    Grain quality characteristics of imported rice in Ghana: Implications for breeding for consumer-preferred varieties

    ABSTRACTRice is the fastest growing food source in Ghana. The country, however, imports about 70 per cent of its rice requirement due to low volumes of production and poor grain quality of domestic rice compared to imported ones.  In the study, 10 popular imported rice brands on the Ghanaian market were characterised for grain quality traits including grain dimensions, apparent amylose content, gelatinisation temperature, paste viscosity properties, and aroma using physiochemical properties and DNA markers. The rice brands came from Asia and the USA. The rice type from Asia was found to be Jasmine-styled aromatic long grained with low amylose content and gelatinisation temperature, whereas those of USA were conventional long grain with intermediate amylose content and gelatinisation temperature.  These findings were confirmed through DNA marker analyses, with the rice types from Asia and the USA revealing the presence of the TAC and GCC of the waxy gene SNP haplotypes, respectively.     Determination of Trace Elements in the Sakumo Wetland Sediments

    Abstract: The objective of this research was to determine the current pollution status of the Sakumo wetland with the aim of identifying factors affecting the long-term integrity of the wetland ecosystem. Sediment samples from the Sakumo wetland were analysed for Cd, Co, Cr, Cu, Fe, Ni, Mn and Zn using Instrumental Neutron Activation Analysis (INAA) coupled with the conventional counting system. The sediment materials exhibited higher concentrations of trace elements Cd (maximum; 0.041 mg/kg), Co (maximum; 0.64 mg/kg), Cr (maximum; 30.73 mg/kg), Cu (maximum; 22.89 mg/kg), Ni (maximum; 11.69 mg/kg) and Zn (maximum; 6.52 mg/kg). In some of the lagoon sediments compared with their levels in world average soils, the average concentrations of the trace elements in general are below or within levels in world average soils/uncontaminated soils. Concentrations of Ni showed positive correlation with Cr whilst Co correlated positively with Cr and Zn. However, lack of correlation between Fe and Cd, suggests that the influence of these parameters on the distribution of trace metals is not important

    Nurturing novelty: Toulmin's greenhouse, journal rankings and the evolution of knowledge

    Reflecting on Toulmin’s 1972 conceptualization of the academic research process, we fast-forward his thinking to the current climate of academic excellence and associated journal ranking lists. We argue that the formal and informal use of such rankings throughout the hierarchy of research institutions creates an artificial environment within which favoured ‘branches’ of knowledge continue to flourish at the expense of new conceptual saplings. This ‘greenhouse’ effect might result in the creation of a knowledge tree which is increasingly unfit to the external world for which it is intended. We thus step back and examine wider implications of these factors on the broader evolution of knowledge in the research process. In sum, we argue for a Toulminian explanation of the process by which journal ranking lists reduce academic innovation and creativity, bias academic selection and constrain dissemination processes in the academic community

    Farmers' perceptions on mechanical weeders for rice production in sub-Saharan Africa

    Competition from weeds is one of the major biophysical constraints to rice (Oryza spp.) production in sub-Saharan Africa. Smallholder rice farmers require efficient, affordable and labour-saving weed management technologies. Mechanical weeders have shown to fit this profile. Several mechanical weeder types exist but little is known about locally specific differences in performance and farmer preference between these types. Three to six different weeder types were evaluated at 10 different sites across seven countries – i.e., Benin, Burkina Faso, Côte d'Ivoire, Ghana, Nigeria, Rwanda and Togo. A total of 310 farmers (173 male, 137 female) tested the weeders, scored them for their preference, and compared them with their own weed management practices. In a follow-up study, 186 farmers from Benin and Nigeria received the ring hoe, which was the most preferred in these two countries, to use it during the entire crop growing season. Farmers were surveyed on their experiences. The probability of the ring hoe having the highest score among the tested weeders was 71%. The probability of farmers’ preference of the ring hoe over their usual practices – i.e., herbicide, traditional hoe and hand weeding – was 52, 95 and 91%, respectively. The preference of this weeder was not related to gender, years of experience with rice cultivation, rice field size, weed infestation level, water status or soil texture. In the follow-up study, 80% of farmers who used the ring hoe indicated that weeding time was reduced by at least 31%. Of the farmers testing the ring hoe in the follow-up study, 35% used it also for other crops such as vegetables, maize, sorghum, cassava and millet. These results suggest that the ring hoe offers a gender-neutral solution for reducing labour for weeding in rice as well as other crops and that it is compatible with a wide range of environments. The implications of our findings and challenges for out-scaling of mechanical weeders are discussed

    The repulsive lattice gas, the independent-set polynomial, and the Lov\'asz local lemma

    We elucidate the close connection between the repulsive lattice gas in equilibrium statistical mechanics and the Lovasz local lemma in probabilistic combinatorics. We show that the conclusion of the Lovasz local lemma holds for dependency graph G and probabilities {p_x} if and only if the independent-set polynomial for G is nonvanishing in the polydisc of radii {p_x}. Furthermore, we show that the usual proof of the Lovasz local lemma -- which provides a sufficient condition for this to occur -- corresponds to a simple inductive argument for the nonvanishing of the independent-set polynomial in a polydisc, which was discovered implicitly by Shearer and explicitly by Dobrushin. We also present some refinements and extensions of both arguments, including a generalization of the Lovasz local lemma that allows for "soft" dependencies. In addition, we prove some general properties of the partition function of a repulsive lattice gas, most of which are consequences of the alternating-sign property for the Mayer coefficients.     Outcomes of laboratory-confirmed SARS-CoV-2 infection during resurgence driven by Omicron lineages BA.4 and BA.5 compared with previous waves in the Western Cape Province, South Africa

    OBJECTIVE: We aimed to compare clinical severity of Omicron BA.4/BA.5 infection with BA.1 and earlier variant infections among laboratory-confirmed SARS-CoV-2 cases in the Western Cape, South Africa, using timing of infection to infer the lineage/variant causing infection. METHODS: We included public sector patients aged ≥20 years with laboratory-confirmed COVID-19 between 1-21 May 2022 (BA.4/BA.5 wave) and equivalent prior wave periods. We compared the risk between waves of (i) death and (ii) severe hospitalization/death (all within 21 days of diagnosis) using Cox regression adjusted for demographics, comorbidities, admission pressure, vaccination and prior infection. RESULTS: Among 3,793 patients from the BA.4/BA.5 wave and 190,836 patients from previous waves the risk of severe hospitalization/death was similar in the BA.4/BA.5 and BA.1 waves (adjusted hazard ratio (aHR) 1.12; 95% confidence interval (CI) 0.93; 1.34). Both Omicron waves had lower risk of severe outcomes than previous waves. Prior infection (aHR 0.29, 95% CI 0.24; 0.36) and vaccination (aHR 0.17; 95% CI 0.07; 0.40 for at least 3 doses vs. no vaccine) were protective. CONCLUSION: Disease severity was similar amongst diagnosed COVID-19 cases in the BA.4/BA.5 and BA.1 periods in the context of growing immunity against SARS-CoV-2 due to prior infection and vaccination, both of which were strongly protective

    Investigation of the midgut structure and ultrastructure in Cimex lectularius and Cimex pipistrelli (Hemiptera, Cimicidae)

    Cimicidae are temporary ectoparasites, which means that they cannot obtain food continuously. Both Cimex species examined here, Cimex lectularius (Linnaeus 1758) and Cimex pipistrelli (Jenyns 1839), can feed on a non-natal host, C. lectularius from humans on bats, C. pipistrelli on humans, but never naturally. The midgut of C. lectularius and C. pipistrelli is composed of three distinct regions—the anterior midgut (AMG), which has a sack-like shape, the long tube-shaped middle midgut (MMG), and the posterior midgut (PMG). The different ultrastructures of the AMG, MMG, and PMG in both of the species examined suggest that these regions must fulfill different functions in the digestive system. Ultrastructural analysis showed that the AMG fulfills the role of storing food and synthesizing and secreting enzymes, while the MMG is the main organ for the synthesis of enzymes, secretion, and the storage of the reserve material. Additionally, both regions, the AMG and MMG, are involved in water absorption in the digestive system of both Cimex species. The PMG is the part of the midgut in which spherites accumulate. The results of our studies confirm the suggestion of former authors that the structure of the digestive tract of insects is not attributed solely to diet but to the basic adaptation of an ancestor

    Estimating Impact Forces of Tail Club Strikes by Ankylosaurid Dinosaurs

    BACKGROUND: It has been assumed that the unusual tail club of ankylosaurid dinosaurs was used actively as a weapon, but the biological feasibility of this behaviour has not been examined in detail. Ankylosaurid tail clubs are composed of interlocking vertebrae, which form the handle, and large terminal osteoderms, which form the knob. METHODOLOGY/PRINCIPAL FINDINGS: Computed tomographic (CT) scans of several ankylosaurid tail clubs referred to Dyoplosaurus and Euoplocephalus, combined with measurements of free caudal vertebrae, provide information used to estimate the impact force of tail clubs of various sizes. Ankylosaurid tails are modeled as a series of segments for which mass, muscle cross-sectional area, torque, and angular acceleration are calculated. Free caudal vertebrae segments had limited vertical flexibility, but the tail could have swung through approximately 100 degrees laterally. Muscle scars on the pelvis record the presence of a large M. longissimus caudae, and ossified tendons alongside the handle represent M. spinalis. CT scans showed that knob osteoderms were predominantly cancellous, which would have lowered the rotational inertia of the tail club and made it easier to wield as a weapon. CONCLUSIONS/SIGNIFICANCE: Large knobs could generate sufficient force to break bone during impacts, but average and small knobs could not. Tail swinging behaviour is feasible in ankylosaurids, but it remains unknown whether the tail was used for interspecific defense, intraspecific combat, or both

    Extrinsic Fluorescent Dyes as Tools for Protein Characterization

    Noncovalent, extrinsic fluorescent dyes are applied in various fields of protein analysis, e.g. to characterize folding intermediates, measure surface hydrophobicity, and detect aggregation or fibrillation. The main underlying mechanisms, which explain the fluorescence properties of many extrinsic dyes, are solvent relaxation processes and (twisted) intramolecular charge transfer reactions, which are affected by the environment and by interactions of the dyes with proteins. In recent time, the use of extrinsic fluorescent dyes such as ANS, Bis-ANS, Nile Red, Thioflavin T and others has increased, because of their versatility, sensitivity and suitability for high-throughput screening. The intention of this review is to give an overview of available extrinsic dyes, explain their spectral properties, and show illustrative examples of their various applications in protein characterization
