    Coronin Is a Component of the Endocytic Collar of Hyphae of Neurospora crassa and Is Necessary for Normal Growth and Morphogenesis

    Coronin plays a major role in the organization and dynamics of actin in yeast. To investigate the role of coronin in a filamentous fungus (Neurospora crassa), we examined its subcellular localization using fluorescent proteins and the phenotypic consequences of coronin gene (crn-1) deletion in hyphal morphogenesis, Spitzenkörper behavior and endocytosis. Coronin-GFP was localized in patches, forming a subapical collar near the hyphal apex; significantly, it was absent from the apex. The subapical patches of coronin colocalized with fimbrin, Arp2/3 complex, and actin, altogether comprising the endocytic collar. Deletion of crn-1 resulted in reduced hyphal growth rates, distorted hyphal morphology, uneven wall thickness, and delayed establishment of polarity during germination; it also affected growth directionality and increased branching. The Spitzenkörper of Δcrn-1 mutant was unstable; it appeared and disappeared intermittently giving rise to periods of hyphoid-like and isotropic growth respectively. Uptake of FM4-64 in Δcrn-1 mutant indicated a partial disruption in endocytosis. These observations underscore coronin as an important component of F-actin remodeling in N. crassa. Although coronin is not essential in this fungus, its deletion influenced negatively the operation of the actin cytoskeleton involved in the orderly deployment of the apical growth apparatus, thus preventing normal hyphal growth and morphogenesis

    MAVS Signaling Is Required for Preventing Persistent Chikungunya Heart Infection and Chronic Vascular Tissue Inflammation

    Chikungunya virus (CHIKV) infection has been associated with severe cardiac manifestations, yet, how CHIKV infection leads to heart disease remains unknown. Here, we leveraged both mouse models and human primary cardiac cells to define the mechanisms of CHIKV heart infection. Using an immunocompetent mouse model of CHIKV infection as well as human primary cardiac cells, we demonstrate that CHIKV directly infects and actively replicates in cardiac fibroblasts. In immunocompetent mice, CHIKV is cleared from cardiac tissue without significant damage through the induction of a local type I interferon response from both infected and non-infected cardiac cells. Using mice deficient in major innate immunity signaling components, we found that signaling through the mitochondrial antiviral-signaling protein (MAVS) is required for viral clearance from the heart. In the absence of MAVS signaling, persistent infection leads to focal myocarditis and vasculitis of the large vessels attached to the base of the heart. Large vessel vasculitis was observed for up to 60 days post infection, suggesting CHIKV can lead to vascular inflammation and potential long-lasting cardiovascular complications. This study provides a model of CHIKV cardiac infection and mechanistic insight into CHIKV-induced heart disease, underscoring the importance of monitoring cardiac function in patients with CHIKV infections

    Airborne nitrogen deposition to the Baltic Sea: past trends, source allocation and future

    Despite significant reductions in nitrogen emissions achieved in Europe during the last three decades, eutrophication remains an environmental concern in the Baltic Sea basin. Recently, a number of comprehensive modelling studies have been conducted for the HELCOM Commission to inform the 2021 update of the Baltic Sea Action Plan. The calculations have focused on trends in airborne nitrogen deposition to the Baltic Sea and its nine sub-basins during the 2000-2017 period, the identification and ranking of the main contributors to deposition, as well as future projections for 2030, assuming compliance with the Gothenburg Protocol and the EU NEC Directive. This paper synthesizes the main results from these studies and puts them into the context of maximum allowable nutrient inputs to the Baltic Sea. According to our results, the airborne annual deposition to the Baltic Sea in 2017 amounted to 122.6 Gg(N) of oxidized nitrogen and 105.3 Gg(N) of reduced nitrogen, corresponding to a decrease since 2000 by, respectively, 39% and 11%. In order to filter out the large inter-annual variability due to meteorology and to better reflect trends in emissions, weather-normalized depositions of nitrogen have been calculated as well, according to which the decreases since 2000 amount to 35%, 7% and 25% for oxidized, reduced and total nitrogen, respectively. In 2017, Germany, Poland and Denmark were the most important contributors to airborne deposition of total (oxidized + reduced) nitrogen to the Baltic Sea. Agriculture contributed most to reduced nitrogen deposition, while the transport sector contributed most to oxidized nitrogen deposition. Agriculture in Germany was the single-most important contributor to nitrogen deposition to the Baltic Sea basin in 2017 (accounting for about 15% of the total), but there are numerous other important sectoral contributions. Emissions of nitrogen from the nine HELCOM Contracting Parties contributed 49%, 76% and 61% to oxidized, reduced and total nitrogen deposition, respectively. Assuming full compliance with the EU NEC Directive and the Gothenburg Protocol, significant further reductions in nitrogen deposition can be achieved by 2030, down to an annual deposition of 72.7 Gg(N) and 84.7 Gg(N) of oxidized and reduced nitrogen, respectively

    A GIS model-based assessment of the environmental distribution of g-hexachlorocyclohexane in European soils and waters

    The MAPPE GIS based multimedia model is used to produce a quantitative description of the behaviour of γ-hexachlorocyclohexane (γ-HCH) in Europe, with emphasis on continental surface waters. The model is found to reasonably reproduce γ-HCH distributions and variations along the years in atmosphere and soil; for continental surface waters, concentrations were reasonably well predicted for year 1995, when lindane was still used in agriculture, while for 2005, assuming severe restrictions in use, yields to substantial underestimation. Much better results were yielded when same mode of release as in 1995 was considered, supporting the conjecture that for γ-HCH, emission data rather that model structure and parameterization can be responsible for wrong estimation of concentrations. Future research should be directed to improve the quality of emission data. Joint interpretation of monitoring and modelling results, highlights that lindane emissions in Europe, despite the marked decreasing trend, persist beyond the provisions of existing legislation.     The Functions of Myosin II and Myosin V Homologs in Tip Growth and Septation in Aspergillus nidulans

    Because of the industrial and medical importance of members of the fungal genus Aspergillus, there is considerable interest in the functions of cytoskeletal components in growth and secretion in these organisms. We have analyzed the genome of Aspergillus nidulans and found that there are two previously unstudied myosin genes, a myosin II homolog, myoB (product = MyoB) and a myosin V homolog, myoE (product = MyoE). Deletions of either cause significant growth defects. MyoB localizes in strings that coalesce into contractile rings at forming septa. It is critical for septation and normal deposition of chitin but not for hyphal extension. MyoE localizes to the Spitzenkörper and to moving puncta in the cytoplasm. Time-lapse imaging of SynA, a v-SNARE, reveals that in myoE deletion strains vesicles no longer localize to the Spitzenkörper. Tip morphology is slightly abnormal and branching occurs more frequently than in controls. Tip extension is slower than in controls, but because hyphal diameter is greater, growth (increase in volume/time) is only slightly reduced. Concentration of vesicles into the Spitzenkörper before incorporation into the plasma membrane is, thus, not required for hyphal growth but facilitates faster tip extension and a more normal hyphal shape
