Isolation of a Housefly Head Protein Fraction that Exhibits High Affinity Binding of Cholinergic Ligands
The purification is described of a protein fraction, isolated from
the central nervous system of housefly heads, that exhibits high
affinity for cholinergic ligands. The purified material was found tq
bind with high affinity acetylcholine, nicotinic ligands such as
nicotine and decamethonium as well as atropine, dexetimide a:qg
pilocarpine which are of a muscarinic nature. With all the ligands
there appeared to be only a single site for binding with measured
dissociation constants varying from 6.2 X 10-s M (dexetimide) to
5.4 x 10-6 M (pilocarpine). The concentration of binding sites was
in the range of 381 nmol g-1 of protein (atropine) to 560 nmol g-1
of protein (pilocarpine)

Optimized pulse source based on a gain-switched laser diode in conjuction with a non-linearly chirped grating for 40 Gbit/s systems
A technique based on the design of an optimized source of pico-second optical pulses, which exhibits excellent temporal and spectral purity, is presented. The procedure entails an initial complete intensity and chirp characterization of pulses, from an externally injected-gain switched laser (EI-GSL), using the technique of frequency resolved optical gating (FROG). This characterization yields the parameters that are required for the design of a non-linearly chirped fibre Bragg grating (NC FBG) with a chirp profile that is opposite to that measured across the pulse. By employing the tailor made NC FBG after the gain-switched laser, direct compression of the gain-switched pulses is achieved, to obtain pedestal-free, near transform-limited, 7 ps pulses. The system viability, characterized by simulations, portrays a 6 dB difference in performance between a pulse source with insufficient TPSR and one that exhibits adequate TPSR &(ge; 30 dB)
ACE-ASIA - Regional climatic and atmospheric chemical effects of Asian dust and pollution
Although continental-scale plumes of Asian dust and pollution reduce the amount of solar radiation reaching the earth's surface and perturb the chemistry of the atmosphere, our ability to quantify these effects has been limited by a lack of critical observations, particularly of layers above the surface. Comprehensive surface, airborne, shipboard, and satellite measurements of Asian aerosol chemical composition, size, optical properties, and radiative impacts were performed during the Asian Pacific Regional Aerosol Characterization Experiment (ACE-Asia) study. Measurements within a massive Chinese dust storm at numerous widely spaced sampling locations revealed the highly complex structure of the atmosphere, in which layers of dust, urban pollution, and biomass-burning smoke may be transported long distances as distinct entities or mixed together. The data allow a first-time assessment of the regional climatic and atmospheric chemical effects of a continental-scale mixture of dust and pollution. Our results show that radiative flux reductions during such episodes are sufficient to cause regional climate change
The influence of methylphenidate on the power spectrum of ADHD children â an MEG study
BACKGROUND: The present study was dedicated to investigate the influence of Methylphenidate (MPH) on cortical processing of children who were diagnosed with different subtypes of Attention Deficit Hyperactivity Disorder (ADHD). As all of the previous studies investigating power differences in different frequency bands have been using EEG, mostly with a relatively small number of electrodes our aim was to obtain new aspects using high density magnetoencephalography (MEG). METHODS: 35 children (6 female, 29 male) participated in this study. Mean age was 11.7 years (± 1.92 years). 17 children were diagnosed of having an Attention-Deficit/Hyperactivity Disorder of the combined type (ADHDcom, DSM IV code 314.01); the other 18 were diagnosed for ADHD of the predominantly inattentive type (ADHDin, DSM IV code 314.0). We measured the MEG during a 5 minute resting period with a 148-channel magnetometer system (MAGNESâą 2500 WH, 4D Neuroimaging, San Diego, USA). Power values were averaged for 5 bands: Delta (D, 1.5â3.5 Hz), Theta (T, 3.5â7.5 Hz), Alpha (A, 7.5â12.5 Hz), Beta (B, 12.5â25 Hz) and Global (GL, 1.5â25 Hz).). Additionally, attention was measured behaviourally using the D2 test of attention with and without medication. RESULTS: The global power of the frequency band from 1.5 to 25 Hz increased with MPH. Relative Theta was found to be higher in the left hemisphere after administration of MPH than before. A positive correlation was found between D2 test improvement and MPH-induced power changes in the Theta band over the left frontal region. A linear regression was computed and confirmed that the larger the improvement in D2 test performance, the larger the increase in Theta after MPH application. CONCLUSION: Main effects induced by medication were found in frontal regions. Theta band activity increased over the left hemisphere after MPH application. This finding contradicts EEG results of several groups who found lower levels of Theta power after MPH application. As relative Theta correlates with D2 test improvement we conclude that MEG provide complementary and therefore important new insights to ADHD
Chemical and Photometric Evolution of Extended Ultraviolet Disks: Optical Spectroscopy of M83 (NGC5236) and NGC4625
We present the results from the analysis of optical spectra of 31
Halpha-selected regions in the extended UV (XUV) disks of M83 (NGC5236) and
NGC4625 recently discovered by GALEX. The spectra were obtained using IMACS at
Las Campanas Observatory 6.5m Magellan I telescope and COSMIC at the Palomar
200-inch telescope, respectively for M83 and NGC4625. The line ratios measured
indicate nebular oxygen abundances (derived from the R23 parameter) of the
order of Zsun/5-Zsun/10. For most emission-line regions analyzed the line
fluxes and ratios measured are best reproduced by models of photoionization by
single stars with masses in the range 20-40 Msun and oxygen abundances
comparable to those derived from the R23 parameter. We find indications for a
relatively high N/O abundance ratio in the XUV disk of M83. Although the
metallicities derived imply that these are not the first stars formed in the
XUV disks, such a level of enrichment could be reached in young spiral disks
only 1 Gyr after these first stars would have formed. The amount of gas in the
XUV disks allow maintaining the current level of star formation for at least a
few Gyr.Comment: 52 pages, 8 tables, 7 figures, accepted for publication in Ap
SNPs for Parentage Testing and Traceability in Globally Diverse Breeds of Sheep
DNA-based parentage determination accelerates genetic improvement in sheep by increasing pedigree accuracy. Single nucleotide polymorphism (SNP) markers can be used for determining parentage and to provide unique molecular identifiers for tracing sheep products to their source. However, the utility of a particular ââparentage SNPââ varies by breed depending on its minor allele frequency (MAF) and its sequence context. Our aims were to identify parentage SNPs with exceptional qualities for use in globally diverse breeds and to develop a subset for use in North American sheep. Starting with genotypes from 2,915 sheep and 74 breed groups provided by the International Sheep Genomics Consortium (ISGC), we analyzed 47,693 autosomal SNPs by multiple criteria and selected 163 with desirable properties for parentage testing. On average, each of the 163 SNPs was highly informative (MAF â„ 0.3) in 48±5 breed groups. Nearby polymorphisms that could otherwise confound genetic testing were identified by whole genome and Sanger sequencing of 166 sheep from 54 breed groups. A genetic test with 109 of the 163 parentage SNPs was developed for matrix-assisted laser desorption/ionizationâ time-of-flight mass spectrometry. The scoring rates and accuracies for these 109 SNPs were greater than 99% in a panel of North American sheep. In a blinded set of 96 families (sire, dam, and non-identical twin lambs), each parent of every lamb was identified without using the other parentâs genotype. In 74 ISGC breed groups, the median estimates for probability of a coincidental match between two animals (PI), and the fraction of potential adults excluded from parentage (PE) were 1.1Ă10(â39) and 0.999987, respectively, for the 109 SNPs combined. The availability of a well-characterized set of 163 parentage SNPs facilitates the development of high-throughput genetic technologies for implementing accurate and economical parentage testing and traceability in many of the worldâs sheep breeds
Attitudes and Performance: An Analysis of Russian Workers
This paper investigates the relationship between locus of control and performance among Russian employees, using survey data collected at 28 workplaces in 2002 in Taganrog and at 47 workplaces in 2003 in Ekaterinburg. We develop a measure that allows us to categorize the Russian employees participating in our survey as exhibiting an internal or external locus of control. We then assess the extent to which there are significant differences between âinternalsâ and âexternalsâ in work-related attitudes that may affect performance. In particular, we focus on (1) attitudes about outcomes associated with hard work, (2) level of job satisfaction, (3) expectation of receiving a desired reward, and (4) loyalty to and involvement with oneâs organization. In each case we identify where gender and generational differences emerge. Our main objective is to determine whether Russian employees who exhibit an internal locus of control perform better than employees with an external locus of control. Our performance measures include earnings, expected promotions, and assessments of the quantity and quality of work in comparison to others at the same organization doing a similar job. Controlling for a variety of worker characteristics, we find that (1) individuals who exhibit an internal locus of control perform better, but this result is not always statistically significant; (2) even among âinternals,â women earn significantly less than men and have a much lower expectation of promotion; (3) even among âinternals,â experience with unemployment has a negative influence on performance.http://deepblue.lib.umich.edu/bitstream/2027.42/40144/3/wp758.pd
Uncovering treatment burden as a key concept for stroke care: a systematic review of qualitative research
<b>Background</b> Patients with chronic disease may experience complicated management plans requiring significant personal investment. This has been termed âtreatment burdenâ and has been associated with unfavourable outcomes. The aim of this systematic review is to examine the qualitative literature on treatment burden in stroke from the patient perspective.<p></p>
<b>Methods and findings</b> The search strategy centred on: stroke, treatment burden, patient experience, and qualitative methods. We searched: Scopus, CINAHL, Embase, Medline, and PsycINFO. We tracked references, footnotes, and citations. Restrictions included: English language, date of publication January 2000 until February 2013. Two reviewers independently carried out the following: paper screening, data extraction, and data analysis. Data were analysed using framework synthesis, as informed by Normalization Process Theory. Sixty-nine papers were included. Treatment burden includes: (1) making sense of stroke management and planning care, (2) interacting with others, (3) enacting management strategies, and (4) reflecting on management. Health care is fragmented, with poor communication between patient and health care providers. Patients report inadequate information provision. Inpatient care is unsatisfactory, with a perceived lack of empathy from professionals and a shortage of stimulating activities on the ward. Discharge services are poorly coordinated, and accessing health and social care in the community is difficult. The study has potential limitations because it was restricted to studies published in English only and data from low-income countries were scarce.<p></p>
<b>Conclusions</b> Stroke management is extremely demanding for patients, and treatment burden is influenced by micro and macro organisation of health services. Knowledge deficits mean patients are ill equipped to organise their care and develop coping strategies, making adherence less likely. There is a need to transform the approach to care provision so that services are configured to prioritise patient needs rather than those of health care systems
