of the advance in the capacity of medical technology. This survival rate has brought about a manifestation of fungal infections at new proportions, which has raised the interest in studying anti-fungal drugs. Ciclopirox olamina is an anti-fungal agent with a wide spectrum of action, with anti-inflammatory and anti-bacterial activities. In this study, method by agar diffusion for the quantification of ciclopirox olamina as topical solution were developed and validated, employing Candida albicans ATCC 10231. Development and validation of an analytical methodology for determination of Sibutramine in capsules
A Sibutramina é classificada terapeuticamente como anorexígeno, sendo indicada para tratamento da obesidade e redução do peso corpóreo, em conjunto com dieta e exercícios. Encontra-se disponível comercialmente no Brasil na forma farmacêutica de cápsulas. Não existem, até o momento, monografias em códigos oficiais para o controle de qualidade desse fármaco em sua forma farmacêutica. O presente trabalho teve por objetivo desenvolver e validar um método espectrofotométrico na região do ultravioleta para quantificação de sibutramina em cápsulas. A análise do teor de sibutramina foi realizada por espectrofotometria na região do ultravioleta no comprimento de onda de 223 nm utilizando metanol e água como solventes. A validação do método analítico seguiu as normas da RE Nº 899 e da International Conference on Harmonization-ICH. O método analítico validado demonstrou ser adequado, simples e rápido para a quantificação das cápsulas.Sibutramine is therapeutically classified as an anorexigen, being recommended in the treatment of obesity and reduction of body weight, in combination with diet and exercise. It is commercially available in Brazil in the pharmaceutical form of capsules. As far of we know, there are no monographs in official codes for drug quality control in this pharmaceutical form. The present study aimed to develop and validate a spectrophotometric method in the ultraviolet area for sibutramine quantification in capsules. The analysis of the sibutramine content was accomplished by spectrophotometry in the ultraviolet area at 223 nm using methanol and water as solvent. The validation of the analytic method followed the RE 899 norms and International Conference on Harmonization - ICH. Use of the ACP® and BTS extenders for cooling at 15°C white-lipped peccary (Tayassu pecari) semen
ABSTRACT: Knowledge about reproduction of white-lipped peccary is of great importance to assist with the conservation of this species and enable its rational use in captivity. This study aimed to evaluate the effect of ACP-103®, ACP-116® and BTS semen extenders on sperm viability during cooling of Tayassu pecari semen. Five ejaculates from four adult males were chilled. The animals were submitted to the protocols of sedation and anesthesia for semen collection by the electroejaculation method. After collection, the semen was macro- and microscopically assessed and diluted to reach 35x106 spermatozoa/mL in each of the three different extenders tested. The fresh-extended semen was packed in a BotuFLEX® thermal box to keep samples at 15°C for 24 hours. After cooling, the following semen parameters were analyzed: sperm motility, functional and structural integrity of sperm membranes, mitochondrial activity, chromatin condensation, and the thermoresistance test was performed. The parameters sperm motility, structural and functional integrity of sperm membranes, mitochondrial activity, and chromatin condensation were preserved after use of the extenders tested, and were similar to those of in natura semen (p>0.05). Curvilinear velocity (VCL) (p<0.05) was the only parameter with reduced values after cooling regardless of the extender used. The percentage of sperm with normal morphology was greater in samples cooled using the BTS extender (p<0.05). The ACP-103®, ACP-116® and BTS extenders can be used for the cooling and preservation of white-lipped peccary semen at 15°C for 24 hours
The ventral premammillary nucleus links fasting-induced changes in leptin levels and coordinated luteinizing hormone secretion
Physiological conditions of low leptin levels like those observed during negative energy balance are usually characterized by the suppression of luteinizing hormone (LH) secretion and fertility. Leptin administration restores LH levels and reproductive function. Leptin action on LH secretion is thought to be mediated by the brain. However, the neuronal population that mediates this effect is still undefined. The hypothalamic ventral premammillary nucleus (PMV) neurons express a dense concentration of leptin receptors and project to brain areas related to reproductive control. Therefore, we hypothesized that the PMV is well located to mediate leptin action on LH secretion. To test our hypothesis, we performed bilateral excitotoxic lesions of the PMV in adult female rats. PMV-lesioned animals displayed a clear disruption of the estrous cycle, remaining in anestrus for 15–20 d. After apparent recovery of cyclicity, animals perfused in the afternoon of proestrus showed decreased Fos immunoreactivity in the anteroventral periventricular nucleus and in gonadotropin releasing hormone neurons. PMV-lesioned animals also displayed decreased estrogen and LH secretion on proestrus. Lesions caused no changes in mean food intake and body weight up to 7 weeks after surgery. We further tested the ability of leptin to induce LH secretion in PMV-lesioned fasted animals. We found that complete lesions of the PMV precluded leptin stimulation of LH secretion on fasting.
