    Evaluation of the New Jersey Access Initiative mentor program

    The purposes of this study were to (a) evaluate the effectiveness of the New Jersey Access Initiative Mentor Program and (b) determine if an increased number of sessions of ancillary, psychosocial services was related to improved treatment retention, periods of abstinence and decreased number of relapses with opiate-addicted participants. Correlational analyses were planned utilizing data collected and recorded in the NJAI database by program staff; however, analyses were unable to be performed due to a large amount of incomplete and missing data. For those participants with data, descriptive results are reported on alcohol and illegal substance usage in past 30 days prior to data collection, living conditions, employment status, educational level, and number of services provided. Limitations of the study design and potential implications for importance of including access to psychosocial, ancillary services in addition to addiction treatment are discussed

We consider next to leading order terms to the Stirling approximation, and show that they are important at small values of the angular momentum ll and other regimes. We employ the uniform approximation.     Glycoproteomic studies of IgE from a novel hyper IgE syndrome linked to PGM3 mutation

    International audienceGlycans serve as important regulators of antibody activities and half-lives. IgE is the most heavily glycosylated antibody, but in comparison to other antibodies little is known about its glycan structure function relationships. We therefore describe the site specific IgE glycosylation from a patient with a novel hyper IgE syndrome linked to mutations in PGM3, which is an enzyme involved in synthesizing UDP-GlcNAc, a sugar donor widely required for glycosylation. A two-step method was developed to prepare two IgE samples from less than 1 mL of serum collected from a patient with PGM3 mutation and a patient with atopic dermatitis as a control subject. Then, a glycoproteomic strategy was used to study the site-specific glycosylation. No glycosylation was found at Asn264, whilst high mannose glycans were only detected at Asn275, tri-antennary glycans were exclusively observed at Asn99 and Asn252, and non-fucosylated complex glycans were detected at Asn99. The results showed similar glycosylation profiles between the two IgE samples. These observations, together with previous knowledge of IgE glycosylation, imply that IgE glycosylation is similarly regulated among healthy control, allergy and PGM3 related hyper IgE syndrome

    Development and Validation of a New Bitumen Fume Generation System which Generates Polycyclic Aromatic Hydrocarbon Concentrations Proportional to Fume Concentrations

    Bitumen fumes emitted during road paving and roofing contain polycyclic aromatic compounds (PACs) of potential health concern. Little information is available for an experimental device devoted to inhalation experiments with animals exposed to bitumen fumes, and in all studies the systems were never validated for a range of fume concentrations, which prohibited their use for toxicological concentration-effect studies. Therefore, the purpose of this study was to validate a new experimental device able to generate bitumen fumes at different total particulate matter (TPM) concentrations with a linear correlation between TPM and the concentrations of different PACs, thus allowing toxicological dose-response studies with fumes representative of those in the field. Atmosphere samples collected from an animal exposure chamber allowed the determination of TPM, toluene soluble matter, polycyclic aromatic hydrocarbons (PAHs) and semi-volatiles. The particulate size distributions were determined in order to assess the deposition pattern in the respiratory tract. The temperature of 170°C was chosen by analogy with the upper range of the temperature used during paving operations. The temperature of the air passing over the fume emission area was regulated to 20°C and stirring of the heated bitumen was restricted to 90 r.p.m. The data show that the objective of developing a static fume generation system that reproducibly produces fumes in the inhalation chamber for specified target concentrations (TPM) were successful. The within-day variation coefficients for TPM were between 2.5 and 6.1%. The day-to-day variations for TPM concentration were between 4.1 and 5.8%. The concentrations of the 4-5 ring PAHs and the polycyclic aromatic sulphur heterocycles were proportional to the TPM concentration. The 2 and 3 ring PAH concentrations showed a deviation from proportionality with the TPM, probably due to their re-evaporation during sampling. The mass median aerodynamic diameter of airborne particles varied from 1.4 µm at a fume concentration of 5 mg/m3 to 3.2 µm at 100 mg/m3. In conclusion, this equipment was suitable for nose-only inhalation studies in the 5-100 mg/m3 range of TPM. Bitumen fumes were generated with a good reproducibility under well-controlled conditions.     DNA damage-induced interaction between a lineage addiction oncogenic transcription factor and the MRN complex shapes a tissue-specific DNA Damage Response and cancer predisposition

    Since genome instability can drive cancer initiation and progression, cells have evolved highly effective and ubiquitous DNA Damage Response (DDR) programs. However, some cells, in skin for example, are normally exposed to high levels of DNA damaging agents. Whether such high-risk cells possess lineage-specific mechanisms that tailor DNA repair to the tissue remains largely unknown. Here we show, using melanoma as a model, that the microphthalmia-associated transcription factor MITF, a lineage addition oncogene that coordinates many aspects of melanocyte and melanoma biology, plays a non-transcriptional role in shaping the DDR. On exposure to DNA damaging agents, MITF is phosphorylated by ATM/DNA-PKcs, and unexpectedly its interactome is dramatically remodelled; most transcription (co)factors dissociate, and instead MITF interacts with the MRE11-RAD50-NBS1 (MRN) complex. Consequently, cells with high MITF levels accumulate stalled replication forks, and display defects in homologous recombination-mediated repair associated with impaired MRN recruitment to DNA damage. In agreement, high MITF levels are associated with increased SNV burden in melanoma. Significantly, the SUMOylation-defective MITF-E318K melanoma predisposition mutation recapitulates the effects of ATM/DNA-PKcs-phosphorylated MITF. Our data suggest that a non-transcriptional function of a lineage-restricted transcription factor contributes to a tissue-specialised modulation of the DDR that can impact cancer initiation

    Precision Measurement of the Neutron Spin Asymmetry A1nA_1^n and Spin-Flavor Decomposition in the Valence Quark Region

    We have measured the neutron spin asymmetry A1nA_1^n with high precision at three kinematics in the deep inelastic region at x=0.33x=0.33, 0.47 and 0.60, and Q2=2.7Q^2=2.7, 3.5 and 4.8 (GeV/c)2^2, respectively. Our results unambiguously show, for the first time, that A1nA_1^n crosses zero around x=0.47x=0.47 and becomes significantly positive at x=0.60x=0.60. Combined with the world proton data, polarized quark distributions were extracted. Our results, in general, agree with relativistic constituent quark models and with perturbative quantum chromodynamics (pQCD) analyses based on the earlier data. However they deviate from pQCD predictions based on hadron helicity conservation.Comment: 5 pages, 2 figures, this is the final version appeared in Phys. Rev. Let
