    Synaptic Proteins Linked to HIV-1 Infection and Immunoproteasome Induction: Proteomic Analysis of Human Synaptosomes

    Infection of the central nervous system with human immunodeficiency virus type 1 (HIV-1) can produce morphological changes in the neocortical synaptodendritic arbor that are correlated with neurocognitive impairment. To determine whether HIV-1 infection influences the protein composition of human synapses, a proteomic study of isolated nerve endings was undertaken. Synaptosomes from frontal neocortex were isolated using isopyknic centrifugation from 19 human brain specimens. Purity and enrichment were assessed by measuring pre- and postsynaptic protein markers. Two-dimensional polyacrylamide gel electrophoresis and matrix-assisted laser desorption ionization time-of-flight mass spectrometry was used to screen for proteins differentially expressed in HIV/AIDS. The concentrations of 31 candidate protein spots were potentially abnormal in HIV-infected decedents with HIV encephalitis and/or increased expression of immunoproteasome subunits. Immunoblots showed that the concentration of some of them was related to HIV-1 infection of the brain and immunoproteasome (IPS) induction. Synapsin 1b and stathmin were inversely related to brain HIV-1 load; 14-3-3ζ and 14-4-4ε proteins were higher in subjects with HIV-1 loads. Perturbed synaptosome proteins were linked with IPS subunit composition, and 14-3-3ζ was histologically colocalized with IPS subunits in stained neocortical neurons. Proteomics illustrates that certain human proteins within the synaptic compartment are involved with changes in the synaptodendritic arbor and neurocognitive impairment in HIV-1-infected people

    Private Narratives and Infant Views: Iconizing 1970s Militancy in Contemporary Argentine Cinema

    This is the author's accepted manuscript, made available with the permission of the publisher.This article analyses the connections between the subjective turn in the representation of militancy, iconicity, and historical examination in Infancia clandestina, a recent Argentine film that portrays the 1970s armed struggle through a child’s lens. Breaking with the leading interpretation that praises the movie because of its original exposition of left-leaning violence, I contend that this coming-of-age story fits within a version of militancy that originated in the mid-1990s and that has become quite common since the advent of the Kirchner administration in 2003. This particular version relies on a privatized and archaic image of activism that is at the core of the global iconization of 1970s militancy. An analysis of the filmic use of an infant perspective and of anime-style cartoons illuminates how contemporary Argentine cinema both registers and participates in this iconizing process

    Creating spatially continuous maps of past land cover from point estimates: A new statistical approach applied to pollen data

    International audienceReliable estimates of past land cover are critical for assessing potential effects of anthropogenic land-cover changes on past earth surface-climate feedbacks and landscape complexity. Fossil pollen records from lakes and bogs have provided important information on past natural and human-induced vegetation cover. However, those records provide only point estimates of past land cover, and not the spatially continuous maps at regional and sub-continental scales needed for climate modelling. We propose a set of statistical models that create spatially continuous maps of past land cover by combining two data sets: 1) pollen-based point estimates of past land cover (from the REVEALS model) and 2) spatially continuous estimates of past land cover, obtained by combining simulated potential vegetation (from LPJ-GUESS) with an anthropogenic land-cover change scenario (KK10). The proposed models rely on statistical methodology for compositional data and use Gaussian Markov Random Fields to model spatial dependencies in the data. Land-cover reconstructions are presented for three time windows in Europe: 0.05, 0.2, and 6 ka years before present (BP). The models are evaluated through cross-validation, deviance information criteria and by comparing the reconstruction of the 0.05 ka time window to the present-day land-cover data compiled by the European Forest Institute (EFI). For 0.05 ka, the proposed models provide reconstructions that are closer to the EFI data than either the REVEALS-or LPJ-GUESS/KK10-based estimates; thus the statistical combination of the two estimates improves the reconstruction. The reconstruction by the proposed models for 0.2 ka is also good. For 6 ka, however, the large differences between the REVEALS-and LPJ-GUESS/KK10-based estimates reduce the reliability of the proposed models. Possible reasons for the increased differences between REVEALS and LPJ-GUESS/KK10 for older time periods and further improvement of the proposed models are discussed

    From the animal house to the field : are there consistent individual differences in immunological profile in wild populations of field voles (Microtus agrestis)?

    Inbred mouse strains, living in simple laboratory environments far removed from nature, have been shown to vary consistently in their immune response. However, wildlife populations are typically outbreeding and face a multiplicity of challenges, parasitological and otherwise. In this study we seek evidence of consistent difference in immunological profile amongst individuals in the wild. We apply a novel method in this context, using longitudinal (repeated capture) data from natural populations of field voles, Microtus agrestis, on a range of life history and infection metrics, and on gene expression levels. We focus on three immune genes, IFN-γ, Gata3, and IL-10, representing respectively the Th1, Th2 and regulatory elements of the immune response. Our results show that there was clear evidence of consistent differences between individuals in their typical level of expression of at least one immune gene, and at most all three immune genes, after other measured sources of variation had been taken into account. Furthermore, individuals that responded to changing circumstances by increasing expression levels of Gata3 had a correlated increase in expression levels of IFN-γ. Our work stresses the importance of acknowledging immunological variation amongst individuals in studies of parasitological and infectious disease risk in wildlife populations

    The XXL Survey IV. Mass-temperature relation of the bright cluster sample

    The XXL survey is the largest survey carried out by XMM-Newton. Covering an area of 50deg2^2, the survey contains 450\sim450 galaxy clusters out to a redshift \sim2 and to an X-ray flux limit of 5×1015ergs1cm2\sim5\times10^{-15}erg\,s^{-1}cm^{-2}. This paper is part of the first release of XXL results focussed on the bright cluster sample. We investigate the scaling relation between weak-lensing mass and X-ray temperature for the brightest clusters in XXL. The scaling relation is used to estimate the mass of all 100 clusters in XXL-100-GC. Based on a subsample of 38 objects that lie within the intersection of the northern XXL field and the publicly available CFHTLenS catalog, we derive the MWLM_{WL} of each system with careful considerations of the systematics. The clusters lie at 0.1<z<0.60.1<z<0.6 and span a range of T15keV T\simeq1-5keV. We combine our sample with 58 clusters from the literature, increasing the range out to 10keV. To date, this is the largest sample of clusters with MWLM_{WL} measurements that has been used to study the mass-temperature relation. The fit (MTbM\propto T^b) to the XXL clusters returns a slope b=1.780.32+0.37b=1.78^{+0.37}_{-0.32} and intrinsic scatter σlnMT0.53\sigma_{\ln M|T}\simeq0.53; the scatter is dominated by disturbed clusters. The fit to the combined sample of 96 clusters is in tension with self-similarity, b=1.67±0.12b=1.67\pm0.12 and σlnMT0.41\sigma_{\ln M|T}\simeq0.41. Overall our results demonstrate the feasibility of ground-based weak-lensing scaling relation studies down to cool systems of 1keV\sim1keV temperature and highlight that the current data and samples are a limit to our statistical precision. As such we are unable to determine whether the validity of hydrostatic equilibrium is a function of halo mass. An enlarged sample of cool systems, deeper weak-lensing data, and robust modelling of the selection function will help to explore these issues further

    Climate Change and the Future of California's Endemic Flora

    The flora of California, a global biodiversity hotspot, includes 2387 endemic plant taxa. With anticipated climate change, we project that up to 66% will experience >80% reductions in range size within a century. These results are comparable with other studies of fewer species or just samples of a region's endemics. Projected reductions depend on the magnitude of future emissions and on the ability of species to disperse from their current locations. California's varied terrain could cause species to move in very different directions, breaking up present-day floras. However, our projections also identify regions where species undergoing severe range reductions may persist. Protecting these potential future refugia and facilitating species dispersal will be essential to maintain biodiversity in the face of climate change
