    Socioeconomic determinants of type 2 diabetes in England

    Background: The rise in diabetes worldwide and its significance as a cause of disability and premature death has increased the interest of researchers in diabetes mellitus. There is a large volume of published studies describing the role of factors such as income, poor education, inadequate housing conditions and unemployment as key social determinants in the development and progression of type 2 diabetes (T2D). This study therefore set out to investigate the main socioeconomic predictors of T2D in England. Methods: Based on the available literature on the socioeconomic determinants of T2D, the variables, unemployment (long-term and short-term), overcrowding, income deprivation and no qualification were selected. Data on the socioeconomic determinants and prevalence of type 2 diabetes were retrieved online from the registry of the Office for National Statistics and the Health and Social Care Information Centre respectively. This was done specifically for the 64 randomly selected districts in England. Following preliminary analysis, multiple regression was conducted to ensure there was no violation of the assumptions of regression. Results: Correlation indicates a significant moderate positive correlation between the prevalence of T2D and each of the variables: long-term unemployment, no qualifications and income deprivation, but no significant relationship with overcrowding. Multiple linear regression analysis indicates that long-term unemployment and no qualification were significant predictors of T2D in England. Conclusions: The evidence from this study suggests that the prioritisation of resources, especially in periods of austerity and welfare reforms, to target these key predictors may achieve much in reducing the prevalence of T2D in England. Hence, it is recommended that a comprehensive data collection of these demographic characteristics are integrated into the existing process, and the collective effects of social, economic and biologic factors are investigated in future studies

    Timing of maternal exposure and fetal sex determine the effects of low-level chemical mixture exposure on the fetal neuroendocrine system in sheep

    We have shown that continuous maternal exposure to the complex mixture of environmental chemicals (ECs) found in human biosolids (sewage sludge), disrupts mRNA expression of genes crucial for development and long-term regulation of hypothalamo-pituitary gonadal (HPG) function in sheep. This study investigated whether exposure to ECs only during preconceptional period or only during pregnancy perturbed key regulatory genes within the hypothalamus and pituitary gland and whether these effects were different from chronic (life-long) exposure to biosolid ECs. The findings demonstrate that the timing and duration of maternal EC exposure influences the subsequent effects on the fetal neuroendocrine system in a sex-specific manner. Maternal exposure prior to conception or during pregnancy only, altered the expression of key fetal neuroendocrine regulatory systems such as GnRH and kisspeptin to a greater extent than when maternal exposure was ‘life-long’. Furthermore, hypothalamic gene expression was affected to a greater extent in males than in females, and following EC exposure, male fetuses expressed more “female-like” mRNA levels for some key neuroendocrine genes. This is the first study to show that “real-life” maternal exposure to low levels of a complex cocktail of chemicals prior to conception can subsequently affect the developing fetal neuroendocrine system. These findings demonstrate that the developing neuroendocrine system is sensitive to EC mixtures in a sex-dimorphic manner likely to predispose to reproductive dysfunction in later life

    Evolution of Melanopsin Photoreceptors: Discovery and Characterization of a New Melanopsin in Nonmammalian Vertebrates

    In mammals, the melanopsin gene (Opn4) encodes a sensory photopigment that underpins newly discovered inner retinal photoreceptors. Since its first discovery in Xenopus laevis and subsequent description in humans and mice, melanopsin genes have been described in all vertebrate classes. Until now, all of these sequences have been considered representatives of a single orthologous gene (albeit with duplications in the teleost fish). Here, we describe the discovery and functional characterisation of a new melanopsin gene in fish, bird, and amphibian genomes, demonstrating that, in fact, the vertebrates have evolved two quite separate melanopsins. On the basis of sequence similarity, chromosomal localisation, and phylogeny, we identify our new melanopsins as the true orthologs of the melanopsin gene previously described in mammals and term this grouping Opn4m. By contrast, the previously published melanopsin genes in nonmammalian vertebrates represent a separate branch of the melanopsin family which we term Opn4x. RT-PCR analysis in chicken, zebrafish, and Xenopus identifies expression of both Opn4m and Opn4x genes in tissues known to be photosensitive (eye, brain, and skin). In the day-14 chicken eye, Opn4m mRNA is found in a subset of cells in the outer nuclear, inner nuclear, and ganglion cell layers, the vast majority of which also express Opn4x. Importantly, we show that a representative of the new melanopsins (chicken Opn4m) encodes a photosensory pigment capable of activating G protein signalling cascades in a light- and retinaldehyde-dependent manner under heterologous expression in Neuro-2a cells. A comprehensive in silico analysis of vertebrate genomes indicates that while most vertebrate species have both Opn4m and Opn4x genes, the latter is absent from eutherian and, possibly, marsupial mammals, lost in the course of their evolution as a result of chromosomal reorganisation. Thus, our findings show for the first time that nonmammalian vertebrates retain two quite separate melanopsin genes, while mammals have just one. These data raise important questions regarding the functional differences between Opn4x and Opn4m pigments, the associated adaptive advantages for most vertebrate species in retaining both melanopsins, and the implications for mammalian biology of lacking Opn4x

    Defining optimal thresholds for digital Brix refractometry to determine IgG concentration in ewe colostrum and lamb serum in Scottish lowland sheep flocks

    This research aimed to define thresholds for ewe colostrum and lamb serum Brix refractometer measurements in lowland Scottish sheep. This would facilitate the use of this convenient, sheep-side test, enabling quick and accurate identification of poor quality colostrum and prevention of failure of transfer of passive immunity (FTPI) in lambs. Secondary aims were to identify risk factors for poor colostrum quality and FTPI in lambs. Serum samples (n = 233) were collected from lambs between 24 and 48hrs after birth, from four lowland Scottish meat sheep farms. Pre-suckle colostrum samples (n = 112) were also collected from ewes on two of these farms. Farmers provided information on litter size, ewe body condition score, ewe breed and dystocia. Duplicate digital Brix refractometer measurements were compared with immunoglobulin G (IgG) radial immunodiffusion (RID) testing for all colostrum and serum samples. Receiver operating characteristic (ROC) curves were used to redefine thresholds for Brix testing in colostrum and serum. Linear regression models were constructed with colostrum and serum IgG concentration as the outcomes of interest. Colostrum and serum IgG concentrations were highly variable. The prevalence of inadequate colostrum quality (using <50 g/L IgG on RID) was 4.5% (95% CI = 1.5 – 10.1) and the prevalence of FTPI (using <15 g/L IgG in serum on RID) was 7.73% (95% CI = 4.64–11.93). A ewe’s colostrum IgG concentration was significantly and positively associated with the serum IgG concentration of her lamb(s) (p = 0.02). ROC analysis defined a Brix threshold for adequate colostrum quality of > 22.10% (sensitivity 80% (95%CI=28.4–99.5), specificity 90% (95%CI=82.3–94.8)). ROC analysis defined a Brix threshold for serum of > 8.65% for adequate passive transfer of immunity in Scottish lambs (sensitivity 94% (95%CI=72.7–99.8), specificity 82% (95%CI=76.6–87.2)). To optimise passive transfer of immunity in lambs, we suggest that ewe colostrum Brix measurements be defined as ‘poor’ (<22%); ‘fair’ (22–26%) and ‘good’ (>26%); and lamb serum as ‘poor’ (<8%); ‘fair’ (8–9%) and ‘good’ (>9%). It is recommended that these tests are used as for flock screening, using samples from multiple animals

    Breast cancer risk and prevention in 2024: An overview from the Breast Cancer UK ‐ Breast Cancer Prevention Conference

    The Breast Cancer UK—Breast Cancer Prevention Conference addressed risk from environmental pollutants and health behaviour-related breast-cancer risk. Epidemiological studies examining individual chemicals and breast cancer risk have produced inconclusive results including endocrine disrupting chemicals (EDCs) Bisphenol A, per- and polyfluorinated alkyl substances as well as aluminium. However, laboratory studies have shown that multiple EDCs, can work together to exhibit effects, even when combined at levels that alone are ineffective. The TEXB-α/β assay measures total estrogenic load, and studies have provided evidence of a link between multiple-chemical exposures and breast cancer. However, prospective studies using TEXB-α/β are needed to establish a causative link. There is also a need to assess real-life exposure to environmental-chemical mixtures during pregnancy, and their potential involvement in programming adverse foetal health outcomes in later life. Higher rates of breast cancer have occurred alongside increases in potentially-modifiable risk factors such as obesity. Increasing body-mass index is associated with increased risk of developing postmenopausal breast cancer, but with decreased risk of premenopausal breast cancer. In contrast, lower rates of breast cancer in Asian compared to Western populations have been linked to soya/isoflavone consumption. Risk is decreased by breastfeeding, which is in addition to the decrease in risk observed for each birth and a young first-birth. Risk is lower in those with higher levels of self-reported physical activity. Current evidence suggests breast-cancer survivors should also avoid weight gain, be physically active, and eat a healthy diet for overall health. A broad scientific perspective on breast cancer risk requires focus on both environmental exposure to chemicals and health behaviour-related risk. Research into chemical exposure needs to focus on chemical mixtures and prospective epidemiological studies in order to test the effects on breast cancer risk. Behaviour-related research needs to focus on implementation as well as deeper understanding of the mechanisms of cancer prevention

    Edge influence on vegetation at natural and anthropogenic edges of boreal forests in Canada and Fennoscandia

    Although anthropogenic edges are an important consequence of timber harvesting, edges due to natural disturbances or landscape heterogeneity are also common. Forest edges have been well studied in temperate and tropical forests, but less so in less productive, disturbance-adapted boreal forests. We synthesized data on forest vegetation at edges of boreal forests and compared edge influence among edge types (fire, cut, lake/wetland; old vs. young), forest types (broadleaf vs. coniferous) and geographic regions. Our objectives were to quantify vegetation responses at edges of all types and to compare the strength and extent of edge influence among different types of edges and forests. Research was conducted using the same general sampling design in Alberta, Ontario and Quebec in Canada, and in Sweden and Finland. We conducted a meta-analysis for a variety of response variables including forest structure, deadwood abundance, regeneration, understorey abundance and diversity, and non-vascular plant cover. We also determined the magnitude and distance of edge influence (DEI) using randomization tests. Some edge responses (lower tree basal area, tree canopy and bryophyte cover; more logs; higher regeneration) were significant overall across studies. Edge influence on ground vegetation in boreal forests was generally weak, not very extensive (DEI usually < 20 m) and decreased with time. We found more extensive edge influence at natural edges, at younger edges and in broadleaf forests. The comparison among regions revealed weaker edge influence in Fennoscandian forests. Synthesis. Edges created by forest harvesting do not appear to have as strong, extensive or persistent influence on vegetation in boreal as in tropical or temperate forested ecosystems. We attribute this apparent resistance to shorter canopy heights, inherent heterogeneity in boreal forests and their adaptation to frequent natural disturbance. Nevertheless, notable differences between forest structure responses to natural (fire) and anthropogenic (cut) edges raise concerns about biodiversity implications of extensive creation of anthropogenic edges. By highlighting universal responses to edge influence in boreal forests that are significant irrespective of edge or forest type, and those which vary by edge type, we provide a context for the conservation of boreal forests.     Revealing the sub-AU asymmetries of the inner dust rim in the disk around the Herbig Ae star R CrA

    Models predict that in the innermost AU of the disk around Herbig Ae/Be star, the dust disk forms a "puffed-up" inner rim, which should result in a strongly asymmetric brightness distribution for disks seen under intermediate inclination. Using the VLTI/AMBER long-baseline interferometer, we obtained 24 near-infrared (H- and K-band) spectro-interferometric observations on the Herbig Ae star R CrA. In the derived visibility function, we detect the signatures of an extended (25 mas) and a compact component (5.8 mas), with the compact component contributing about 2/3 of the total flux. The brightness distribution is highly asymmetric, as indicated by the strong closure phases (up to 40 deg) and the detected position angle dependence of the visibilities and closure phases. To interpret these asymmetries, we employ geometric as well as physical models, including a binary model, a skewed ring model, and a puffed-up inner rim model with a vertical or curved rim shape. Our curved puffed-up rim model can reasonably well reproduce the interferometric observables and the SED simultaneously and suggests a luminosity of 29 L_sun and the presence of relatively large (> 1.2 micron) Silicate dust grains. Perpendicular to the disk, two bow shock-like structures appear in the associated reflection nebula NGC 6729, suggesting that the resolved sub-AU size disk is the driving engine of a large-scale outflow. Detecting, for the first time, strong non-localized asymmetries in the inner regions of a Herbig Ae disk, our study supports the existence of a puffed-up inner rim in YSO disks.Comment: 17 pages; 15 figures; Accepted by A&

To effectively study these complex and evolving phenomena, methods that enable autonomous platforms to detect and respond to targeted phenomena are required. Such capabilities allow for directed sensing and water sample acquisition in the most relevant and informative locations, as compared against static grid surveys. To meet this need, we have designed algorithms for autonomous underwater vehicles that detect oceanic features in real time and direct vehicle and sampling behaviors as dictated by research objectives. These methods have successfully been applied in a series of field programs to study a range of phenomena such as harmful algal blooms, coastal upwelling fronts, and microbial processes in open-ocean eddies.
